In recent years,with the continuous improvement and development of molecular technology in the application process,different types of DNA molecular markers have made rapid progress in the study of genetic diversity of rapeseed. The study of genetic diversity is conducive to the correct formulation of the strategy of collecting and in situ preservation of genetic resources of rapeseed,and it is the genetic basis for the improvement of rapeseed varieties. This article mainly starts with the three DNA molecular markers( SSR,RAPD,AFLP) widely used in the study of genetic diversity of rapeseed. By introducing the application principles and characteristics of SSR,RFPD and AFLP molecular markers,research progress of these three marker technologies in genetic diversity of rapeseed is briefly described. 展开更多

Long non-coding RNAs(lncRNAs)are members of the non-protein coding RNA family longer than 200 nucleotides.They participate in the regulation of gene and protein expression influencing apoptosis,cell proliferation and immune responses,thereby playing a critical role in the development and progression of various cancers,including colorectal cancer(CRC).As CRC is one of the most frequently diagnosed malignancies worldwide with high mortality,its screening and early detection are crucial,so the identification of disease-specific biomarkers is necessary.LncRNAs are promising candidates as they are involved in carcinogenesis,and certain lncRNAs(e.g.,CCAT1,CRNDE,CRCAL1-4)show altered expression in adenomas,making them potential early diagnostic markers.In addition to being useful as tissue-specific markers,analysis of circulating lncRNAs(e.g.,CCAT1,CCAT2,BLACAT1,CRNDE,NEAT1,UCA1)in peripheral blood offers the possibility to establish minimally invasive,liquid biopsy-based diagnostic tests.This review article aims to describe the origin,structure,and functions of lncRNAs and to discuss their contribution to CRC development.Moreover,our purpose is to summarise lncRNAs showing altered expression levels during tumor formation in both colon tissue and plasma/serum samples and to demonstrate their clinical implications as diagnostic or prognostic biomarkers for CRC. 展开更多

As an excellent local crop variety of Huanggang,"Huangzhou radish" has been widely promoted and its market influence is increasing day by day.Recently,the purity of "Huangzhou radish" has declined deeply,so it is urgent to improve and strengthen the purity and viability of "Huangzhou radish" including bad phenomena such as degeneration of varieties after self-pollination.At present,there was no SSR molecular marker can be used for genetic diversity identification of "Huangzhou radish".SSR molecular marker can provide a favorable tool to study "Huangzhou radish".In this study,"Xiakang","Hongbao"and "Huangzhou radish" were used as experimental materials,100 SSR primers were screened,and 25 pairs of SSR primers were selected.Finally,two pairs of primers were selected for identification of the purity of "Huangzhou radish" by non-denatured polyacrylamide gel electrophoresis (PAGE).The results showed that R-102 and R-112 primer pairs could amplify the special bands for "Huangzhou radish" from above three kinds of radishes.These two pairs of primers could be developed as SSR markers to identify the specificity and purity of "Huangzhou radish".Furthermore,this study provides convenient conditions for promoting the economic and social value of "Huangzhou radish". 展开更多

外周血淋巴细胞circRNA在脊柱结核患者中表达的初步筛选及功能分析 预览

目的:探寻对脊柱结核有诊断及治疗价值的特异性环状RNA(circRNA)及分析其功能。方法:选取2016年9月至2018年9月广西医科大学第一附属医院收治的25例脊柱结核患者作为实验组,25例健康志愿者作为对照组,并从中随机抽取3例脊柱结核患者和3... 目的:探寻对脊柱结核有诊断及治疗价值的特异性环状RNA(circRNA)及分析其功能。方法:选取2016年9月至2018年9月广西医科大学第一附属医院收治的25例脊柱结核患者作为实验组,25例健康志愿者作为对照组,并从中随机抽取3例脊柱结核患者和3例健康志愿者,分别提取外周血淋巴细胞中总RNA,进行circRNA基因芯片检测,筛选出脊柱结核患者和健康志愿者中差异表达的circRNAs,随后提取两组外周血淋巴细胞总RNA,并用RT-PCR方法验证所筛选的部分差异circRNA分子的表达情况。结果:通过circRNA基因芯片初步筛选出89个表达有差异的circRNAs,其中46个表达上调,43个表达下调。挑选在实验组中的上调和下调表达差异倍数最大的各5条circRNA进行RT-PCR验证,结果显示,与对照组相比,实验组中hsa_circRNA_100632、hsa_circRNA_103747、hsa_circRNA_102051、hsa_circRNA_104375、hsa_circRNA_405571表达上调,hsa_circRNA_104791、hsa_circRNA_400770、hsa_circRNA_403882、hsa_circRNA_104936、hsa_circRNA_404758表达下调,与基因芯片检测结果一致。结论:脊柱结核患者外周血淋巴细胞中存在一些特异性的circRNA表达,而且其有可能对脊柱结核的诊断及治疗有重要的意义。 展开更多

Insect body pigmentation and coloration are critical to adaption to the environment. To explore the mechanisms that drive pigmentation, we used the clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 (Cas9) genome editing system to target the ebony gene in the non-model insect Spodoptera litura. Ebony is crucial to melanin synthesis in insects. By directly injecting Cas9 messenger RNA and ebony-specific guide RNAs into S. litura embryos, we successfully induced a typical ebony-deficient phenotype of deep coloration of the puparium and induction of melanin formation during the pupal stage. Polymerase chain reaction-based genotype analysis demonstrated that various mutations had occurred at the sites targeted in ebony. Our study clearly demonstrates the function of ebony in the puparium coloration and also provides a potentially useful marker gene for functional studies in S. litura as well as other lepidopteran pests. 展开更多

As the most important complementary medication against a variety of diseases,traditional Chinese medicines(TCMs)have been extensively applied over thousands of years.Current quality control of herbal medicines,however,is in great dispute.Unlike chemical drugs,which have clear and validated quality standards,the content of only one(or a few)compounds of many herbs and preparations is currently assessed as an indicator of quality,even though the assessed compound(s)is neither closely associated with the efficacy nor representative of the medicine as a whole.Based on the clinical use,compatibility of multiple component prescriptions,and manufacturing process of TCM,the new concept of a TCM quality marker that was proposed in previous work is discussed further here.In addition,practical technological approaches are described for the qualitative analysis and quantification of TCMs including herbs,processed products,and preparations,which lead to the discovery and identification of specific chemicals as quality markers and new quality control patterns.The progress that has been made in TCM quality control is also addressed.This work provides useful information for the quality control of herbal medicines in the future. 展开更多

目的探讨CellSearch系统与单标志物[上皮细胞黏附分子(EpCAM)]实时定量聚合酶链反应(qRT-PCR)平台、多标志物[EpCAM、CD133、CD90、细胞角蛋白(CK19)]qRT-PCR平台检测肝细胞肝癌(HCC)患者外周血循环肿瘤细胞(CTC)的阳性率与相关性。方法... 目的探讨CellSearch系统与单标志物[上皮细胞黏附分子(EpCAM)]实时定量聚合酶链反应(qRT-PCR)平台、多标志物[EpCAM、CD133、CD90、细胞角蛋白(CK19)]qRT-PCR平台检测肝细胞肝癌(HCC)患者外周血循环肿瘤细胞(CTC)的阳性率与相关性。方法 分别采用CellSearch系统、单标志物qRT-PCR平台和多标志物qRT-PCR平台检测32例HCC患者外周血CTC并计算阳性率。采用Spearman相关分析评估各方法间的相关性。结果 单标志物(EpCAM)qRT-PCR平台检测CTC的阳性率为46.9%(15/32),多标志物(EpCAM、CD133、CD90、CK19)qRT-PCR平台检测CTC的阳性率为65.6%(21/32);CellSearch系统检测CTC的阳性率为28.1%(9/32)。单标志物qRT-PCR平台与CellSearch系统的阳性符合率为75.0%(24/32,P=0.002),多标志物qRT-PCR平台与CellSearch系统的阳性符合率为56.3%(18/32,P=0.034),单标志物qRT-PCR平台与多标志物qRT-PCR平台的阳性符合率为81.3%(26/32,P=0.000)。采用多标志物qRT-PCR平台检测CTC时,不同患者4项标志物所占百分比各不相同,有明显异质性。结论多标志物qRT-PCR平台能明显提高HCC患者外周血CTC的阳性检出率,可更直观地反映肿瘤的异质性,适用于多样化的检测需求。 展开更多

As an importantwild relative ofwheat,Agropyron cristatumhas been successfully used forwheat improvement.Currently,a few useful agronomic traits of A.cristatum,such as high grain number per spike and resistance to diseases,have been transferred into common wheat.However,the effective detection of small A.cristatum segmental introgressions in common wheat is still difficult.The objective of this study was to identify A.cristatum-specific single nucleotide polymorphisms (SNPs) for the detection of small alien segments in wheat.The transcriptome sequences of A.cristatum were aligned against wheat coding DNA sequences (CDS) for SNP calling.As a result,we discovered a total of 167,613 putative SNPs specific to the P genome of A.cristatum compared with the common wheat genomes.Among 230 selected SNPs with functional annotations related to inflorescence development and stress resistance,68 were validated as P genome-specific SNPs inmultiple wheat backgrounds using Kompetitive Allele Specific PCR (KASP) assays.Among them,55 SNPs were assigned to six homoeologous groups of the P genome using wheat-A.cristatum addition lines,and 6P-specific SNP markers were further physically mapped on different segments of chromosome 6P in 6P translocation lines.The P genome-specific SNPs were also validated by Sanger sequencing and used to detect the P chromatin in wheat-A.cristatum cryptic introgression lines.Two SNP markers (Unigene20217-182 and Unigene20307-1420) were detected in two wheat-A.cristatum introgression lines that showed enhanced grain number per spike and high resistance to powdery mildew.Together,thedeveloped P genome-specific SNPmarkers will accelerate thedetection of large numbers of wheat-A.cristatum derivatives andwill be helpful formarker-assisted transfer of desirable traits from A.cristatum into adapted wheat cultivars in wheat breeding programs. 展开更多

目的探讨胫骨骨折髓内钉固定患者围术期血清Ⅰ型胶原氨基端末肽(NTX)、碱性磷酸酶(ALP)、人胰岛素生长因子-1(IGF-1)的变化对术后骨不连的预测价值。方法湖北省随州市中心医院(湖北医药学院附属随州医院)2015年1月-2017年1月收治71例胫... 目的探讨胫骨骨折髓内钉固定患者围术期血清Ⅰ型胶原氨基端末肽(NTX)、碱性磷酸酶(ALP)、人胰岛素生长因子-1(IGF-1)的变化对术后骨不连的预测价值。方法湖北省随州市中心医院(湖北医药学院附属随州医院)2015年1月-2017年1月收治71例胫骨骨折患者,其中男性38例,女性33例;年龄21~65岁,平均37.8岁。均采用髓内钉固定治疗。分析患者术前、术后1、4周的血清NTX、ALP、IGF-1水平,记录2015年1月-2018年1月患者骨不连发生情况,并比较有骨不连患者与无骨不连患者围手术期血清NTX、ALP、IGF-1水平,用受试者工作绘制特征(ROC)曲线分析各指标对术后骨不连的预测价值。结果71例患者术后1、4周血清NTX、ALP及IGF-1水平均高于术前,差异有统计学意义(P<0.05),术后1年内骨不连发生率为21.13%(15/71)。术前,术后1、4周有骨不连的患者血清NTX、ALP及IGF-1水平均低于无骨不连者(P<0.05)。ROC曲线显示,胫骨骨折髓内钉患者术前,术后1、4周血清NTX、ALP及IGF-1水平对术后骨不连均有一定预测价值,其中术后4周血清NTX、ALP及IGF-1联合曲线下面积高于术后1周及术前(P<0.05)。结论胫骨骨折髓内钉固定患者围术期血清NTX、ALP及IGF-1对术后骨不连有一定预测价值,可能为早期评估术后骨不连的参考指标,进一步指导临床治疗。 展开更多

Application of Fingerprint Combined with QAMS in Quality Evaluation of Zhenrongdan Mixture 预览

[Objectives] The research aimed to evaluate the quality of Zhenrongdan mixture by fingerprint combining QAMS.[Methods] The quality evaluation method was established and validated with echinacoside as internal reference to determine the contents of other components(ferulic acid, salvianolic acid B, and icariin) according to the relative correction factor. The accuracy and feasibility of QAMS were evaluated by comparison on the results between the measured value and calculation value by external standard method and QAMS.[Results] A common pattern of characteristic fingerprint of Zhenrongdan mixture by HPLC was established. Thirteen common peaks were identified, and they account for 91% of the total peak area, and four components were verified in five batches of Zhenrongdan mixture. Good similarities with correlation coefficients higher than 0.99 were found in the fingerprints. There was no significant difference between the quantitative results of the four ingredients in the five batches by external standard method and QAMS.[Conclusions] The method of fingerprint combined with QAMS could be used for the quality control of multiple components determination and fingerprint chromatography for Zhenrongdan mixture. 展开更多

Recently, a novel green fluorescent protein eYGFPuv has been identified in the marine organism Chiridius poppei which displays high fluorescence intensity and can be visible by eyes in dark. Although strong green fluorescence was achieved in transgenic petunia, 3 expression cassettes (about 8 kb) complicate its application. In this study, to confirm whether 1 expression cassette could be used as a transgenic marker in prokaryotes and eukaryotes, eYGFPuv was cloned into prokaryotic expression vector pET28α-eYGFPuv- His and plant binary expression vector 35S::eYGFPuv. Compared to EGFP, eYGFPuv protein exhibited stronger dazzling green fluorescence in E. coli under excited light at 365 nm and maintains steadily over a long period of time without degradation. When transiently expressed in tobacco leaves, eYGFPuv protein displayed strong green fluorescence. Moreover, the fluorescence of eYGFPuv protein also could be directly observed in living plant, and thus can be used easily as a marker to screen transformed lines in transgenic research. Overall, compared to previous studies on eYGFPuv tandem repeats, our data confirmed that single eYGFPuv sequence still possesses high fluorescence intensity and quenching resistance. Furthermore, because of small size of expression cassette,it is suitable for efficient transformation in both prokaryotic and eukaryotic organisms. 展开更多

For these lesions, it is imperative to identify mucinous cysts (intraductal papillary mucinous neoplasms and mucinous cystic neoplasms) due to the risk of their progression to malignancy. However, no single imaging modality alone is sufficient for a definitive diagnosis of all PCLs. The cyst fluid obtained by endoscopic ultrasound-guided fine needle aspiration provides additional information for the differential diagnosis of PCLs. Current recommendations suggest sending cyst fluid for cytology evaluation and measurement of carcinoembryonic antigen (CEA) levels. Unfortunately, the sensitivity of cytology is greatly limited, and cyst fluid CEA has demonstrated insufficient accuracy as a predictor of mucinous cysts. More recently, cyst fluid glucose has emerged as an alternative to CEA for distinguishing between mucinous and nonmucinous lesions. 目的 分析甲状腺癌病理分化类型与EB病毒感染及感染标志物水平的关联性,阐明感染性疾病在恶性肿瘤发生和发展中的作用机制。方法 选取2017年2月-2018年2月80例医院收治的甲状腺癌患者作为研究组,选取同期80例甲状腺腺瘤患者作为对照组,... 目的 分析甲状腺癌病理分化类型与EB病毒感染及感染标志物水平的关联性,阐明感染性疾病在恶性肿瘤发生和发展中的作用机制。方法 选取2017年2月-2018年2月80例医院收治的甲状腺癌患者作为研究组,选取同期80例甲状腺腺瘤患者作为对照组,对两组患者的EB病毒感染率、血清白介素-6(IL-6)、C-反应蛋白(CRP)及外周血白细胞计数(WBC)水平进行观察和比较。结果 研究组患者EB病毒感染率及血清IL-6、CRP和WBC水平分别为46.25%、(0.56±0.18)μg/L、(5.26±0.71)mg/L、(7.65±2.82)×109/L,均高于对照组(P<0.05)。乳头状癌、滤泡状癌患者血清IL-6、CRP和WBC水平分别为(0.81±0.23)μg/L、(7.81±0.96)mg/L、(9.25±3.16)×109/L;(0.82±0.22)μg/L、(7.76±0.98)mg/L、(9.56±4.63)×109/L,高于髓样癌、未分化癌患者(P<0.05)。合并EB病毒感染甲状腺癌患者血清IL-6、CRP和WBC水平分别为(0.95±0.21)μg/L、(9.24±1.56)mg/L、(10.06±4.18)×109/L,均高于未合并EB感染患者(P<0.05)。结论 甲状腺癌患者的EB病毒感染率和感染标志物水平均高于良性甲状腺肿瘤患者,EB病毒感染可能与甲状腺癌患者的炎症反应具有关联性,某些病理分化类型甲状腺癌患者的EB病毒感染率较高,但尚未观察到与其他病毒类型存在显著的差异,还需要进一步的研究予以证实和讨论。 展开更多
