蛋白质在溶液中是多种构象并存的一个热力学系统,不同的构象以一定的概率在不同的态之间互相转变。作为一个热力学体系,不同构象存在的概率受到热力学变量的影响,压强作为一个基本的热力学变量,其变化将影响到蛋白质的构象分布和动力学... 蛋白质在溶液中是多种构象并存的一个热力学系统,不同的构象以一定的概率在不同的态之间互相转变。作为一个热力学体系,不同构象存在的概率受到热力学变量的影响,压强作为一个基本的热力学变量,其变化将影响到蛋白质的构象分布和动力学行为。压强的增大通常会减小蛋白质的体积,从而可以通过增大压强开展对常压下难以观察的构象进行研究,这些构象对于揭示蛋白质的生物学功能具有重要意义。以朊蛋白为研究对象,通过分子动力学模拟方法研究高压诱导下蛋白质的构象变化。研究结果表明:水分子在高压下的排列趋向于有序,水分子的有序排列直接影响到与水分子有密切接触残基的动力学行为;高压下水分子能够渗入二级结构的疏水核心,破坏β片层的二级结构。揭示高压诱导下朊蛋白构象转化的分子机制,为蛋白质的高压变性研究提供理论依据。 展开更多

【目的】探讨高钙高蛋白日粮对海兰褐蛋鸡肝脏的影响。【方法】将80只40日龄海兰褐蛋鸡随机均分为4组,1组饲喂基础日粮(对照组),2、3和4组分别饲喂高钙、高蛋白和高钙+高蛋白饲料(试验组)。试验第95天,各组随机取6只鸡采集血清,检测生... 【目的】探讨高钙高蛋白日粮对海兰褐蛋鸡肝脏的影响。【方法】将80只40日龄海兰褐蛋鸡随机均分为4组,1组饲喂基础日粮(对照组),2、3和4组分别饲喂高钙、高蛋白和高钙+高蛋白饲料(试验组)。试验第95天,各组随机取6只鸡采集血清,检测生化指标,随后扑杀,采集肝脏组织检测黄嘌呤氧化酶活性(XOD)、白细胞介素1(IL-1)、白细胞介素6(IL-6)和肿瘤坏死因子α(TNF-α),并将肝脏包埋切片进行HE和油红染色。【结果】各试验组与对照组相比,血清尿酸水平和肝中XOD活性升高;天门冬氨酸氨基转移酶、丙氨酸氨基转移酶升高,2、4组的总蛋白、白蛋白和球蛋白升高;总胆红素、直接胆红素和间接胆红素降低;高密度脂蛋白、低密度脂蛋白和胆固醇降低,甘油三酯升高;肝中IL-1、IL-6和TNF-α升高,而高密度脂蛋白、低密度脂蛋白、胆固醇、甘油三酯与对照组相近。显微镜下观察发现:肝内有坏死灶和炎性细胞浸润,肝细胞索排列紊乱,肝细胞空泡变性,少数鸡肝细胞内有脂肪滴。【结论】饲喂高钙和/或高蛋白饲料使鸡肝的结构和功能不同程度受损,高钙+高蛋白组受损更为严重。 展开更多

Objectives To evaluate the distribution by age and sex of serum high-sensitivity C-reactive protein(hsCRP)in an urban Chinese population and to provide a profile prediction for the risk of bacterial infection,inflammatory diseases,or tissue damages in the body.Methods Serum hsCRP was determined using the Roche Tina-quant immuno-turbidimetric assay on a Hitachi 7600–010 automatic biochemical analyzer(Roche Diagnostics)in 1,572 males and 1,800 females,including 78 pregnant women,who were derived from the National Health and Nutrition Survey in2010–2012.Results The average hsCRP concentration in urban China was 0.68 mg/L for males and 0.65 mg/L for females.Significant differences in hsCRP were found among different age groups(P<0.05).Monitoring results showed no significant differences among the 6–11,45–59,and≥60-year-old groups in the comparison of hsCRP between males and females in large cities.However,hsCRP concentration was significantly higher in men aged 12–17 and 18–44 years than in women.Conclusion The distribution of the hsCRP status of residents in large cities in China was influenced by age and gender,and the hsCRP levels of both sexes increased gradually with age.In addition,hsCRP concentration was higher in healthy pregnant women than in non-pregnant women.Basing on our results,we recommend that this parameter be included in future national and international screening for early detection of various illnesses. 展开更多

Fertile chicken eggs were used as an alternative model for large animals to evaluate suspect toxic dietary ingredients for fetal loss disorders associated with mare reproductive loss syndrome(MRLS) and fetal losses in other livestock.Nitrate,ammonia,and sulfate may react with proteinaceous compounds to enable the formation of abiotic pathogenic nanoparticles which were constant findings in pathognomonic placental lesions associated with non-infectious fetal losses of previously unknown etiology in mares,chickens and other livestock.The pathogenic nanoparticles may be produced naturally by toxic elements associated with air pollution that affect pasture forages or crops,uninte ntionally by reactions of these elements in protein-mineral mixes in dietary rations,or endogenously within tissues of fetuses and adult animals.The nanoparticles may form niduses in small vessels and predispose animals to a host of secondary opportunistic diseases affecting the reproductive,respiratory and gastrointestinal tracts of animals.The newly recognized abiotic pathogenic micro and nanoparticles are associated with MRLS.The discovery of the pathogenic nanoparticles led to the identification of nitrate,ammonium,and sulfur,in the form of sulfate,that seemingly enable the formation of the pathogenic nanoparticles in embryonic and fetal tissues. 展开更多

灌浆结实期温度与氮肥施用量是影响稻米品质的两个重要生态因子,尤其是与稻米蛋白含量及米饭食味关系密切。本文以多个水稻主栽品种为材料,通过灌浆结实期的人工控温试验、大田长期定位点的施氮处理试验和盆栽条件下的温氮两因素复合处... 灌浆结实期温度与氮肥施用量是影响稻米品质的两个重要生态因子,尤其是与稻米蛋白含量及米饭食味关系密切。本文以多个水稻主栽品种为材料,通过灌浆结实期的人工控温试验、大田长期定位点的施氮处理试验和盆栽条件下的温氮两因素复合处理试验,探讨了水稻灌浆结实期温度对稻米贮藏蛋白含量与组分影响及其有别于氮肥处理效应的差异规律,并分析了温度与氮肥两个因素对稻米贮藏蛋白及其组分影响的交互作用特点。结果表明,高温胁迫和增施氮肥均引起水稻籽粒总蛋白及其谷蛋白组分含量(%)的显著增加,但两者对稻米醇溶蛋白影响却存在明显差别。其中,高温处理引起醇溶蛋白含量显著下降,提高稻米谷蛋白/醇溶蛋白比值,而增施氮肥引起稻米谷蛋白和醇溶蛋白含量明显增加,但对谷蛋白/醇溶蛋白比值与贮藏蛋白各亚基的组成比例影响相对较小。在高温处理下,谷蛋白的57 kD前体亚基组分含量有所提高,而37 kD酸性亚基和22 kD碱性亚基随温度处理的差异变化却因品种而异,且高温处理对水稻籽粒蛋白绝对含量(mg grain–1)的影响程度也远没有其对蛋白相对含量(%)的影响明显。高氮×高温处理组合对稻米总蛋白与谷蛋白含量的影响程度显著大于单一高温或高氮处理,但在高氮水平下由高温引起稻米醇溶蛋白含量的下降幅度却小于其低氮对照,有利于稻米醇溶蛋白含量在不同温度处理下的相对稳定。 展开更多

Percutaneous coronary intervention is a well-established technique used to treat coronary artery disease,but the risk of coronary artery in-stent restenosis following percutaneous coronary intervention is still high.Previous studies revealed that high mobility group protein B1(HMGB1)plays a critical role in neointima formation.In this study,we aimed to investigate the role of glycyrrhizic acid(GA),an HMGB1 inhibitor,in the process of neointima formation and the potential mechanisms.We investigated the role of GA in neointima formation through an iliac artery balloon injury model in rabbits.Proliferation,migration,and phenotype transformation of human vascular smooth muscle cells(VSMCs)were observed.Besides,infl ammation and receptor for advanced glycosylation end products(RAGE)signaling pathways were studied.The results indicate that GA attenuated neointima formation and downregulated HMGB1 expression in injured artery in rabbits.HMGB1 promoted proliferation,migration,and phenotype transformation through the activation of RAGE signaling pathways in VSMCs,and blockade of HMGB1 by GA(1,10,and 100μM)could attenuate those processes and reduce proliferation of human VSMCs.In conclusion,the HMGB1 inhibitor GA might be useful to treat proliferative vascular diseases by downregulating RAGE signaling pathways.Our results indicate a new and promising therapeutic agent for restenosis. 展开更多

Background Coronary artery disease(CAD)remains a leading cause of morbidity and mortality.Cytokines play a potential role in atherosclerosis pathogenesis and progression.We investigated the association between high sensitive C-reactive protein(hs CRP)and severity of CAD.Methods CAD patients were stratified according to hs CRP cut-off value into high levels hs CRP group(≥8.4 mg/L)and low levels hs CRP group(<8.4 mg/L).Severity of CAD was assessed according to artery stenosis degree and the number of vessel involved.Statistical analysis was performed using Statistical Package for the Social Sciences(SPSS,version 23.0).Results The mean age was 60.3±11.0 years.The level of hs CRP was increased and ranged from 0.2 to 1020.0 mg/L.Biochemical risk factors and severity of CAD didn’t show significant differences between the two groups.In multivariate linear analysis,cardiac troponin I(c Tn I)and serum amyloid A(SAA)were predictors of hs CRP.As shown in receiver operating characteristic(ROC)curve analysis performed in patients with ST-segment elevation myocardial infarction(STEMI)and compared to myonecrosis biomarkers,hs CRP(area under the curve(AUC):0.905;95%CI:0.844-0.966;P<0.001)could be a powerful predictor marker in evaluating the infarct size after myocardial infarction but not better than c Tn I.Conclusions Hs CRP levels were not associated with the severity of CAD but could be useful in the evaluation of myocardial necrosis in patients with STEMI. 展开更多

大豆高蛋白基因分子标记辅助育种的应用 认领

为了在周口地区大豆遗传背景的基础上筛选出有效的高蛋白基因分子标记,选用高蛋白大豆周豆25号和高油周豆18号作为亲本杂交构建F2分离群体,选择144对SSR引物,运用BSA方法进行大豆高蛋白基因SSR分子标记筛选,并利用Mapmaker/Exp V3.0软... 为了在周口地区大豆遗传背景的基础上筛选出有效的高蛋白基因分子标记,选用高蛋白大豆周豆25号和高油周豆18号作为亲本杂交构建F2分离群体,选择144对SSR引物,运用BSA方法进行大豆高蛋白基因SSR分子标记筛选,并利用Mapmaker/Exp V3.0软件构建分子连锁图谱和定位分析。结果表明,在亲本间具有多态性引物32对;这32对引物在高、低蛋白基因池间有4对引物(satt182、satt419、satt239、satt598)表现多态性,但这4对引物与蛋白相关的标记不连锁,其中,引物satt182贡献率最高(3.1%)。利用标记satt182对来自河南、北京等地的50份高蛋白大豆材料进行检测,验证分子标记辅助育种方法的有效性,结果显示,50份大豆材料中检测出38份与相应的特异性条带相同,检测符合度为76.0%,检测结果有效性较高、准确性较好。 展开更多

短时高温胁迫对中华地鳖蛋白质和氨基酸含量的影响 认领

土鳖虫体内富含蛋白质和氨基酸,是一种重要的药用昆虫。高温对昆虫生命活动产生重要影响。为探明短时高温胁迫对土鳖虫体内蛋白质和氨基酸含量的影响,本试验以中华地鳖Eupolyphaga sinensis Walker 8龄雌若虫为研究对象,通过人工设置29... 土鳖虫体内富含蛋白质和氨基酸,是一种重要的药用昆虫。高温对昆虫生命活动产生重要影响。为探明短时高温胁迫对土鳖虫体内蛋白质和氨基酸含量的影响,本试验以中华地鳖Eupolyphaga sinensis Walker 8龄雌若虫为研究对象,通过人工设置29℃、33℃、37℃为高温处理组,以25℃为对照,测定2 h短时高温胁迫对中华地鳖存活率以及体内蛋白质和氨基酸含量的影响。结果表明:29℃、33℃和37℃短时高温处理对中华地鳖存活率无影响,蛋白质含量分别为76.57%±3.29%、79.60%±0.92%、77.00%±0.71%,与对照(84.50%±1.57%)相比均明显下降;短时高温处理后,29℃处理组土鳖虫总氨基酸含量为206.97±7.42 mg/g,6种人体必需的游离氨基酸苏氨酸8.23±0.32 mg/g,甲硫氨酸3.98±0.49 mg/g,异亮氨酸8.27±0.35 mg/g,亮氨酸16.38±0.86 mg/g,苯丙氨酸8.34±0.40 mg/g,以及赖氨酸12.56±0.64 mg/g的含量较对照和33℃、37℃高温处理组均显著升高,表明29℃短时高温处理可显著提高中华地鳖体内人体必需游离氨基酸含量。本研究是国内外对高温胁迫下药用昆虫体内活性成分的首次报道,研究结果将为进一步明确高温对土鳖虫的药用价值的影响,以及为土鳖虫人工养殖条件提供理论依据。 展开更多

A response surface method was employed to study the effect of α-amylase concentration, hydrolysis temperature and time on the production of high protein glutinous rice flour(HPGRF). The suspension of glutinous rice flour(15%) that contained 6.52% protein was gelatinized and subsequently hydrolyzed by thermostable α-amylase. The hydrolysis yielded 0.144–0.222 g/g HPGRF with 29.4%–45.4% protein content. Hydrolysis time exerted a significant effect, while enzyme concentration and hydrolysis temperature showed insignificant effect on the protein content and production yield of HPGRF. The result of response surface method showed that the optimum condition for the production of HPGRF that contained at least 36% protein was treating gelatinized 15% glutinous rice flour suspension with 0.90 Kilo Novo α-amylase Unit(KNU)/g α-amylase at 80 oC for 99 min. By carrying out the predicted hydrolysis condition, HPGRF with 35.9% protein and 61.8% carbohydrates was resulted. The process yielded 0.172 g/g HPGRF. HPGRF contained higher amount of essential amino acids compared to glutinous rice flour. HPGRF had higher solubility and lower swelling power, and also showed no pasting peak compared with glutinous rice flour. 展开更多

目的:探讨在高糖培养的肾小管上皮细胞中过氧化物酶体增殖物激活受体γ(PPARγ)对PTEN/AKT/FAK通路的调控机制及对肾小管上皮细胞上皮-间充质转化(EMT)的影响。方法:以高糖培养的肾小管上皮细胞NRK52E为研究对象,采用过表达及shRNA干扰... 目的:探讨在高糖培养的肾小管上皮细胞中过氧化物酶体增殖物激活受体γ(PPARγ)对PTEN/AKT/FAK通路的调控机制及对肾小管上皮细胞上皮-间充质转化(EMT)的影响。方法:以高糖培养的肾小管上皮细胞NRK52E为研究对象,采用过表达及shRNA干扰技术分别上调和下调肾小管上皮细胞中PPARγ的表达,并培养48 h,应用细胞免疫荧光、Western blot和RT-qPCR方法检测PTEN及EMT相关蛋白的表达变化;然后,在过表达及敲减PTEN的情况下,分别给予PPARγ抑制剂GW9662及激动剂罗格列酮干预,进一步观察PPARγ对AKT/FAK通路的调节是否通过PTEN实现的。结果:过表达PPARγ组PPARγ和PTEN的mRNA及蛋白表达增加,E-cad⁃herin的表达明显上调,而vimentin和α-SMA蛋白表达显著下调(P<0.05);PPARγ敲减组PPARγ及PTEN的mRNA及蛋白表达水平明显降低,E-cadherin的表达也明显下调,而vimentin和α-SMA蛋白表达显著升高(P<0.05)。GW9662干预组PTEN的表达减少,而p-AKT(Thr308)、FAK及p-FAK(Tyr397)蛋白水平均明显升高(P<0.05);与GW9662组相比,GW9662+过表达PTEN组PTEN表达明显增加,并伴随p-AKT(Thr308)、FAK及p-FAK(Tyr397)蛋白水平的降低(P<0.05);罗格列酮干预组PTEN的表达增多,而p-AKT(Thr308)、FAK和p-FAK(Tyr397)蛋白水平降低(P<0.05);与罗格列酮组相比,罗格列酮+PTEN shRNA组PTEN的表达减少,相应的p-AKT(Thr308)、FAK和p-FAK(Tyr397)蛋白水平升高(P<0.05)。结论:PPARγ能调节肾小管上皮细胞PTEN的mRNA及蛋白表达水平,并影响肾小管上皮细胞的EMT;PPARγ对AKT及FAK通路的调节及对细胞EMT的影响主要是通过PTEN实现的。 展开更多

[Objectives]This study was conducted to investigate the relationship between HSPs and the response of high temperature stress.[Methods]Molecular biological techniques were applied to clone and analyze the gene sequence of DpHsp70 gene.The changes in the expression of DpHsp70 gene under high temperature stress and exogenous salicylic acid(SA)were observed and further analyzed by qRT PCR.[Results]The coding region of the Dahlia DpHsp70 gene was 705 bp,encoding 234 amino acid residues(GenBank accession number:MH102288).Aligned with Compositae plants,the Dahlia DpHsp70 gene shared more than 83%homology in gene sequence while 99%-100%homology in amino acid sequence.Under the 35℃high temperature stress,the expression of DpHsp70 gene in Dahlia petals significantly increased.Meanwhile,the expression of DpHsp70 gene further increased under SA at 35℃temperature,which was significantly higher than those of the control group and the 35℃high temperature stress treatment group.It was demonstrated that the Hsp70 gene in Compositae is with highly conservative property and its expression could be up-regulated in response to high temperature stress.It can also be concluded that applying exogenous SA can improve the high temperature resistance of Dahlia.[Conclusions]This study provides a new experimental basis for elucidating the physiological function and mechanism of Dahlia in response to high temperature stress. 展开更多

Foods with a low glycemic index provide lower postprandial glucose spikes and induce satiety. The objective of this study was to assess the Glycemic index (GI) without milk and Glycemic response (GR) with milk of two different flavours of a plant-based supplement which is high in protein and fibre, along with a subjective assessment of satiety. Methods: Fifteen overweight/obese subjects aged 18 - 45 years were recruited. After overnight fasting, blood samples were drawn at 5 mins before food consumption (-5), 0, 15, 30, 45, 60, 90 and 120 minutes. Participants underwent 3 days of reference food testing and 1 day of test food in random order with 2 days of wash-out period. The GI was assessed using a validated protocol recognized by FAO/WHO, as well as the guidelines by the International Dietary Carbohydrate Task Force for GI Methodology. The satiety index was measured using the Visual Analog Scale (VAS). The dietary intake of the subjects was measured by 24-hour dietary recall. The Incremental Area Under the Curve (IAUC) was calculated using the trapezoid rule. Results: Both the flavours of the supplement had low GI & GR. The GI and GR of Flavour 1 were 27.3 ± 4.8 & 16.4 ± 2.6 (Mean ± SEM) respectively. For Flavour 2 the GI and GR were 36.7 ± 4.4 & 25.7 ± 2.3 (Mean ± SEM). For Flavour 1, 60% and for flavour 2 66.7% of subjects reported feeling hungry only after 3 hours, showing good satiety. Conclusion: The plant-based high fibre high protein supplement in both flavours showed a low glycemic index and hence may be useful to include in the diets to reduce the postprandial glycemic response and could improve satiety. 展开更多
