BACKGROUND Nucleic acid isolation from formalin-fixed, paraffin-embedded tissue(FFPET)samples is a daily routine in molecular pathology laboratories, but extraction from FFPET is not always easily achieved. Choosing the right extraction technique is key for further examinations.AIM To compare the performance of four commercially available kits used for DNA extraction in routine practice.METHODS DNA isolation was performed on 46 randomly selected formalin-fixed, paraffinembedded(FFPE) colorectal adenocarcinoma(CRC) surgical specimens. Four commercially available extraction kits were used: two for manual DNA extraction(the Pure Link Genomic DNA Mini Kit from Invitrogen and the High Pure FFPE DNA Isolation Kit from Roche) and two for automated DNA extraction(the i Prep Genomic DNA Kit from Invitrogen and the Magna Pure LC DNA Isolation Kit from Roche). The DNA concentration and quality(odds ratio) among the four systems were compared. The results were correlated with the clinicopathological aspects of CRC cases: age, gender, localization, macro-and microscopic features,lymph node metastases, and the lymph node ratio.RESULTS The highest DNA concentration was obtained using the manual kits: 157.24 ±62.99 ng/μL for the Pure Link Genomic DNA Mini Kit and 86.64 ng/μL± 43.84 for the High Pure FFPE DNA Isolation Kit(P < 0.0001). Lower concentrations were obtained with automated systems: 20.39 ± 21.19 ng/μL for the Magna Pure LC DNA Isolation Kit and 8.722 ± 6.408 ng/μL for the i Prep Genomic DNA Kit,with differences between the systems used(P < 0.0001). The comparison between age, gender, tumor localization, pT or pN stage and the lymph node ratio indicated no statistically significant difference in DNA concentration using any of the nucleic acid isolation kits. DNA concentration was influenced by the macroscopic features and grade of differentiation. Foamed concrete has a good energy absorption capability and can be used as seismic isolation material for tunnels.This study aims to investigate the mechanical properties and associated seismic isolation effects of foamed concrete layer in rock tunnel.For this,a series of uniaxial/triaxial compression tests was conducted to understand the effects of concrete density,confining stress and strain rate on the mechanical properties of foamed concrete.The direct shear tests were also performed to investigate the effects of concrete density and normal stress on the nonlinear behaviors of foamed concrete layer-lining interface.The test results showed that the mechanical properties of foamed concrete are significantly influenced by the concrete density.The foamed concrete also has high volumetric compressibility and strain-rate dependence.The peak stress,residual stress,shear stiffness and residual friction coefficient of the foamed concrete layer-lining interface are influenced by the foamed concrete density and normal stress applied.Then,a crushable foam constitutive model was constructed using ABAQUS software and a composite exponential model was also established to study the relationship between shear stress and shear displacement of the interface,in which their parameters were fitted based on the experimental results.Finally,a parametric analysis using the finite element method(FEM)was conducted to understand the influence of foamed concrete layer properties on the seismic isolation effect,including the density and thickness of the layer as well as the shear stiffness and residual friction coefficient of the interface.It was revealed that lower density and greater thickness in addition to smaller shear stiffness or residual friction coefficient of the foamed concrete layer could yield better seismic isolation effect,and the influences of the first two tend to be more significant. 展开更多

隔震技术在大跨度复杂建筑中的应用现状及关键问题

BACKGROUND Open abdomen (OA) has been generally accepted for its magnificent superiority and effectiveness in patients with severe trauma, severe intra-abdominal infection, and abdominal compartment syndrome. In the meantime, OA calls for a mass of nursing and the subsequent enteroatomospheric fistula (EAF), which is one of the most common complications of OA therapy, remains a thorny challenge. CASE SUMMARY Our team applied thermoplastic polyurethane as a befitting material for producing a 3D-printed “fistula stent” in the management of an EAF patient, who was initially admitted to local hospital because of abdominal pain and distension and diagnosed with bowel obstruction. After a series of operations and OA therapy, the patient developed an EAF. CONCLUSION Application of this novel “fistula stent” resulted in a drastic reduction in the amount of lost enteric effluent and greatly accelerated rehabilitation processes. 展开更多

淀粉酶作为水解淀粉和糖原的酶类,在生物的新陈代谢和工业化生产中起着广泛而重要的作用,尤其是高温淀粉酶在利用过程中备受关注.从云南大理市弥渡县石夹泉热泉的43℃底泥中分离并筛选到1株高温淀粉酶的高产菌,进行显微形态及生理生化... 淀粉酶作为水解淀粉和糖原的酶类,在生物的新陈代谢和工业化生产中起着广泛而重要的作用,尤其是高温淀粉酶在利用过程中备受关注.从云南大理市弥渡县石夹泉热泉的43℃底泥中分离并筛选到1株高温淀粉酶的高产菌,进行显微形态及生理生化特征、16S rRNA基因序列分析,将其初步鉴定为芽孢杆菌属(Bacillus sp.)的一株菌,命名为Bacillus sp. Amy-43.对其生长条件及酶学性质进行了研究,结果表明:该菌株耐高温性较好,菌株在55℃仍能生长,菌株最适生长温度为37℃.所产淀粉酶最适酶活温度为75℃,最适反应p H值为4.0.在37℃以下,能保持良好的稳定性,Zn2+和Mn2+对该酶有一定的抑制作用,而Ca2+、K+、Mg2+、Fe2+、Cu2+均对该酶活力起到促进作用,其中Cu2+的促进效果最为明显.这些特性预示着Bacillus sp.Amy-43来源的淀粉酶为工业运用提供了重要的酶源. 展开更多

陇西腊肉是一种传统发酵风味肉制品。以陇西腊肉为材料,通过经典形态学分类和生理生化鉴定,对其加工过程中的优势乳酸菌(lactic acid bacteria,LAB)进行分离、鉴定。通过测定菌株的产酸能力、生长特性、耐氯化钠和亚硝酸钠能力以及产气... 陇西腊肉是一种传统发酵风味肉制品。以陇西腊肉为材料,通过经典形态学分类和生理生化鉴定,对其加工过程中的优势乳酸菌(lactic acid bacteria,LAB)进行分离、鉴定。通过测定菌株的产酸能力、生长特性、耐氯化钠和亚硝酸钠能力以及产气、产黏液、产蛋白酶和脂肪酶的能力等,筛选具有优良发酵性能的菌株。随后,开展人工接种优良菌株的模拟腊肉加工试验,分析其对腊肉感官品质的影响。结果表明,从陇西腊肉中筛选获得7株乳酸菌,根据菌种形态学分类,初步鉴定分别为植物乳杆菌(Lactobacillus plantarum)、米酒乳杆菌(L.sake)、乳酸乳球菌(Lactococcus lactis)、戊糖片球菌(Pediococcus pentosaceus)、肠膜明串珠菌(Leuconostoc mesenteroides)、食品乳杆菌(L.alimentarius)和弯曲乳杆菌(L.curvatus),其中,菌株R-1(植物乳杆菌)产酸能力强、生长速度快,对氯化钠和亚硝酸钠均具有较好的耐受性。经模拟腊肉发酵试验发现,添加菌株R-1后,陇西腊肉的风味得到了较大改善,表现出更好的感官可接受性。研究表明,植物乳杆菌R-1具有优良的发酵性能,可作为传统陇西腊肉工艺现代化改造的菌株来源。 展开更多

Sheep pox is an infectious viral disease that affects specifically sheep and it is caused by the Capripoxvirus genus.The clinical signs include fever,diarrhea,difficulty breathing,nodules,lung lesions and death.In Morocco,the 2010 epidemic of sheep pox was characterized by the emergence of a nodular form of the disease.The local strain was isolated and the analysis of affected animals was positively confirmed by virus isolation and real-time polymerase chain reaction(RT-PCR).The epidemiological analysis of 911 data records showed that the virus is endemic in the country;an average of 350 cases per year with an epizootic evolution was observed in 2010.The incidence varies depending on provinces and the disease appears confined to the central and the eastern regions of the country where a very intensive sheep breeding activity is taking place.The statistical analysis showed that there is a positive correlation between the endemicity and the significant factor of the rural market(p=0.006).The annual average morbidity and mortality rates were 2.96%(1.26%to 4.32%)and 0.71%(0.41%to 0.94%),respectively.The clinical findings associated to the epidemiological data analysis confirmed the presence of sheep pox in its nodular form and suggest that new pathogenic strains may have been introduced from Mauritania.The purpose of this work was to provide a better description of the spatiotemporal evolution of sheep pox disease based on some epidemiological indicators and to put forward plausible hypotheses regarding the emergence of the virus in order to implement an adequate control strategy. 展开更多

Drug susceptibility test was conducted using 15 kinds of drug susceptibility papers. The E. coli was sensitive to florfenicol, enrofloxacin, ceftriaxone, norfloxacin;intermediately sensitive to amikacin, gentamicin;and strongly resistant to penicillin, ampicillin,cefradine, sulfamethoxazole, lincomycin, streptomycin and amoxicillin.[Conclusion] It is difficult to treat E. coli causing fox pneumonia with traditional antibiotics clinically. 展开更多

为了给川北地区养猪场提供副猪嗜血杆菌病的科学防控及合理的治疗用药方案,对川北地区西充、三台、绵阳等地规模化猪场保育猪发生的副猪嗜血杆菌病病原进行分离鉴定。采集副猪嗜血杆菌病疑似病死猪的肺、肝、脾、淋巴结、心包液、腹水... 为了给川北地区养猪场提供副猪嗜血杆菌病的科学防控及合理的治疗用药方案,对川北地区西充、三台、绵阳等地规模化猪场保育猪发生的副猪嗜血杆菌病病原进行分离鉴定。采集副猪嗜血杆菌病疑似病死猪的肺、肝、脾、淋巴结、心包液、腹水、关节肿液等样本55例,通过细菌的分离纯化,对分离菌进行了染色镜检、V因子需要试验、致病性试验、PCR鉴定、药敏试验和同源性分析。结果表明,分离出了20株副猪嗜血杆菌,分离株革兰染色为阴性,有荚膜,分离菌株与GenBank公布的12株副猪嗜血杆菌的16S rRNA序列有高度同源性,且分离株对β-内酰胺类、四环素类、磺胺类的耐药率最高。试验结果对于川北地区养猪生产中副猪嗜血杆菌病的有效防治提供依据。 展开更多

In order to isolate and identify the pathogenic bacteria causing dead chickens in a chicken farm in Qinhuangdao area, the liver, heart and other organs of dead chickens suspected of salmonella disease were collected aseptically, and streaked on SS agar medium and chromagar medium. Transparent colonies were observed on SS agar medium, and purple and transparent colonies on CAS medium. The isolate was conducted purification, staining microscopy, biochemical tests, and 16 S rRNA sequence analysis, and the results showed that four strains of the isolated bacte-ria were salmonella. The 16 S rRNA sequence analysis of four strains of salmonella showed that the isolates shared more than 99% homology. Drug susceptibility test was performed using paper method, and the results showed that most of the strains were resistant to tilmicosin, cefradine and sul-famethoxazole, but were sensitive to ceftriaxone. 展开更多

为了解百香果果浆的内生细菌,该研究采用平板计数琼脂(PCA)培养基从百香果果浆中分离内生细菌,通过形态观察及分子生物学技术对其进行鉴定,并对其生长条件进行研究。结果表明,从百香果果浆中分离得到3株菌落形态单一、均呈浅黄色的内生... 为了解百香果果浆的内生细菌,该研究采用平板计数琼脂(PCA)培养基从百香果果浆中分离内生细菌,通过形态观察及分子生物学技术对其进行鉴定,并对其生长条件进行研究。结果表明,从百香果果浆中分离得到3株菌落形态单一、均呈浅黄色的内生细菌,分别编号为GJ2-5、GJ2-13、GJ2-19,均呈革兰氏阳性。3株内生细菌均为泛菌属(Pantoea),其中,菌株GJ2-5和GJ2-13均为分散泛菌(Pantoea dispersa),菌株GJ2-19为Pantoea eucrina。3株内生细菌的最适生长培养基均为LB液体培养基,最适生长pH值均为5.5,最适生长温度均为35℃,生长周期为迟滞期(0~6 h)、对数期(6~18 h)、平稳期(18~22 h)和衰亡期(22 h以后)。 展开更多
