Development of natural based nanomaterial immense interest of researchers involved in eco-friendly biosynthesis of nanoparticles. This research set out to investigate the potential of Allium ampeloprasum leaves extract to synthesize silver nanoparticles (AgNPs), and evaluate their antifungal activity against some toxigenic strains of Aspergillus isolates. In order to achieve this, AgNPs were prepared using plant extracts at room temperature and the formation of AgNPs was visually detected after the color changed to dark brown within few minutes. The biosynthesized AgNPs were characterized using the UV-vis spectroscopy, which confirmed the formation of AgNPs with a maximum peak at 437 nm due to the Plasmon resonance. The main active reduction agents were detected by Fourier Transmission infrared spectroscopy (FTIR). Also, the nanoparticles were characterized using Scanning electron microscope (SEM). The antifungal activity of AgNPs was investigated by agar well diffusion method, which revealed inhibition zones of 8 mm, 10 mm, 11 mm, 11 mm and 14 mm against Aspergillus flavus 1, A. parasiticus, A. flavus 2, A. ochraceus, and A. niger. The minimum inhibition concentration (MIC) was determined using micro broth dilution method. MIC values of AgNPs ranged from 652 - 2500 μg/ml. The fungi treated with AgNPs, were examined with (SEM), it was observed that the treated fungi were damaged. These results suggest that AgNPs have the potential to be used as an ideal eco-friendly approach to control toxigenic fungi. 展开更多

Permeation was evaluated through a synthetic membrane. The study provided information on the rate of diffusion and percentage of drug release and also compared the penetration dynamics in the layers of the stratum corneum of both formulations. Liposome TH cream was able to deliver the active ingredient to a larger number of SC layers, with a higher amount of drug, which is very promising for the treatment of superficial mycoses. 展开更多

Intracranial Fungal Infections: Overview from Two Large Tertiary Hospital in Upper Egypt and Literature Review 预览

Purpose: Fungal infections of the central nervous system (CNS) are potentially lethal conditions with high morbidity and mortality. In this review, we summarise the most common clinical manifestations, diagnostic methods, and treatment strategies for intracranial fungal infection at two tertiary care teaching hospitals. Material and methods: Prospective hospital study is carried out at Department of Neurosurgery;Assiut and Suhaj University Hospitals between January2010 to January 2018 (Minimum 12-months follow-up). Radiographs and hospital data of 74 patients with proven intracranial fungal infections were gathered and analyzed. There were no exclusion criteria: age, gender, clinical presentations, immunity status, radiological findings, laboratory, and microbiological data, types of management and outcome. In surgically treated patients, diagnosis was confirmed by pathologic evaluation. Gathered data were coded and entered into a computer and analyzed using SPSS version 22. Results: The greatest number of the patients had 40 to 60 years old (49;66%) and the mean age was 44 years. There was an overwhelming male patient’s ranged preponderance 66%;49 cases. Sixty-three patients (85%) were immunosuppressed;11 cases (15%) were immunocompetent. The most common causes of immunosuppression were diabetes 27 patients;43%, on chemotherapeutic agents 19 patients;31%, on corticosteroid 16 patients;25% and AIDS in one patient;1%. Five different fungal types were identified but Cryptococcus spp. was the most common cause of CNS fungal infection, occurring in 39 patients (53%). This was followed by Candida spp. in 14 patients (19%), Aspergillus in 11 patients (15%), Blastomyces in 7 patients (9%) and Coccidiosis in 3 patients (4%). Headache was the most common presenting symptom, occurring in 33 patients (45%). Other relatively common symptoms were nausea or vomiting 11 patients (15%), fever 10 patients;(13%), seizures 9 patients (12%), acute mental status changes 8 patients;(11%) and stroke like Symptoms 3patients (4%). Bioassay results showed that some of them exhibited good activity against Pyricularia oryzae (P. oryzae). It was found that the compound 5q (benzyl (6-(tert-butyl)-8-fluoro-2,3-dimethylquinolin-4-yl) carbonate) possessed good activity against P. oryzae whatever protective activity (10 mg·L^-1) or curative activity (25 mg·L^-1), which was better than that of control tebufloquin. In addition, the frontier molecular orbit results revealed that the compound held higher activity against P. oryzae when the total energy was low and the ClogP was high, which may provide useful information for further design novel fungicides. 展开更多

Objective: To evaluate the anti Candida activity of Hyptis martiusii decoction and its major compound, caffeic acid alone or in the presence of fluconazole, as well as their cytotoxic effect. Methods: The decoction was characterized using high performance liquid chromatography coupled with diode array detector. For the antifungal activity, the minimum inhibitory concentration(MIC) and the potential effect of the decoction with the fluconazole were evaluated by microdilution method using 96-well microtiter trays. The osmotic fragility test was performed using erythrocytes under saline stress. All tests were performed in triplicate. Results: The chemical characterization of the decoction was performed by high performance liquid chromatography and revealed the presence of seven compounds, including caffeic acid as major constituent. The antifungal tests demonstrated that both decoction(DHm) and caffeic acid obtained from Hyptis martiusii presented MIC and MFC ≥4096 μg/mL against Candida albicans and Candida tropicalis strains. However, in the presence of fluconazole, DHm and caffeic acid presented IC50 of 2.60 and 2.53 μg/mL respectively, demonstrating significant synergistic effects against Candida strains. The modulator activity of DHm might be due to the presence of caffeic acid. Moreover, DHm and caffeic acid did not cause significant hemolytic effects, indicating that they present low cytotoxicity. Conclusions: These data indicate that DHm potentiates the activity of the fluconazole, without enhancement of the toxicity, encouraging further toxicological, pharmacological and phytochemical studies to provide consistent evidence of the potential of this plant to be used in drug development. 展开更多

A series of novel 1,3,5-thiadiazine-2-thione derivatives bearing a hydrazide moiety were designed, synthesized and evaluated for their antifungal activities against Rhizoctonia solani (Rs), Fusarium graminearum (Fg), Botrytis cinerea (Bc) and Colletotrichum capsici (Cc). The in vitro antifungal bioassays indicated that most of title compounds displayed good selectivity and specificity aganist Rs relative to Fg, Bc and Cc. Strikingly, the title compound N'-(4-chlorophenyl)-2-(5-phenyl-6-thioxo-1,3,5-thiadiazinan-3-yl)acethydrazide (5b) obviously inhibited the Rs growth in vitro with the EC50 value of 0.24μg/mL, which is approximately 2-folds more effective than the commercialized fungicide carbendazim (0.55 μg/mL). The in vivo anti-Rs effects of title compound 5b were further evaluated on rice leaves with control efficacies of 98.58% at 200 μg/mL and 61.27% at 100 μg/mL. The above researches provide a significant reference for the further structural optimization of 1,3,5-thiadiazine-2-thione derivatives bearing a hydrazide moiety as potential fungicides. 展开更多

However, no clear guidelines have been deve... BACKGROUND Non-albicans Candida prosthetic joint infections (PJIs) are rare. Optimal treatment involves a two-stage revision surgery in combination with an antifungal agent. However, no clear guidelines have been developed regarding the agent or treatment duration. Hence, a broad range of antifungal and surgical treatments have been reported so far. AIM To clarify treatment of non-albicans Candida PJIs. METHODS A literature review of all existing non-albicans Candida PJIs cases through April 2018 was conducted. Information was extracted about demographics, comorbidities, responsible species, duration and type of antifungal treatment, type of surgical treatment, time between initial arthroplasty and symptom onset, time between symptom onset and definite diagnosis, outcome of the infection and follow-up. RESULTS A total of 83 cases, with a mean age of 66.3 years, were located. The causative yeast isolated in most cases was C. parapsilosis (45 cases;54.2%), followed by C. glabrata (18 cases;21.7%). The mean Charlson comorbidity index was 4.4 ± 1.5. The mean time from arthropalsty to symptom onset was 27.2 ± 43 mo, while the mean time from symptom onset to culture-confirmed diagnosis was 7.5 ± 12.5 mo. A two stage revision arthroplasty (TSRA), when compared to one stage revision arthroplasty, had a higher success rate (96% vs 73%, P = 0.023). Fluconazole was the preferred antifungal agent (59;71%), followed by amphotericin B (41;49.4%). CONCLUSION The combination of TSRA and administration of prolonged antifungal therapy after initial resection arthroplasty is suggested on the basis of limited data. 展开更多

Objective: To isolate and identify the antifungal compounds from Curcuma amada. Methods: The antifungal activity was measured by the diameter of colonies grown on Petri dish,microscopic observation,and CLSI microdilution methods. The antifungal compounds were isolated through bioactivity guided purification by using silica gel and high-performance liquid chromatography. Structural identification of the antifungal compounds was conducted using 1 H NMR,13 C NMR,and liquid chromatography-tandem mass spectrometry. Results: The purified antifungal compounds were zederone and furanodienone. These two compounds showed dose-dependent antifungal activity against Fusarium solani sensu lato. The concentration required for 50% growth inhibition(IC50) of FSSL ranged from 115 to 129 μM and 82 to 91 μM for zederone and furanodienone,respectively. Conclusion: This study suggested that the isolated compounds from Curcuma amada could be promising natural antifungal agents to control the diseases caused by Fusarium solani sensu lato. 展开更多

Bifurcaria bifurcata is a seaweed of the Order Fucales(Ochrophyta,Phaeophyceae)that can be found all year round along the Portuguese Atlantic shore.Although it is considered edible in some countries,its biotechnological potential has not yet been assessed in detail and here we report its antimicrobial potential against human dermatophyte fungi.Three samples were harvested from Baleal Island(Peniche,Portugal)harvested in winter(BbPe);from Aguda Beach(Arcozelo,Vila Nova de Gaia,Portugal),harvested in autumn(BbAg)and a sample obtained by culturing the Aguda sample under laboratory conditions(BbLC).A broth macrodilution assay was applied to determine the MIC(minimum inhibitory concentration)and MLC(minimum lethal concentration)of the B.bifurcata extracts.Results show that the BbPe MeOH extracts were the most effective and had higher antifungal activity against all the tested dermatophyte strains than the BbLC and BaAg extracts[Epidermophyton floccosum FF9(100–200μg/mL MIC;200μg/mL MLC),Microsporum canis FF1(400μg/mL MIC;≥400μg/mL MLC),Trichophyton mentagrophytes FF7(100μg/mL MIC;>800μg/mL MLC),M.gypseum CECT 2908(800μg/mL MIC;≥800μg/mL MLC),T.mentagrophytes var.interdigitale CECT 2958(800μg/mL MIC;≥800μg/mL MLC),T.rubrum CECT 2794(200μg/mL MIC;≥400μg/mL MLC)].In fact,only E.floccosum FF9 was sensitive to BbAg and BbLC MeOH extracts.To our knowledge,this is the first report of antifungal activity of B.bifurcata against human dermatophyte fungi. 展开更多
