Meanwhile the outbreak of the Covid-19 since December, 2019 in China, it has killed more than a hundred thousand of people of all ages and sex across the globe in a short span of time. On the bases of this study the nearest family member of the virus and its receptor binding domain of S protein including its model structure and function of its active sites were naked through Multiple Sequence Alignment, modelling and molecular docking software accordingly its repository genome databases. The virus was genetically associated and molecular evolutionary related with (<em>RaTG</em>13) and it scores 96.12% homology with 99% query coverage followed by <em>bat-SL-CoVZC</em>45 and<em> bat-SL-CoVZXC</em>21 notch 89.12% and 88.65% respectively. However, SARS and MERS corona type virus those outbreak earlier respectively less likely family members of 2019-nCoV. Though the virus has a close genetic association with those previous SARS coronaviruses, and certainly the spike protein used as a binding receptor to fight against human receptor protein of ACE 2, but on the basis of FRODOC and HDOCK server analysis multi favorable active sites of S protein was discovered such GLN493 shown as a finest key in both model and possessed a unique traits on it resulting unexpected rate of transmission and number of people died while compared to the previous one. TYR500, ASN501, GLN498 and others residues preferably contemplate site also. In particular, the diversity of the virus in the world may be due to the genome structure of the virus and S gene changed over the time, across the world against to host of human genetic diversity, which may be more robust, and may be a new and unique feature. This is because it is characterized close to contact with distance divergence between wild type novel coronavirus which was risen from China against to the genomes from Lebanon, India, Italy, and USA and so on. Protein adsorption onto polymer surfaces is a very complex and ubiquitous phenomenon whose integrated process impacts essential applications in our daily lives such as food packaging materials,health devices,diagnostic tools,and medical products.Increasingly,novel polymer materials with greater chemical intricacy and reduced dimensionality are used for various applications involving adsorbed proteins on their surfaces.Hence,the nature of protein-surface interactions to consider is becoming much more complicated than before.A large body of literature exists for protein adsorption.However,most of these investigations have focused on collectively measured,ensemble-averaged protein behaviors that occur on macroscale and chemically unvarying polymer surfaces instead of direct measurements at the single protein or sub-protein level.In addition,interrogations of protein-polymer adsorption boundaries in these studies were typically carried out by indirect methods,whose insights may not be suitably applied for explaining individual protein adsorption processes occurring onto nanostructured,chemically varying polymer surfaces.Therefore,an important gap in our knowledge still exists that needs to be systematically addressed via direct measurement means at the single protein and sub-protein level.Such efforts will require multifaceted experimental and theoretical approaches that can probe multilength scales of protein adsorption,while encompassing both single proteins and their collective ensemble behaviors at the length scale spanning from the nanoscopic all the way to the macroscopic scale.In this review,key research achievements in nanoscale protein adsorption to date will be summarized.Specifically,protein adsorption studies involving polymer surfaces with their defining feature dimensions and associated chemical partitions comparable to the size of individual proteins will be discussed in detail.In this regard,recent works bridging the crucial knowledge gap in protein adsorption will be highlighted.New findings of intri 展开更多

With increased demand for plant based proteins by the consumers, the food manufacturers appeal for the new plant proteins with predetermined characteristics. This study aims at isolating the protein fraction from jackfruit seeds and characterizing the protein powder for functional and physicochemical properties. The protein part of the seeds was separated through pH treatments and centrifugation process and finally, the concentrate was converted into powder by spray drying method. The functional properties such as solubility, gelling capacity and emulsion properties and the physicochemical properties such as crystallinity, morphology and particle size distribution of the jackfruit seeds protein isolate (JSPI) were studied. The secondary structural elements of JSPI were also determined by Fourier-transform infrared (FTIR) spectroscopy. About 76.89% protein was estimated in the prepared JSPI with 78.44% solubility in an aquatic solvent. The least gelation concentration of JSPI was 12% in a salt solution. The pH of the solvent significantly affected the emulsifying and foaming properties. The protein isolate possessed amorphous structure, moderate bulk density and almost 75% of the particles fell in a similar size distribution range. The conformational study reported that the β-sheet is the dominant secondary structural element with the highest content of 50.28%. The observed features suggest that the JSPI holds satisfactory functional and physicochemical characteristics for being used in protein-enriched foods. 展开更多

目的:研究蛋白Z(PZ)、蛋白Z依赖蛋白酶抑制剂(ZPI)及其他凝血指标[蛋白C(PC)、蛋白S(PS)]与妊娠期高血压疾病(HDP)的相关性。方法:收集2017年6月2018年12月在郑州大学第三附属医院产科收治的HDP患者60例作为研究对象(HDP组),其中妊娠期... 目的:研究蛋白Z(PZ)、蛋白Z依赖蛋白酶抑制剂(ZPI)及其他凝血指标[蛋白C(PC)、蛋白S(PS)]与妊娠期高血压疾病(HDP)的相关性。方法:收集2017年6月2018年12月在郑州大学第三附属医院产科收治的HDP患者60例作为研究对象(HDP组),其中妊娠期高血压组(25例)、子痫前期组(PE组,35例),另外选取健康孕妇50例作为正常妊娠组,未孕妇女30例作为正常未孕组。比较各组血清PZ、ZPI及血浆PS、PC水平,研究其与HDP的相关性,并采用受试者工作特征(ROC)曲线分析其对HDP的诊断价值。结果:①HDP组的血清PZ、ZPI水平均低于正常妊娠组(P<0.05),正常妊娠组高于正常未孕组(P<0.05),血清PZ、ZPI水平与HDP呈负相关(r分别为-0.687和-0.444,P<0.05),ROC曲线下面积分别为0.901(95%CI:0.853~0.948)、0.759(95%CI:0.676~0.841),最佳诊断截断值分别为31.52 ng/mL和14.54 ng/mL。②血浆PS、PC在HDP组低于正常妊娠组和正常未孕组(P<0.05),正常妊娠组与正常未孕组相比差异无统计学意义(P>0.05),血浆PC水平与HDP呈负相关(r为-0.304,P=0.000),PC对HDP诊断的最佳截断值为108.08%,AUC为0.677(95%CI:0.586~0.769)。血浆PS水平与HDP无相关性(P>0.05)。③HDP组2个亚组相比,PE组血清PZ、ZPI水平均低于妊娠期高血压组(P<0.05),而2组血浆PS、PC水平比较差异无统计学意义(P>0.05)。结论:检测妊娠期高血压疾病患者血清中PZ、ZPI及血浆中PS、PC水平,可以及早地发现凝血指标的异常,进行早期干预治疗,最大程度减少HDP的并发症。 展开更多

A large number of chemokines,cytokines,other trophic factors and the extracellular matrix molecules form a favorable microenvironment for peripheral nerve regeneration.This microenvironment is one of the major factors for regenerative success.Therefore,it is important to investigate the key molecules and regulators affecting nerve regeneration after peripheral nerve injury.However,the identities of specific cytokines at various time points after sciatic nerve injury have not been determined.The study was performed by transecting the sciatic nerve to establish a model of peripheral nerve injury and to analyze,by protein microarray,the expression of different cytokines in the distal nerve after injury.Results showed a large number of cytokines were up-regulated at different time points post injury and several cytokines,e.g.,ciliary neurotrophic factor,were downregulated.The construction of a protein-protein interaction network was used to screen how the proteins interacted with differentially expressed cytokines.Kyoto Encyclopedia of Genes and Genomes pathway and Gene ontology analyses indicated that the differentially expressed cytokines were significantly associated with chemokine signaling pathways,Janus kinase/signal transducers and activators of transcription,phosphoinositide 3-kinase/protein kinase B,and notch signaling pathway.The cytokines involved in inflammation,immune response and cell chemotaxis were up-regulated initially and the cytokines involved in neuronal apoptotic processes,cell-cell adhesion,and cell proliferation were up-regulated at 28 days after injury.Western blot analysis showed that the expression and changes of hepatocyte growth factor,glial cell line-derived neurotrophic factor and ciliary neurotrophic factor were consistent with the results of protein microarray analysis.The results provide a comprehensive understanding of changes in cytokine expression and changes in these cytokines and classical signaling pathways and biological functions during Wallerian degeneration,as well as a bas 展开更多

为了评价传统风干牦牛肉在加工过程中肌原纤维蛋白消化率的变化,并探讨蛋白质氧化影响其消化性的潜在机制。在牦牛肉自然风干加工过程中(40 d)采集样本,测定肌原纤维蛋白羰基、巯基、二硫键、二聚酪氨酸、粒径分布、体外消化率及脂质氧... 为了评价传统风干牦牛肉在加工过程中肌原纤维蛋白消化率的变化,并探讨蛋白质氧化影响其消化性的潜在机制。在牦牛肉自然风干加工过程中(40 d)采集样本,测定肌原纤维蛋白羰基、巯基、二硫键、二聚酪氨酸、粒径分布、体外消化率及脂质氧化等指标,并分析脂质氧化和蛋白质氧化对蛋白质消化性的影响。结果显示,脂质初级氧化产物(Primary Oxidation Value,POV)、硫代巴比妥酸反应物(Thiobarbituric Acid Reaction Substrates,TBARS)和蛋白质羰基化合物随加工时间的延长而显著增加(P<0.05),并且相互间存在极显著相关性(P<0.01)。此外,二硫键和二聚酪氨酸含量显著增加(P<0.05),这是蛋白质交联的主要形式。蛋白质被胃蛋白酶和胰蛋白酶总的水解率在整个加工过程中降低了19.58%(P<0.05),蛋白质的水解率与羰基、二硫键、二聚酪氨酸和蛋白质粒径呈现极显著负相关性(P<0.01)。综上所述,在风干牛肉加工期间,脂质氧化产物促进了蛋白质氧化,蛋白质的羰基化和交联导致蛋白酶对蛋白质的水解率降低。研究结果对了解风干牦牛肉传统加工方式和进一步改进加工工艺具有积极的促进作用。 展开更多

In plants,RNA interference(RNAi)plays a pivotal role in growth and development,and responses to environmental inputs,including pathogen attack.The intercellular and systemic trafficking of small interfering RNA(siRNA)/microRNA(miRNA)is a central component in this regulatory pathway.Currently,little is known with regards to the molecular agents involved in the movement of these si/miRNAs.To address this situation,we employed a biochemical approach to identify and characterize a conserved SMALL RNA-BINDING PROTEIN 1(SRBP1)family that mediates non-cell-autonomous small RNA(sRNA)trafficking.In Arabidopsis,AtSRBP1 is a glycine-rich(GR)RNA-binding protein,also known as AtGRP7,which we show binds single-stranded siRNA.A viral vector,Zucchini yellow mosaic virus(ZYMV),was employed to functionally characterized the AtSRBP1-4(AtGRP7/2/4/8)RNA recognition motif and GR domains.Cellular-based studies revealed the GR domain as being necessary and sufficient for SRBP1 cell-to-cell movement.Taken together,our findings provide a foundation for future research into the mechanism and function of mobile sRNA signaling agents in plants. 展开更多

The objective of this study was to evaluate the effect of rice bran quality and dietary supplement source on growing pig performance. A total of 60 castrated male pigs (Yorkshire × Duroc, initially 16.1 kg) were used for 52-day trial. Pigs were allotted to pens in a completely randomized design (4 pigs per pen) and assigned to four dietary treatments in a 2 × 2 factorial with factors being rice bran source (high or low-quality) and source of protein, vitamins, and minerals (supplement or soybean meal and base mix). Analyzed protein and fat level for the high- and low-quality rice bran was 11.07% and 5.86% and 11.20% and 4.50%, respectively. Crude fiber content for high- and low-quality rice bran was 13.44% and 27.79%. Diets were formulated in two phases (phase 1: 16 to 27 and phase 2: 27 to 48 kg) to adjust to the pig’s nutrient requirements. There was no evidence for an interaction (P > 0.05) between rice bran quality × supplement type or main effect of supplement type (soybean meal + base mix vs protein supplement) for final body weight, average daily gain (ADG), average daily feed intake (ADFI), and feed efficiency (F/G). However, pigs fed high-quality rice bran had improved (P < 0.001) ADG and F/G and lower ADFI compared to pigs fed low protein rice bran. In conclusion, rice bran quality as measured by protein, crude fiber, and fat influenced growing pig performance, while the type of dietary protein supplement did not. Abnormal protein expression or activities are associated with many diseases,especially cancer.Therefore,down-regulating the proteins involved in cancer cell survival proved to be an effective strategy for cancer treatment—a number of drugs based on proteolysis-targeting chimaera(PROTAC)mechanism have demonstrated clinical efficacy.Recent progress in the PROTAC strategy includes identification of the structure of the first ternary eutectic complex,extra-terminal domain-4-PROTAC-VonHippel-Lindau(BRD4-PROTAC-VHL),and PROTAC ARV-110 has entered clinical trials for the treatment of prostate cancer in 2019.These discoveries strongly proved the value of the PROTAC strategy.In this review,we summarize recent meaningful research of PROTACs,including the molecular design and optimization strategy as well as clinical application of candidate molecules.We hope to provide useful insights for rational design of PROTACs. 展开更多

Kolbroek pigs have numerous advantages as an indigenous breed including its contributions to food security. However, there are numerous concerns over the current feed regime that compromises its growth performance and carcass quality. Therefore, this study was conducted to evaluate the growth performance traits, attainment of puberty, backfat thickness (BFT), and carcass traits when boar fed diets supplemented with 10% (standard diet), 13% and 16% crude protein. Fifteen pigs per protein diets were individually housed and fed <em>ad-libitum</em> for eight weeks in a complete randomised design. Average daily gain (ADG), average daily feed intake (ADFI) and feed conversion ratio (FCR) were measured on a weekly basis. At the end of the trial, all boars were slaughtered and carcass quality parameters measured. Pearson’s correlation coefficients were calculated between growth performance parameters. Our data revealed that there was an increase in final body weight, ADG, ADFI accompanied by decreases in BFT and the age of attainment of puberty. There was also a positive correlation between ADFI and ADG (r = 0.78) accompanied by a low positive correlation between ADG and BFT (r = 0.12) with a high negative correlation between FCR and ADG (r = -0.94). The water holding capacity (WHC), dressing percentage and marbling mass appeared to decrease while the eye muscle area increased with protein supplementation of the diet. There were also increases in the lungs, liver, small plus large intestines and stomach with no changes in the pancreas and heart tissues. Accompanying these was observations that the meat colour lightness improved accompanied by a decline in meat redness and yellowness. In conclusion, supplementation of boar diet with increasing crude protein improved the carcass quality and growth performance of Kolbroek boars. 展开更多

Background:This study was conducted to determine protein molecular structure profiles and quantify the relationship between protein structural features and protein metabolism and bioavailability of blend pel eted products(BPP)based on co-products(canola or carinata)from processing with different proportions of pulse pea screenings and lignosulfonate chemical compound.Method:The protein molecular structures were determined using the non-invasive advanced vibrational molecular spectroscopy(ATR-FT/IR)in terms of chemical structure and biofunctional groups of amides(ⅠandⅡ),α-helix andβ-sheet.Results:The results showed that increasing the level of the co-products in BPP significantly increased the spectral intensity of the amide area and amide height.The products exhibited similar protein secondaryα-helix toβ-sheet ratio.The protein molecular structure profiles(amidesⅠandⅡ,α-helix toβ-sheet)were highly associated with protein degradation kinetics and intestinal digestion.In conclusion,the non-invasive vibrational molecular spectroscopy(ATR-FT/IR)could be used to detect inherent structural make-up characteristics in BPP.Conclusion:The molecular structural features related to protein biopolymer were highly associated with protein utilization and metabolism. 展开更多

PP2C (2C type protein phosphatase)是一类广泛存在于生物体中、具有重要调控作用的蛋白磷酸酶,在植物抵抗逆境胁迫及激素响应等过程中具有重要作用,而在毛竹(Phyllostachys edulis)中的研究尚未充分开展。本研究以毛竹为材料,利用生物信息学方法,从毛竹全基因组中鉴定出125个具有完整PP2C保守结构域的基因,这些基因编码蛋白长度为110~1 086 aa,分子量为12.07~121.02 kD;系统进化分析表明,125个毛竹PP2C基因共可分为9个亚家族,其中4个基因(PH02Gene36278.t1, PH02Gene48541.t1,PH02Gene37946.t1, PH02Gene29918.t1)未被分入任何亚家族,而是单独形成分枝;基于转录组数据的组织特异性分析显示,121个PP2C基因在毛竹不同组织及不同发育阶段中都有不同程度的表达,多数PP2C基因在毛竹笋的不同发育阶段呈现较高的表达,也有部分PP2C基因在根与叶片等部位具有较高的表达;qRT-PCR结果显示,毛竹经过脱落酸处理后有9个PP2C基因上调表达、1个下调表达;在盐和茉莉酸甲酯处理后,各有6个基因上调表达、5个下调表达,说明PP2C基因在参与毛竹非生物胁迫及不同激素调控通路时可能发挥不同的作用。本研究可为毛竹PP2C基因家族功能的进一步研究提供参考依据。 展开更多

目的比较子痫前期(PE)与健康孕妇血浆游离蛋白S(fPS)、蛋白C (PC)、抗凝血酶Ⅲ(ATⅢ)活性的水平,探究孕妇血浆fPS、PC及ATⅢ活性改变是否与子痫前期发病有关。方法采用随机排列表抽取2018年1月至2019年4月郑州大学第一附属医院定期产前... 目的比较子痫前期(PE)与健康孕妇血浆游离蛋白S(fPS)、蛋白C (PC)、抗凝血酶Ⅲ(ATⅢ)活性的水平,探究孕妇血浆fPS、PC及ATⅢ活性改变是否与子痫前期发病有关。方法采用随机排列表抽取2018年1月至2019年4月郑州大学第一附属医院定期产前检查、临床资料完整的PE患者89例,其中早发型PE组(<34周)53例,晚发型PE组(≥34周)36例。抽取同期本院定期产前检查74例健康孕妇为对照组,其中<34周对照组30例,≥34周对照组44例。采用凝固法检测血浆fPS活性,发色底物法检测血浆PC、ATⅢ活性,比较孕妇血浆fPS、PC、ATⅢ活性。结果①早发型PE组、<34周对照组、晚发型PE组、≥34周对照组孕妇血浆fPS活性分别为(44.10±1.8)%、(49.7±1.8)%、(42.3±3.0)%、(46.7±1.7)%,PC活性分别为(116.0±3.8)%、(111.4±2.7)%、(107.8±3.7)%、(109.4±2.9)%,ATⅢ活性分别为(72.1±1.7)%、(95.0±3.0)%、(75.0±2.4)%、(91.1±2.5)%,早发型PE组较与<34周对照组孕妇血浆fPS、ATⅢ活性均降低,(P均<0.05)。②晚发型PE组孕妇血浆fPS、ATⅢ活性与≥34周对照组均降低(P均<0.05)。③早发型PE组孕妇血浆fPS、PC及ATⅢ活性与晚发型PE组比较,差异均无统计学意义(P均>0.05);结论孕妇血浆fPS、ATⅢ活性降低可能与PE发病有关;早发型、晚发型PE患者体内可能均存在血浆fPS、ATⅢ活性降低引发的易栓状态。 展开更多
