A single-nucleotide polymorphism(SNP)is an alteration in one nucleotide in a certain position within a genome.SNPs are associated with disease susceptibility.However,the influences of SNPs on the pathogenesis of neonatal hypoxic-ischemic brain damage remain elusive.Seven-day-old rats were used to establish a hypoxic ischemic encephalopathy model.SNPs and expression profiles of mRNAs were analyzed in hypoxic ischemic encephalopathy model rats using RNA sequencing.Genes exhibiting SNPs associated with hypoxic ischemic encephalopathy were identified and studied by gene ontology and pathway analysis to identify their possible involvement in the disease mechanism.We identified 89 up-regulated genes containing SNPs that were mainly located on chromosome 1 and 2.Gene ontology analysis indicated that the up-regulated genes containing SNPs are mainly involved in angiogenesis,wound healing and glutamatergic synapse and biological processing of calcium-activated chloride channels.Signaling pathway analysis indicated that the differentially expressed genes play a role in glutamatergic synapses,long-term depression and oxytocin signaling.Moreover,intersection analysis of high throughput screening following PubMed retrieval and RNA sequencing for SNPs showed that CSRNP1,DUSP5 and LRRC25 were most relevant to hypoxic ischemic encephalopathy.Significant up-regulation of genes was confirmed by quantitative real-time polymerase chain reaction analysis of oxygen-glucose-deprived human fetal cortical neurons.Our results indicate that CSRNP1,DUSP5 and LRRC25,containing SNPs,may be involved in the pathogenesis of hypoxic ischemic encephalopathy.These findings indicate a novel direction for further hypoxic ischemic encephalopathy research.This animal study was approved on February 5,2017 by the Animal Care and Use Committee of Kunming Medical University,Yunnan Province,China(approval No.kmmu2019038).Cerebral tissue collection from a human fetus was approved on September 30,2015 by the Ethics Committee of Kunming Medical University,Chin 展开更多

HECT, UBA and WWE domain-containing 1(Huwe1), an E3 ubiquitin ligase involved in the ubiquitin-proteasome system, is widely expressed in brain tissue. Huwe1 is involved in the turnover of numerous substrates, including p53, Mcl-1, Cdc6 and N-myc, thereby playing a critical role in apoptosis and neurogenesis. However, the role of Huwe1 in brain ischemia and reperfusion injury remains unclear. Therefore, in this study, we investigated the role of Huwe1 in an in vitro model of ischemia and reperfusion injury. At 3 days in vitro, primary cortical neurons were transduced with a control or shRNA-Huwe1 lentiviral vector to silence expression of Huwe1. At 7 days in vitro, the cells were exposed to oxygen-glucose deprivation for 3 hours and reperfusion for 24 hours. To examine the role of the c-Jun N-terminal kinase(JNK)/p38 pathway, cortical neurons were pretreated with a JNK inhibitor(SP600125) or a p38 MAPK inhibitor(SB203508) for 30 minutes at 7 days in vitro, followed by ischemia and reperfusion. Neuronal apoptosis was assessed by TUNEL assay. Protein expression levels of JNK and p38 MAPK and of apoptosis-related proteins(p53, Gadd45 a, cleaved caspase-3, Bax and Bcl-2) were measured by western blot assay. Immunofluorescence labeling for cleaved caspase-3 was performed. We observed a significant increase in neuronal apoptosis and Huwe1 expression after ischemia and reperfusion. Treatment with the shRNA-Huwe1 lentiviral vector markedly decreased Huwe1 levels, and significantly decreased the number of TUNEL-positive cells after ischemia and reperfusion. The silencing vector also downregulated the pro-apoptotic proteins Bax and cleaved caspase-3, and upregulated the anti-apoptotic proteins Gadd45 a and Bcl-2. Silencing Huwe1 also significantly reduced p-JNK levels and increased p-p38 levels. Our findings show that downregulating Huwe1 affects the JNK and p38 MAPK signaling pathways as well as the expression of apoptosis-related genes to provide neuroprotection during ischemia and reperfusion. With the digestive endoscopic tunnel technique(DETT),many diseases that previously would have been treated by surgery are now endoscopically curable by establishing a submucosal tunnel between the mucosa and muscularis propria(MP).Through the tunnel,endoscopic diagnosis or treatment is performed for lesions in the mucosa,in the MP,and even outside the gastrointestinal(GI)tract.At present,the tunnel technique application range covers the following:(1)Treatment of lesions originating from the mucosal layer,e.g.,endoscopic submucosal tunnel dissection for oesophageal large or circular early-stage cancer or precancerosis;(2)treatment of lesions from the MP layer,per-oral endoscopic myotomy,submucosal tunnelling endoscopic resection,etc.;and(3)diagnosis and treatment of lesions outside the GI tract,such as resection of lymph nodes and benign tumour excision in the mediastinum or abdominal cavity.With the increasing number of DETTs performed worldwide,endoscopic tunnel therapeutics,which is based on DETT,has been gradually developed and optimized.However,there is not yet an expert consensus on DETT to regulate its indications,contraindications,surgical procedure,and postoperative treatment.The International DETT Alliance signed up this consensus to standardize the procedures of DETT.In this consensus,we describe the definition,mechanism,and significance of DETT,prevention of infection and concepts of DETTassociated complications,methods to establish a submucosal tunnel,and application of DETT for lesions in the mucosa,in the MP and outside the GI tract(indications and contraindications,procedures,pre-and postoperative treatments,effectiveness,complications and treatments,and a comparison between DETT and other operations). 展开更多

Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. We study the hadronic decays of ∧c^+ to the final states ∑^+η and ∑^+η, using an e^+e^- annihilation data sample of 567 pb^-1 taken at a center-of-mass energy of 4.6 GeV with the BESIII detector at the BEPCII collider. We find evidence for the decays ∧^+→∑^+η and ∑^+η' and with statistical significance of and , respectively. Normalizing to the reference decays ∧^+→∑^+π^0 and ∑^+ω, we obtain the ratios of the branching fractions B(∧C^+→∑^+η)/V(∧^→∑^+π^0)and B(∧C^+→∑^+η)/B(∧C^+→∑^+ω)to be and , respectively. The upper limits at the 90% confidence level are set to be B(∧C^→∑^+η)/V(∧^+→∑^+π^0)<0.58 and B(∧C^+→∑^+η)/B(∧C^+→∑^+ω)<1.2. Using BESIII measurements of the branching fractions of the reference decays, we determine B(∧C^+→∑^+η)=(0.41±019±0.05)%(<0.68%) and B(∧C^+→∑^+η)=(1.34+0.53+0.19)%(<1.9%). Here, the first uncertainties are statistical and the second systematic. The obtained branching fraction of ∧C^+→∑^+η is consistent with the previous measurement, and the branching fraction of ∧C^+→∑^+η is measured for the first time. 展开更多

Intraocular pressure(IOP) is a major risk factor for glaucoma. Genetic determinants of intraocular pressure can provide critical insights into the genetic architecture of glaucoma and, as a result, open new avenues for therapeutic intervention. We performed a genome-wide association study and replication analysis of 8,552 Chinese participants. In the genome-wide association study, we identified 51 loci that surpassed the significance of P<9×10-7, and we formally replicated these loci. A combined discovery and replication meta-analysis identified 21 genome-wide loci that surpassed the genome-wide significance of P<5×10-8, including 4 previously reported loci: rs145063132(7 p21.2, ETV1/DGKB), rs548030386(7 q31.2, ST7 near CAV1/CAV2), rs7047871(9 p24.2, GLIS3), and rs2472494(9 q31.1, ABCA1/SLC44 A1). Of the 17 newly identified loci, five were reported to have ocular related phenotypes: PTCH2(rs7525308 in 1 p34.1), LRIF1/DRAM2(rs1282146 in 1 p13.3), COLEC11(rs201143466 in 2 p25.3),SPTBN1(rs4514918 in 2 p16.2), and CRK(rs11078446 in 17 p13.3). The genetic loci identified in this study not only increase our understanding of the genes involved in intraocular pressure but also provide important genetic markers to improve future genetic screening and drug discovery for intraocular pressure disorders. 展开更多

Objective To investigate the molecular characteristics and intracellular growth ability of Legionella pneumophila(L. pneumophila) strains from 1989 to 2016 in Sichuan Province, China. Methods Seventy-nine isolates of L. pneumophila were collected from environmental and clinical sources, including cooling towers, hot springs, bath water, fountains, and patients, and identified with 16 S rRNA gene analysis and serum agglutination assay. The isolates were then typed by Sequence-Based Typing(SBT), and Genotyping of forty-two LP1 strains were analyzed by means of multiple-locus VNTR analysis with 8 loci(MLVA-8). All strains were further analyzed for two virulence genes: Legionella vir homologue(lvh) and repeats in structural toxin(rtx A). The intracellular growth ability of 33 selected isolates was determined by examining their interaction with J774 cells. Results All isolates were identified to L. pneumophila including 11 serogroups, among which the main serogroup were LP1, accounting for 54.43%. Thirty-three different sequence types(STs) from five main clonal groups and five singletons were identified, along with 8 different MLVA patterns. Both the lvh and rtx A loci were found in all 79 strains. Thirty isolates showed high intracellular growth ability in J774 cells. Conclusion L. pneumophila is a potential threat to public health, and effective control and prevention strategies are urgently needed. 展开更多

Molecular, morphological and ecological data show distinct differences from known species, herein we describe these specimens as a new species Trimeresurus yingjiangensis sp. nov Chen, Ding, Shi and Zhang, 2018. Thin-film composite (TFC) nanofiltration (NF) membranes were fabricated via the interfacial polymerization of piperazine (PIP) and 1,3,5-benzenetricart)oiiyl trichloride on polysulfone (PSf) support membranes blended with K^+-responsive poly(N-isopropylacryamideco- acryloylamidobenzo-15-crown-5)(P(NIPAM-co- AAB15C5)). Membranes were characterized by attenuated total reflection Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, atomic force microscope, scanning electron microscope, contact angle, and filtration tests. The results showed that:(1) Under K^+-free conditions, the blended P(NIPAM-co-AAB15C5)/PSf supports had porous and hydrophilic surfaces, thereby producing NF membranes with smooth surfaces and low MgSO4 rejections;(2) With K^+ in the PIP solution, the surface roughness and water permeability of the resultant NF membrane were increased due to the K^+-induced transition of low-content P(NIPAM-co-AAB15C5) from hydrophilic to hydrophobic;(3) After a curing treatment at 95℃, the improved NF membrane achieved an even higher pure water permeability of 10.97 L·m^-2·h^-1 - bar1 under 200 psi. Overall, this study provides a novel method to improve the performance of NF membranes and helps understand the influence of supports on TFC membranes. 展开更多

The Hexi Inland River Basin in an arid region of northwestern China was chosen as the study area for this research.The authors define the vulnerability of an oasis social-ecological system to glacier change;select 16 indicators from natural and socioeconomic systems according to exposure,sensitivity,and adaptive capacity;and construct a vulnerability-assessment indicator system aimed at an inland river basin in the arid region of Northwestern China.Vulnerability of the oasis socialecological system affected by glacier change in the study area is evaluated by Spatial Principal Component Analysis(SPCA)under the circumstance of glacier change.The key factors affecting the vulnerability are analyzed.The vulnerability of the oasis social-ecological system in the Hexi Inland River Basin affected by glacier change is of more than medium grade,accounting for about 48.0%of the total number of counties in the study area.In terms of the spatial pattern of the vulnerability,the oasis economic belt is the most vulnerable.With the rapid development of the area's society and economy,the exposure of the system to glacial changes is significantly increased;and an increase in glacial meltwater is not enough to overcome the impact of increased exposure,which is the main reason for the high vulnerability.Based on the result of the vulnerability analysis and combined with the present industrial structure in the Hexi Inland River Basin,near-,medium-,and long-term adaptation initiatives are put forward in the article. 展开更多

Antipsychotic-induced metabolic disturbance(AIMD)is a common adverse effect of antipsychotics with genetics partly underpinning variation in susceptibility among schizophrenia patients.Melanocortin4 receptor(MC4R)gene,one of the candidate genes for AIMD,has been under-studied in the Chinese patients.We conducted a pharmacogenetic study in a large cohort of Chinese patients with schizophrenia.In this study,we investigated the genetic variation of MC4R in Chinese population by genotyping two SNPs(rs489693 and rs17782313)in 1,991 Chinese patients and examined association of these variants with the metabolic effects that were often observed to be related to AIMD.Metabolic measures,including body mass index(BMI),waist circumference(WC),glucose,triglyceride,high-density lipoprotein(HDL),and low-density lipoprotein(LDL)levels were assessed at baseline and after 6-week antipsychotic treatment.We found that interaction of SNP×medication status(drug-na?ve/medicated)was significantly associated with BMI,WC,and HDL change %,respectively.Both SNPs were significantly associated with baseline BMI and WC in the medicated group.Moderate association of rs489693 with WC,Triglyceride,and HDL change % were observed in the whole sample.In the drug-naive group,we found recessive effects of rs489693 on BMI gain more than 7%,WC and Triglyceride change %,with AA incurring more metabolic adverse effects.In conclusion,the association between rs489693 and the metabolic measures is ubiquitous but moderate.Rs17782313 is less involved in AIMD.Two SNPs confer risk of AIMD to patients treated with different antipsychotics in a similar way. 展开更多

Investigating and monitoring the area of cultivated land reclaimed from rural settlements is important to optimize rural land use and understand spatial patterns. Measuring cultivated land area is costly and inefficient, however, as this land use type is often widely dispersed and scattered. A new method is therefore explored in this study that utilizes a Phantom2 Vision +(P2V), one kind of Dajiang(DJI) unmanned aerial vehicle(UAV). The method proposed here includes generating rural settlement images using a P2V UAV, subsequently correcting them using a camera lens model, matching them with geo-coded high resolution alternatives, mosaicking them, measuring the area of cultivated land reclaimed from rural settlements, evaluating measurement accuracy, and analyzing overall efficiency.The results of this study show that use of a P2V UAV is reasonable in price, less than 8000 yuan(RMB), and that this method is able to measure cultivated land area reclaimed from rural settlements with 99% accuracy. This method is therefore low cost, highly efficient, and low risk, as well as being easy to learn and use. This UAV-based approach is also likely to be easily popularized and be particularly useful both for application across plains and flats as well as over mountains and hills. The method proposed in this study is also likely to prove beneficial for monitoring and managing rural land use and future consolidation. 展开更多

Active seismic sources are critical for obtaining high resolution images of the subsurface.For active imaging in urban areas,environment friendly and green seismic sources are required.In present work,we introduce a new type of green active source based on the gaseous detonation of methane and oxygen.When fired in a closed container,the chemical reaction,i.e.gaseous detonation,will produce high pressure air over 150 MPa.Seismic waves are produced when high pressure air is quickly released to impact the surroundings.The first field experiment of this active source was carried out in December,2017 in Jingdezhen,Jiangxi Province,where a series of active sources were excited to explore their potential in mine exploration.In current work,we analyzed the seismic waves recorded by near-field accelerators and a dense short-period seismic array and compared them with those from a mobile airgun source,another kind of active source by releasing high pressure air into water.The results demonstrate that it can be used for high resolution near surface imaging.Firstly,the gaseous detonation productions are harmless CO2 and water,making it a green explosive source.Secondly,the dominant seismic frequencies are 10-80 Hz and a single shot can be recorded up to 15 km,making it suitable for local structure investigations.Thirdly,it can be excited in vertical wells,similar to traditional powder explosive sources.It can also act as an additional on-land active source to airgun sources,which requires a suitable water body as intermediate media to generate repeating signals.Moreover,the short duration and high frequency signature of the source signals make it safe with no damage to nearby buildings.These make it convenient to excite in urban areas.As a new explosive source,the excitation equipment and conditions,such as gas ratio,sink depth and air-releasing directions,need further investigation to improve seismic wave generation efficiency. 展开更多

Grain weight and grain number are two important traits directly determining grain yield in rice. To date,a lot of genes related to grain weight and grain number have been identified;however, the regulatory mechanism underlying these genes remains largely unknown. In this study, we studied the biological function of OsSPL18 during grain and panicle development in rice. Knockout (KO) mutants of OsSPL18exhibited reduced grain width and thickness, panicle length and grain number, but increased tiller number. Cytological analysis showed that OsSPL18 regulates the development of spikelet hulls by affecting cell proliferation. qRT-PCR and GUS staining analyses showed that OsSPL18 was highly expressed in developing young panicles and young spikelet hulls, in agreement with its function in regulating grain and panicle development. Transcriptional activation experiments indicated that OsSPL18is a functional transcription factor with activation domains in both the N-terminus and C-terminus, and both activation domains are indispensable for its biological functions. Quantitative expression analysis showed that DEP1, a major grain number regulator, was significantly down-regulated in OsSPL18 KO lines.Both yeast one-hybrid and dual-luciferase (LUC) assays showed that OsSPL18 could bind to the DEP1promoter, suggesting that OsSPL18 regulates panicle development by positively regulating the expression of DEP1. Sequence analysis showed that OsSPL18 contains the OsmiR156k complementary sequence in the third exon;5'RLM-RACE experiments indicated that OsSPL18 could be cleaved by OsmiR156k. Taken together, our results uncovered a new OsmiR156k-OsSPL18-DEP1 pathway regulating grain number in rice. 展开更多

The single prefrontal configuration has historically been used as an important diagnostic character for many natricine taxa. For example, the genus Trimerodytes Cope, 1895 was long been regarded as a junior synonym of Opisthotropis Günther, 1872 for their similar prefrontal configurations and the type species, T. balteatus Cope, 1895, has been assigned to the genus Opisthotropis. However, as the number and arrangement of prefrontal vary frequently both at species and generic level, it is questionable whether the synonymization of Trimerodytes reflects their evolutionary relationships. On the basis of recently collected specimens of O. balteata, the generic status of the species was assessed using both molecular and morphological data. Opisthotropis was recovered as polyphyletic with reference to O. balteata, because O. balteata is nested within the genus Sinonatrix Rossman and Eberle, 1977 and is the sister species of the type species of Sinonatrix. Consequently, we herein resurrect the long-overlooked synonym Trimerodytes from Opisthotropis and synonymize the junior generic nomen Sinonatrix with Trimerodytes. In addition, based on morphological similarities between the monotypic genus Paratapinophis Angel, 1929 and Trimerodytes, we doubt about the validity of Paratapinophis. Following taxonomic changes in this work, the taxonomic account of the genus Trimerodytes, updated descriptions of its type species, and diagnostic key to Trimerodytes species are provided. 展开更多

Preparation of pseudo-pure states for NMR quantum computing with one ancillary qubit

Quantum state preparation plays an equally important role as quantum operations and measurements in quantum information processing. The previous methods for initialization require either an exponential number of experiments, or cause signal reduction or place restrictions on molecular structures. In this study, we propose three types of quantum circuits for preparing the pseudo-pure states of(n-1) qubits in the n-coupled Hilbert space, which simply needs the assistance of one ancilla spin and two different experiments independent of n. Most importantly, our methods work well on homo-nuclear and hetero-nuclear molecules without the reduction of signals in the gradient field. As a proof-of-principle demonstration, we experimentally prepared the pseudo-pure states of heteronuclear 2-qubit and homonuclear 4-qubit molecules using a nuclear magnetic resonance quantum information processor. 展开更多
