Orientin is a flavonoid monomer.In recent years,its importance as a source of pharmacological active substance is growing rapidly due to its properties such as anti-myocardial ischemia,anti-apoptosis,anti-radiation,anti-tumor,and anti-aging.However,the neuroprotective effects of Orientin on stroke injury have not been comprehensively evaluated.The aim of the present study was thus to investigate the neuroprotective capacity and the potential mechanisms of Cyperus esculentus L.orientin(CLO)from Cyperus esculentus L.leaves against ischemia/reperfusion(I/R)injury using standard orientin as control.For in vitro studies,we treated HT22 cells with CoCl2 as an in vitro ischemic injury model.HT22 cells in the control group were treated with CoCl2.For in vivo studies,we used rat models of middle cerebral artery occlusion,and animals that received sham surgery were used as controls.We found that CLO protected CoCl2-induced HT22 cells against ischemia/reperfusion injury by lowering lipid peroxidation and reactive oxygen species formation as well as decreasing protein oxidation.However,CLO did not reduce the release of lactate dehydrogenase nor increase the activity of superoxide dismutase.Results showed that CLO could decrease neurological deficit score,attenuate brain water content,and reduce cerebral infarct volume,leading to neuroprotection during cerebral ischemia-reperfusion injury.Our studies indicate that CLO flavonoids can be taken as a natural antioxidant and bacteriostastic substance in food and pharmaceutical industry.The molecular mechanisms of CLO could be at least partially attributed to the antioxidant properties and subsequently inhibiting activation of casepase-3.All experimental procedures and protocols were approved on May 16,2016 by the Experimental Animal Ethics Committee of Xinjiang Medical University of China(approval No.IACUC20160516-57). 展开更多

Our purpose was to determine the impact of ascorbic acid and α-tocopherol on bovine sperm motility following short-term storage or cryopreservation. Semen was collected from mature Angus bulls (n = 4). The experimental design was a randomized complete block, with bull serving as block, and treatments were structured as a 4 × 4 factorial with four concentrations of ascorbic acid (0, 5, 10, 20 mM) and four concentrations of α-tocopherol (0, 0.05, 0.5, 5 mM). Sperm motility characteristics were evaluated using computer assisted sperm analyses at 0, 4, and 8 h of incubation (39?C) and post-cryopreservation. Initial sperm motility was (79.6% ± 1.6) and decreased to (6.1% ± 1.6%) after cryopreservation. Cryopreserved spermatozoa had lower (P P > 0.1) by α-tocopherol. Addition of 20 mM ascorbic acid to storage media decreased (P P α-tocopherol. Stepwise regression models predicted (P α-tocopherol was not beneficial for short-term storage of spermatozoa;however, our results were inconclusive with regards to inclusion of ascorbic acid and α-tocopherol in egg yolk-based cryopreservation media. 展开更多

There are good reasons to include vegetables in human diet since they are enriched in bioactive compounds and by this reason they may help reduce the risk of some diseases. In this paper it was analyzed the nutrition quality and effect on disease prevention of vegetables. Each vegetable family and each vegetable contain a unique combination of bioactive compounds. The health benefit of vegetables should not be linked to one type of vegetable. It is presented some experimental research evidences that vegetables exert anti-oxidative, anti-carcinogenic, anti-diabetic and cardiovascular disease lowering effects. The mechanism by which vegetable bioactive compounds decrease the risk of some of these diseases is complex and sometimes unknown. 展开更多

The present study was carried out to determine whether the consumption of epigallocatechin (EGCG), the major bioactive green tea catechin, exerts a positive effect on lowering in vivo lipid peroxidation, a measure of oxidative stress, in healthy postmenopausal women. Urinary excretion of secondary lipid peroxidation products, a measure of in vivo lipid peroxidation, was determined in 40 participants randomly assigned to consume a green tea catechin extract (843.0 ± 44.0 mg EGCG/d) or placebo capsules for 12 months. Urine samples were analyzed for individual polar and nonpolar lipophilic aldehydes and related carbonyl compounds by high-performance liquid chromatography (HPLC) at the beginning and at the end of the 12-month intervention period. Results show that two nonpolar aldehydes, nonanal and decatrienal, were both 48% lower (p in vivo antioxidant activity exists with long-term EGCG consumption, which could slightly limit oxidative damage associated with lipid peroxidation and the onset and progression of chronic diseases. 展开更多

Antioxidants are substances that can prevent or slow damage to cells caused by free radicals, unstable molecules that the body produces as a reaction to environmental and other pressures. Free radicals may play a role in heart disease, cancer and other diseases. If the body cannot process and remove free radicals efficiently, oxidative stress can result. This can harm cells and body function. Free radicals are also known as reactive oxygen species (ROS). In this research, Triptorelin&#174;(TRP) conjugates of triphenylmethanol derivatives (TPMs) were synthesized to evaluate their in vitro lipid peroxidation potency. Comparative lipid peroxidation assays between TRP-TPMs conjugates and the corresponding TPMs derivatives were measured using thiobarbituric reactive substance (TBARS) in a dose- and time-dependent manner following the Fenton’s pathway. Overall, TBARS decreased between 20% - 30% for the treated samples of synthesized conjugates compared to their respective control physical mixtures. These data suggest that TRP-TPMs derivatives can be used to improve the biological activity of TRP. 展开更多

In the present work, the experimental formula was designed to remove these blockers. It was applied on 120 Swiss albino mice which were inoculated intraperitoneally and subcutaneously with Ehrlich Ascites Carcinoma cells;1 × (106) cell/mouse. The activity of the cell death programs of the tumor was detected by measuring the volume of Ascites fluid, counting the number of dead cancer cells, measuring the size of the tumor, detecting the positive reaction of caspase enzyme in cancer cells and presence of macrophages and apoptotic bodies in tumor tissue. The experimental formula succeeded in removing the blockers of the cell death program in cancer cells returning the cell death program to work again. 展开更多

This study describes the phytochemical profile and antioxidant activity of an extract from the leaves of Pouteria caimito (Sapotaceae). The extract in ethanol was obtained by maceration at room temperature and subjected to the liquid-liquid partition to obtain fractions in hexane and ethyl acetate. Steroids, triterpenes, saponins, alkaloids and flavonoids were identified by the phytochemical prospection of extracts and fractions from the leaves. The analysis of the ethyl acetate fraction by UPLC-QTOF-MS allowed us to identify eight triterpenes, namely, euscaphic acid (1), hyptadienic acid (2), betulinic acid (3), oleanolic acid (4), ursolic acid (5), 3β-(O-p-coumaroyl)- alphitolic acid (6), 3β-(O-p-coumaroyl)-maslinic acid (7) and 3β-(O-p-coumaroyl)-2-hydroxy-urs-12-en-28-oic acid (8). The ethanol extract and ethyl acetate fraction presented total phenolic contents of 10.6 ± 0.1 and 11.4 ± 0.3 mg GAE g-1, respectively, and considerable antioxidant activity in the DPPH assay with EC50 values of 299.4 ± 1.5 and 391.8 ± 0.9 μg·ml-1, respectively. 展开更多

Comparative Study of the Antioxidant and Antimicrobial Activities of Compounds Isolated from Solvent Extracts of the Roots of <i>Securinega virosa</i> 预览

There are several lines of indications on the importance of plants as accepted sources of therapeutic agents since most synthetic antioxidants and antibiotics have been associated with cytotoxicity and/or microbial resistance. In this study, the antioxidant and antimicrobial activities of a brown compound recovered from the ethanol extractable fraction (EEF) and white crystals isolated from the chloroform-methanol elution (CME) following column chromatography of the ethanolic extracts of roots of Securinega virosa were examined. Preparations from the white crystalline compound recorded greater antioxidant activity and antimicrobial activities than the crude ethanol extracts of S. virosa. Both the ethanol extract and the crystals showed significant antimicrobial activities against all microbes employed in this study. The crystalline compound demonstrated the highest zone of inhibition against the microbes Enterococcus faecalis with varied inhibitory activities with respect to other microorganisms. The least inhibition was against Pseudomonas fluorescens. The overall range was 8.0 - 22.5 mm. The EEF of the roots of S. virosa recorded its highest activity against Staphylococcus aureus and Escherichia coli. There was varied inhibitory activity against other organisms;the least was against Micrococcus liteus. The zone of inhibition ranged 6.33 - 17.67 mm. Similar to the trends in susceptibility test, the EEF showed appreciable minimum inhibitory concentration (MIC), ranging from 3.13 - 25 mg/mL against test microorganisms while the crystalline preparation had a MIC range of 1.5 - 25 mg/mL except against Salmonella typhii which was 50 mg/mL. Both extracts demonstrated their highest activity against E. faecalis reducing significantly in a dose-dependent manner by each sample against the rest of the test microbes. However, the standard broad-spectrum antibiotic, chloramphenicol, used elicited a zone of inhibition ranging from 0 - 30 mm but with no activity against Streptococcus thermophilus. The pecan nut is considered one of the most important dried fruits in the world by its classification as healthy food. The nutritional balance in the harvest is crucial for its impact on the production, quality and content of bioactive compounds. This research was carried out in the city of Aldama, Chihuahua (Mexico), where doses of mineral and organic fertilization were tested in L25 Taguchi structure with 6 factors and 5 levels by factor: nitrogen (N) 0 - 240 kg·ha-1, phosphate (P2O5) 0 - 120 kg·ha-1, potassium (K2O) 0 - 100 kg·ha-1, calcium (CaO) 0 - 400 kg·ha-1, liquid humus 0 - 3600 L·ha-1 and solid humus 0 - 8000 kg·ha-1. An average yield of 2.2 t·ha-1 was obtained;145 nuts per kilogram, 60% of the edible part of the nut. The total phenolic content was 225.9 mg gallic acid g-1 and the antioxidant capacity was 180.9 mg Trolox g-1. It is concluded that the factor with the greatest impact on yield, quality and antioxidant capacity in pecan nut was nitrogen. It was also found that the combination of mineral and organic fertilization helps to maintain the production and quality ranges of the nut. In turn, these factors contribute to the increase in the phenolic strength and antioxidant capacity. The optimal fertilization dosis to obtain the maximum levels in production of 3.2 t·ha-1;decrease the number of nuts per kilogram to 135, increase the percentage of the edible part of the nut to 61.8%, the increase in the total phenolic content to 318.6 mg of gallic acid g-1 and in the antioxidant capacity with 187.2 mg of Trolox g-1, was 184 kg·ha-1 of N, 107.4 kg·ha-1 of P2O5, 50 kg·ha-1 of K2O y 2777 L·ha-1 of liquid humus. Finally, the mineral fertilization complemented with organic fertilization is considered a good fertilization strategy for pecan trees and its possible benefits to health and the environment. 展开更多

Oxidative stress represents a risk factor and plays a key role at several steps of atherosclerosis. Formation of atherosclerotic plaques is the main reason for coronary artery disease. Aim: This study is aimed to assess the impact of the imbalance between body oxidants and antioxidants (oxidative stress) and its involvement in developing and severity of atherosclerosis in atherosclerotic with and without diabetes Libyan patients. Methods: The study sample comprised of atherosclerosis with and without diabetes patients (n = 73), as well as healthy controls (n = 27). Blood samples were collected to determine the levels of malonaldehyde (MDA) as oxidative biomarker, serum selenium level, total antioxidant status (TAS) level and some antioxidant enzymes including glutathione peroxidase (GPx), Catalase (Cat) and superoxide dismutase (SOD). Additionally, oxidant/antioxidant status was compared between atherosclerotic patients with and without diabetes in comparison to controls. In vitro antioxidant activity was performed via different assays and antimicrobial potential was observed via disc diffusion method. Cytotoxic activity was carried out using brine shrimps’ assay.RESULTS: Phytochemical studies revealed the presence of alkaloids, flavonoids, glycosides, phenols,steroids, saponins, tannins, anthocyanins and coumarins in most of the plant extracts. Maximum antioxidant and antimicrobial potential were observed in Cymbopogon citratus methanol extract and Cymbopogon citratus n-hexane extract along with significant number of total flavonoids and phenols contents. However, Polypogon monspeliensis methanol extract and Polypogon monspeliensis n-hexane extract showed minimum antioxidant as well as antimicrobial potential. Moreover, methanol extracts showed a cytotoxic effect with their effectiveness ranked Polypogon monspeliensis methanol extract > Cymbopogon citratus methanol extract > Phalaris minor n-hexane extract > Aristida funiculata methanol extract > Stipagrostis plumosa methanol extract > Cenchrus ciliaris methanol extract > Panicum antidotale methanol extract. Similarly, n-hexane plant extracts revealed cytotoxic activity in decreasing order Cenchrus ciliaris n-hexane extract > Stipagrostis plumosa n-hexane extract > Phalaris minor n-hexane extract > Aristida funiculata n-hexane extract > Polypogon monspeliensis n-hexane extract > Panicum antidotale n-hexane extract > Cymbopogon citratus n-hexane extract respectively.CONCLUSION: Methanol extracts exhibit significant antioxidant and antimicrobial potential which can be correlated to their medicinal values. Scavenging reactive oxygen species(ROS) by antioxidants is the important therapy to cerebral ischemia-reperfusion injury(CIRI) in stroke. The antioxidant with novel dual-antioxidant mechanism of directly scavenging ROS and indirectly through antioxidant pathway activation may be a promising CIRI therapeutic strategy. In our study, a series of chalcone analogues were designed and synthesized, and multiple potential chalcone analogues with dual antioxidant mechanisms were screened. Among these compounds, the most active 33 not only conferred cytoprotection of H2 O2-induced oxidative damage in PC12 cells through scavenging free radicals directly and activating NRF2/ARE antioxidant pathway at the same time, but also played an important role against ischemia/reperfusion-related brain injury in animals. More importantly, in comparison with mono-antioxidant mechanism compounds, 33 exhibited higher cytoprotective and neuroprotective potential in vitro and in vivo. Overall, our findings showed compound 33 couldemerge as a promising anti-ischemic stroke drug candidate and provided novel dual-antioxidant mechanism strategies and concepts for oxidative stress-related diseases treatment. 展开更多

建立高效液相色谱-串联质谱法(high performance liquid chromato graphy-tanelem mass spectrometry,HPLC-MS/MS)法同时测定酵母产品中4种抗氧化剂和5种对羟基苯甲酸酯类防腐剂的定性定量分析方法。样品经溶解脱脂,用正己烷饱和的乙腈... 建立高效液相色谱-串联质谱法(high performance liquid chromato graphy-tanelem mass spectrometry,HPLC-MS/MS)法同时测定酵母产品中4种抗氧化剂和5种对羟基苯甲酸酯类防腐剂的定性定量分析方法。样品经溶解脱脂,用正己烷饱和的乙腈提取,浓缩至干流动相复溶,经C 18净化、过膜后供仪器测试。采用电喷雾离子源、多重反应监测模式检测,以保留时间和特征离子对比较定性定量。9种分析物在0.1~10 mg/L相关系数(r^2)均大于0.992,方法定量限为0.10~0.50 mg/kg,酵母产品中3水平6平行的加标回收实验,回收率在78.9%~108.6%,相对标准偏差为4.2%~15.8%。该方法快速简便、准确可靠,适用于酵母产品中4种抗氧化剂和5种对羟基苯甲酸酯类防腐剂的测定,为高通量检测此类物质提供了技术支持。 展开更多
