红花提取物磷脂复合物的制备及其透膜性研究

目的优化红花提取物磷脂复合物的制备工艺并研究其透膜性能。方法在单因素实验基础上,利用响应曲面分析法,以红花提取物与磷脂形成的磷脂复合物的复合率为指标,优化制备工艺。利用紫外-可见吸收光谱和红外光谱方法进行表征。采用改良Fr... 目的优化红花提取物磷脂复合物的制备工艺并研究其透膜性能。方法在单因素实验基础上,利用响应曲面分析法,以红花提取物与磷脂形成的磷脂复合物的复合率为指标,优化制备工艺。利用紫外-可见吸收光谱和红外光谱方法进行表征。采用改良Franz扩散池,评价红花提取物及不同药脂比的磷脂复合物的体外透膜性能。结果红花提取物磷脂复合物的最佳制备工艺为以甲醇为复合溶剂,红花提取物的质量浓度为5.0mg/m L,与磷脂质量比为1∶1,反应时间为1.5h,反应温度为55℃。透膜实验结果显示,红花提取物及药脂比分别为2、1、0.5的磷脂复合物24h累积渗透量(Q24)分别为(15.07±1.24)、(15.61±0.92)、(21.94±1.54)、(21.05±1.39)μg/cm2。结论优选的制备工艺合理可行,红花提取物磷脂复合物能明显改善其透膜性能。 展开更多

The heavy metal ATPase(HMA)subfamily is mainly involved in heavy metal(HM)tolerance and transport in plants,but an understanding of the definite roles and mechanisms of most HMA members are still limited.In the present study,we identified 14 candidate HMA genes named BrrHMAl—BrrHMA8 from the turnip genome and analyzed the phylogeny,gene structure,chromosome distribution,and conserved domains and motifs of HMAs in turnip(Brassica rapa var.rapa).According to our phylogenetic tree,the BrrHMAs are divided into a Zn/Cd/Co/Pb subclass and Cu/Ag subclass.The BrrHMA members show similar structural characteristics within subclasses.To explore the roles of BrrHMAs in turnip,we compared the gene sequences and expression patterns of the BrrHMA genes between a Cd-tolerant landrace and a Cd-sensitive landrace.Most BrrHMA genes showed similar spatial expression patterns in both Cd-tolerant and Cd-sensitive turnip landraces;some BrrHMA genes,however,were differentially expressed in specific tissue in Cd-tolerant and Cd-sensitive turnip.Specifically,BrrHMA genes in the Zn/Cd/Co/Pb subclass shared the same coding sequence but were differentially expressed in Cd-tolerant and Cd-sensitive turnip landraces under Cd stress.Our findings suggest that the stable expression and up-regulated expression of BrrHMA Zn/Cd/Co/Pb subclass genes under Cd stress may contribute to the higher Cd tolerance of turnip landraces. 展开更多

为了研究促黄体激素释放激素(LHRH)介导的TAT蛋白核心肽PTD的跨膜作用,测定目的蛋白的转导效率,试验以pET28a—PTD—GFP质粒为模板,设计含LHRH基因序列的特异性PER引物,经PCR扩增后双酶切,并克隆到原核表达载体pET28a中,构建重组质粒并... 为了研究促黄体激素释放激素(LHRH)介导的TAT蛋白核心肽PTD的跨膜作用,测定目的蛋白的转导效率,试验以pET28a—PTD—GFP质粒为模板,设计含LHRH基因序列的特异性PER引物,经PCR扩增后双酶切,并克隆到原核表达载体pET28a中,构建重组质粒并在E.coli BL21(DE3)感受态细胞中诱导表达,超声破碎后进行硫酸铵梯度沉淀,取上清液用Phenyl—HP疏水层析柱和Q阴离子交换层析柱纯化,将纯化后的目的蛋白加到体外培养的HeLa细胞中,观察转导情况,分析目的蛋白浓度对转导效率的影响。结果表明:PER扩增得到目的基因LHRH—PTD-GFP,并成功构建出重组质粒pET28a—LHRH—PTD-GFP,在E.coli BL21(DE3)感受态细胞中成功诱导表达出目的蛋白,经Phenyl—HP疏水层析柱和Q阴离子交换层析柱成功纯化出目的蛋白,在荧光显微镜下HeLa细胞内显现明显的绿色荧光,经分析转导效率与目的蛋白浓度之间呈正相关,回归系数为0.096 6,决定系数(R^2)为0.937 8。说明LHRH介导的TAT蛋白核心肽PTD具有很好的跨膜特性,转导效率对目的蛋白浓度具有依赖性,在一定浓度范围内随着目的蛋白浓度的增加转导效率增大。 展开更多

BACKGROUND Cystic fibrosis(CF)is rare in Asian populations relative to the Caucasian population.In this paper,we report the cystic fibrosis transmembrane conductance regulator(CFTR)variation in a family of Chinese CF patients,and systematically review the previous literature.CASE SUMMARY Here we report a 30-month-old Chinese girl who was diagnosed with CF based on her history and symptoms such as recurrent productive cough,wheezing with repeated infection of Pseudomonas aeruginosa,and parasinusitis.Chest computed tomography(CT)scanning revealed obvious exudative lesions and bilateral bronchiectasis.Liver CT scanning revealed a low-density lesion in the left lobe of the liver.A diagnosis of CF was made based upon CFTR gene tests.The CFTR gene was sequenced using the blood samples of her and her parents and showed a heterozygous novel missense mutation of c.753_754delAG in exon 7.In addition,a heterozygous c.1240 C>T mutation was found in exon 10 of the CFTR.The mutation c.753_754delAG was verified to have been inherited from her mother,and the c.1240 C>T mutation was from her father who was diagnosed with congenital absence of vas deferens.CONCLUSION A novel mutation of CFTR,c.753_754delAG,was found in a Chinese CF child.c.2909G>A is the most common mutation among Chinese CF patients. 展开更多

目的研究具有高对称结构纳米载体的跨膜转运过程,对比不同结构纳米载体的直接跨膜优势。方法使用分子动力学中介观模拟常用的耗散粒子动力学方法,研究具有高对称结构(如正二十面体、正十二面体、截角八面体等)的纳米载体的直接跨膜过程... 目的研究具有高对称结构纳米载体的跨膜转运过程,对比不同结构纳米载体的直接跨膜优势。方法使用分子动力学中介观模拟常用的耗散粒子动力学方法,研究具有高对称结构(如正二十面体、正十二面体、截角八面体等)的纳米载体的直接跨膜过程,并在3个粒径水平对其进行模拟验证。结果纳米载体的形状对跨膜转运过程有非常重要的影响,在几种常见的高对称结构中,正二十面体形状载体具有较好的穿透能力,正二十面体和正十二面体颗粒的易位过程比球形和椭球颗粒明显更流畅迅速;载体的体积效应规律并不明显,在模拟粒径范围内体积可能并不是影响载体易位的直接因素。结论纳米载体形状因素是影响跨膜转运过程的主要因素,粒径因素可能只起到间接的辅助作用。 展开更多

Study on application value of HBV DNA, AFP and GP73 in diagnosis of chronic hepatitis B severity and liver fibrosis

Objective: To explore the application of hepatitis B virus (HBV) DNA, alpha-fetoprotein (AFP), and Golgi transmembrane glycoprotein 73 (GP73) in the diagnosis of chronic hepatitis B (CHB) severity and liver fibrosis. ... Objective: To explore the application of hepatitis B virus (HBV) DNA, alpha-fetoprotein (AFP), and Golgi transmembrane glycoprotein 73 (GP73) in the diagnosis of chronic hepatitis B (CHB) severity and liver fibrosis. Methods: A total of 160 patients with CHB were selected from August 2017 to August 2018. The severity of the disease was classified into mild, moderate, severe and hepatic failure according to the standard of the guidelines for the prevention and treatment of chronic hepatitis B. Fibrosis was classified according to Masson staining, and S2-S4 was fibrosis. The 40 healthy persons in the same period were selected as the control group. The levels of HBV DNA, AFP, and GP73 in peripheral blood of all patients were detected. The healthy person in the same period was selected as the control group. Results: The levels of AFP and GP73 in CHB patients were significantly higher than those in the control group (P<0.05). And with the increase of severity, the levels of AFP and GP73 also increased significantly (P<0.05). With the increase of liver fibrosis in CHB patients, the levels of HBV DNA, AFP and GP73 were significantly increased (P<0.05). There was a significant positive correlation between the severity of AFP, GP73, and CHB and degree of fibrosis (P<0.05). There was a positive correlation between HBV DNA and fibrosis (P<0.05). The AUC of AFP and GP73 on the severity of CHB were 0.711 and 0.729, respectively. The AUC of HBV DNA, AFP, and GP73 on CHB liver fibrosis were 0.534, 0.745, and 0.758, respectively. Conclusions:AFP, GP73, and CHB are positively correlated with severity and fibrosis, and they have certain diagnostic value for CHB disease and fibrosis. HBV DNA also has a certain value for the degree of liver fibrosis in patients with CHB. 展开更多

However,later studies revealed that in addition to the ITIiVI,the transmembrane (TM)domain of FcyRIIB is also indispensable for its inhibitory function.Indeed,recent epidemiological studies revealed that a non-synonymous single nucleotide polymorphism (rs1050501) within the TM domain of FcyRIIB,responsible for the 1232T substitution,is associated with the susceptibility to systemic lupus erythematosus (SLE).In this review, we will summarize these epidemiological and functional studies of FcyRIIB-1232T in the past few years,and will further discuss the mechanisms accounting for the functional loss of FcyRIIB-1232T.Our review will help the reader gain a deeper understanding of the importance of the TM domain in mediating the inhibitory function of FcyRIIB and may provide insights to a new therapeutic target for the associated diseases. 展开更多

Objective:To study the transmembrane transport of chemical components of Chinese herbs and to explore the function of platycodin D (PD) on biomembranes.Methods:Interaction between PD and the dipalmitoylphosphatidylcholine (DPPC) bilayer was reproduced by molecular dynamics simulation with the Martini force field.A model validation and methodological study were first performed,and were based on simulation investigations of transmembrane transport for three herbal compounds with distinct hydrophilic properties.Results:PD increased the mobility of the DPPC bilayer since its aglycone strongly interacted with the hydrophobic layer,which broke the structure of the gate layer,and weakened the ordered performance of hydrophobic tails.Conclusion:The Martini force field was successfully applied to the study of the interaction between herbal compounds and a biological membrane.By combining the dynamics equilibrium morphology,the distribution of drugs inside and outside the biomembrane,and the interaction sites of drugs on the DPPC bilayer,factors influencing transmembrane transport of drugs were elucidated and the function of platycodin D in a biological membrane was reproduced. 展开更多

Structure and Positioning of Three Transmembrane Segments from Slcllal in SDS Micelles

Slcllal is an integral membrane protein with 12 predicted transmembrane domams(TM). In the presentwork, the secondary structures and positioning of the peptides associated with SIclIaI-TM2, TM3 andTM4(wild-type peptides and fimctional mutants) in sodium dodecyl sulfate(SDS) were analyzed using circulardichroism(CD) and fluorescence techniques. The results indicate that TM3 is significantly different in secondarystructure and topology from TM2 and TM4 in SDS. The peptide TM3 is less structured and is embedded in the SDSless deeply than TM2 and TM4 at pH=4, 5.5 and 7. In addition, the position of TM3 is remarkably shifted towards thedirection of the SDS surface with increasing pH, whereas the locations of TM2 and TM4 in SDS are less affected bypH. The E 139A substitution in TM3 significantly impairs the pH dependence of the buried depth of TM3 and causes adecrease in helicity in SDS at the three pH values. The G169D mutation has little effect on the topological arrange-ment of TM4 in SDS. In contrast, TM2 and TM4 are topologically similar. 展开更多

The case of a 32-year-old black woman of African descent who suffered from repeated episodes of acute pancreatitis,initially triggered when flying on airplanes,is reported.She did not drink alcohol or smoke.Genetic analysis was negative for cationic trypsinogen,serine protease inhibitor Kazal type 1 and chymotrypsin C.However,hemoglobin F was elevated.Sequencing of the thalassemia gene revealed a novel alteration in the 5’region indicative of a functional abnormality of the molecule.Sequencing the cystic fibrosis transmembrane conductance regulator(CFTR)gene revealed a heterozygote sequence variant.The combination of a hemoglobin gene mutation known for thalassemia in conjunction with the hitherto undescribed CFTR mutation is suggested to pave the road for initial and repetitive pancreatitis attacks.This will be discussed. 展开更多

Cystic fibrosis,a common autosomal recessive genetic disorder among Caucasians,is caused by defects in the transmembrane conductance regulatory(CFTR)gene.The analysis of CFTR gene mutations is useful to better characterize the disease,and for preconceptional screening,prenatal and preimplantation genetic diagnosis.Here we report the results of a genetic analysis in a 16-year-old boy from southwestern Iran diagnosed as having cystic fibrosis in infancy based on gastrointestinal and pulmonary manifestations,with positive sweat chloride tests.He lacked both normal and mutant forms of the fragment corresponding to theΔF508 allele in initial genetic studies.Multiplex ligationdependent probe amplification-based testing revealed a homozygous deletion spanning exons 4 to 10 of the CFTR gene.We predict an in-frame deletion removing 373 amino acids based on our sequencing results.Determining CFTR gene mutations in patients and their family members would be helpful to prevent the occurrence of new cases,especially in populations in which consanguinity is common. 展开更多

Osteoclasts are the bone resorbing cells essential for bone remodeling.Osteoclasts are formed from hematopoietic progenitors in the monocyte/macrophage lineage.Osteoclastogenesis is composed of several steps including progenitor survival,differentiation to mononuclear pre-osteoclasts,fusion to multi-nuclear mature osteoclasts,and activation to bone resorbing osteoclasts.The regulation of osteoclastogenesis has been extensively studied,in which the receptor activator of NF-κB ligand(RANKL)-mediated signaling pathway and downstream transcription factors play essential roles.However,less is known about osteoclast fusion,which is a property of mature osteoclasts and is required for osteoclasts to resorb bone.Several proteins that affect cell fusion have been identified.Among them,dritic cell-specific transmembrane protein(DC-STAMP)is directly associated to osteoclast fusion in vivo.Cytokines and factors influence osteoclast fusion through regula regulation of DC-STAMP.Here we review the recently discovered new factors that regulate osteoclast fusion with specific focus on DC-STAMP.A better understanding of the mechanistic basis of osteoclast fusion will lead to the development of a new therapeutic strategy for bone disorders due to elevated osteoclast bone resorption.Cell-cell fusion is essential for a variety of cellular biological processes.In mammals,there is a limited number of cell types that fuse to form multinucleated cells,such as the fusion of myoblasts for the formation of skeletal muscle and the fusion of cells of the monocyte/macrophage lineage for the formation of multinucleated osteoclasts and giant cells.In most cases,cellcell fusion is beneficial for cells by enhancing function.Myoblast fusion increases myofiber size and diameter and thereby increases contractile strength.Multinucleated osteoclasts have far more bone resorbing activity than their mono-nuclear counterparts.Multinucleated giant cells are much more efficient in the removal of implanted materials and bacteria due to chronic infection tha 展开更多
