[Objectives]This study was conducted to determine kaempferol content in ginkgo(Ginkgo biloba L.)leaves subjected to microbial fermentation.[Methods]Bacillus licheniformis was selected for solid-state fermentation of ginkgo leaves,and the content of kaempferol in ginkgo leaves was determined by RP-HPLC method.At first,methanol was used to extract flavonoid glycosides,which were then hydrolyzed by hydrochloric acid solution.HPLC was performed with Platisil ODS column C 18(150 mm×4.6 mm,5μm)using mobile phase V methanol∶V water(0.4%phosphoric acid solution)=55∶45 at a flow rate of 1 ml/min,and the eluate was detected with a shimadzu HPLC ultraviolet detector at 360 nm.[Results]With kaempferol as the reference substance,the correlation coefficient was 0.999 2 in the range of 0.001 06-0.016 96 g/L.The content in the fermented product was less than that in the non-fermented product by 28%.[Conclusions]The method is simple,accurate,and is suitable for determination of kaempferol.This study will provide an experimental basis for the development and utilization of ginkgo. 展开更多

[目的]研究银杏叶斑病病叶中黄酮含量的变化。[方法]选择患不同程度银杏叶斑病的银杏病叶,采用乙醇浸提法提取叶中黄酮,同时用紫外分光光度法测定叶中黄酮含量,探讨黄酮在叶斑病致病过程中含量变化规律,确定患叶斑病叶片是否适合采摘再利用,同时也为阐明黄酮在叶斑病致病过程中是否可能起到抵抗病原菌侵染作用提供初步研究。[结果]随着叶斑病患病程度的加深,叶片上侵染面积的增大,叶中黄酮含量有所增加;但当损伤达到一定程度时,黄酮含量不再增加。[结论]患病程度轻的银杏叶片具有采摘再利用的价值,同时因为黄酮具有较强的抗菌活性,说明黄酮含量升高可能是银杏叶抵抗叶斑病病原菌侵染机制之一。 展开更多

葡萄酒的抗氧化功能一直是葡萄酒消费者与爱好者较为关心的问题,适量饮用葡萄酒对心脏病、动脉粥样硬化、血小板凝聚等心血管疾病以及中风和某些癌症有一定的预防与抑制的作用,主要是因为葡萄酒中含有具有抗氧化、清除自由基功能的各种... 葡萄酒的抗氧化功能一直是葡萄酒消费者与爱好者较为关心的问题,适量饮用葡萄酒对心脏病、动脉粥样硬化、血小板凝聚等心血管疾病以及中风和某些癌症有一定的预防与抑制的作用,主要是因为葡萄酒中含有具有抗氧化、清除自由基功能的各种多酚类物质。银杏中含有多酚类、黄酮类等多种生物活性物质。以葡萄酒为依托酿造银杏葡萄酒,开发一种新型保健酒,不但具有较好的营养保健作用,而且风味口感协调。试验对总酚含量以及抗氧化活性进行测定,并研究两者之间的相关性,为消费者选择有益于身体健康的葡萄酒提供有力参考。结果表明:银杏葡萄酒的总酚含量变化范围介于1 588.76~2 130.71 mg/L之间,均高于对照葡萄酒,当银杏酶解液添加量为20%时,银杏葡萄酒中总酚含量最高,达到2 130.71 mg/L,且自由基清除活性较高。根据研究结果可知银杏葡萄酒中多酚类物质与其抗氧化活性之间关系密切,并呈现出良好的相关性。 展开更多

Analysis of spontaneous adverse drug reactions to echinacea, valerian,black cohosh and ginkgo in Australia from 2000 to 2015

Objective: Assessing adverse drug reactions(ADRs) is a proven method to estimate the safety of medicines. The ADRs to herbal medicines in Australia(and by inference, the safety of herbal medicines in Australia) remain unknown. This study examines spontaneous ADR cases to four of the most popular herbs in Australia from 2000 to 2015: echinacea(Echinacea purpurea), valerian(Valeriana officinalis), black cohosh(Actaea racemosa) and ginkgo(Ginkgo biloba).Methods: ADRs of echinacea, valerian, black cohosh and ginkgo reported to the Australian Therapeutic Goods Administration(TGA) between 2000 and 2015 were obtained from the TGA database. Data were collated and analysed according to age, sex, severity, type of ADR and body system affected. Statistics were calculated using Graph Pad Prism software.Results: Most ADRs were mild or moderate. However, every herbal medicine was associated with lifethreatening ADRs. In each life-threatening case, the herbal medicine was taken concomitantly with prescription medications. Black cohosh was associated with a significant number of severe ADRs(30.3% of the total), with 39.4% of these ADRs being associated with abnormal hepatic function, hepatitis or hepatotoxicity.Conclusion: This study highlights the lack of public awareness with regard to herb–drug interactions,since most of the severe ADRs involved a herb–drug interaction. 展开更多

[Objectives] To optimize the extraction of total flavone in folium Ginkgo biloba leaves.[Methods] Using the ultrasonic extraction method, the three levels of maximum impact were selected by each factor selection, and the optimal extraction technology was obtained by box-benhnken test design analysis.[Results] After analyzing the results, the optimal process was as follows: a time of 28.89 min and liquid/material ratio of 31.38∶ 1, the temperature was 40.74℃. In this way, NaNO 2-Al (NO 3) 3-NaOH was used as color developing agent, and the theoretical extraction of flavone in ginkgo leaf can reach 11.74 mg/g.[Conclusions] This method can be used to extract total flavonoids from G. biloba leaves with simple operation, low cost and good repeatability. 展开更多

[Objectives]This study was conducted to determine the content of total flavonoids in ginkgo ( Ginkgo biloba L.) leaves.[Methods]The content of total flavonoids from ginkgo leaves was determined by reversed phase-high performance liquid chromatography (RP-HPLC).The flavonoid glycosides were first extracted with methanol,and hydrolyzed with hydrochloric acid solution to prepare a test solution.Platisil ODS C18 column (150 mm×4.6 mm,5 μm) and the mobile phase V methanol ∶ V water (0.4% phosphoric acid solution)=85∶ 15 were selected for HPLC separation.The HPLC separation was performed with the column at a column temperature of 25℃ using the mobile phase at a flow rate of 1 ml/min.The sample size was 10 μl,and detection was performed with an Agilent HPLC ultraviolet detector at 360 nm.[Results]The reference substance,quercetin,had good linearity in the range of 0.002 6-0.052 0 g/L,with a correlation coefficient of 0.999 7;and the RSD was 1.26%.[Conclusions]The determination method has rapid and simple operation with accurate results and is good in repeatability.This method is suitable for the determination of content of total flavonoids in ginkgo leaves. 展开更多

银杏柠檬酸淀粉酯的制备及加工特性研究

银杏种仁的主要营养成分是淀粉。以银杏淀粉为原料,柠檬酸为酯化剂制备银杏柠檬酸淀粉酯,考察柠檬酸添加量、pH、反应温度、反应时间对柠檬酸淀粉酯取代度(DS)的影响,通过正交试验确定了银杏柠檬酸淀粉酯的最佳制备工艺,并分析比较了银... 银杏种仁的主要营养成分是淀粉。以银杏淀粉为原料,柠檬酸为酯化剂制备银杏柠檬酸淀粉酯,考察柠檬酸添加量、pH、反应温度、反应时间对柠檬酸淀粉酯取代度(DS)的影响,通过正交试验确定了银杏柠檬酸淀粉酯的最佳制备工艺,并分析比较了银杏淀粉和银杏柠檬酸淀粉酯的透明度、膨胀率、溶解度、持水率等加工特性。结果表明:柠檬酸添加量和pH对DS的影响显著(p<0.05),反应温度对DS的影响不显著(p>0.05)。银杏柠檬酸淀粉酯的最佳制备工艺为:柠檬酸添加量8mL,pH2.0,反应温度132℃,反应时间6.0h。在此条件下制备得到的银杏柠檬酸淀粉酯平均取代度DS为0.355。银杏柠檬酸淀粉酯的透光率、溶解度、持水率和膨胀率分别比银杏淀粉提高了51.35%、35.66%、120.37%、16.10%。这些加工特性的提高,适应了食品工业的要求,提高了淀粉的加工性能。 展开更多

目的:研究银杏叶提取物EGB761对缺氧原代乳鼠心肌细胞凋亡相关基因的影响及分子机制。方法:大鼠乳鼠1d提取心肌原代细胞培养,分对照组、缺氧组、缺氧+EGB7615μg/ml组、缺氧+EGB76110μg/ml组、缺氧+EGB76120μg/ml组,MTT法检测细胞存... 目的:研究银杏叶提取物EGB761对缺氧原代乳鼠心肌细胞凋亡相关基因的影响及分子机制。方法:大鼠乳鼠1d提取心肌原代细胞培养,分对照组、缺氧组、缺氧+EGB7615μg/ml组、缺氧+EGB76110μg/ml组、缺氧+EGB76120μg/ml组,MTT法检测细胞存活率、ELISA法检测细胞上清LDH及细胞匀浆中SOD的活性变化、Hoechst染色检测细胞凋亡情况、PCR及Western-blot法检测PPAR-γ、NF-κB-p65、I-κB、Bax、Bcl-2、Caspase-3mRNA及蛋白表达。结果:和对照组相比,缺氧6h时细胞存活率明显下降,且随着加入EGB761浓度增加,细胞存活率逐渐升高。EGB761(10μg/ml和20μg/ml浓度)可有效降低(P<0.05)缺氧造成的LDH漏出和凋亡增加,不同浓度EGB761均可升高细胞内SOD活性(P<0.05)。缺氧造成PPAR-γ、I-kb、抗凋亡因子Bcl-2表达下降;NF-κB-p65基因、促凋亡因子Bax、Caspase-3表达上升,不同浓度的EGB761可逆转缺氧引起的相关因子表达变化(P<0.05)。结论:银杏叶提取物EGB761可调控PPAR-γ、NF-κB-p65及凋亡关键基因的表达,并逆转缺氧引起的心肌细胞凋亡增加、存活率下降等损伤效应。 展开更多

Stem cell transplantation has brought new hope for the treatment of neurological diseases.The key to stem cell therapy lies in inducing the specific differentiation of stem cells into nerve cells.Because the differentiation of stem cells in vitro and in vivo is affected by multiple factors,the final differentiation outcome is strongly associated with the microenvironment in which the stem cells are located.Accordingly,the optimal microenvironment for inducing stem cell differentiation is a hot topic.EGb761 is extracted from the leaves of the Ginkgo biloba tree.It is used worldwide and is becoming one of the focuses of stem cell research.Studies have shown that EGb761 can antagonize oxygen free radicals,stabilize cell membranes,promote neurogenesis and synaptogenesis,increase the level of brain-derived neurotrophic factors,and replicate the environment required during the differentiation of stem cells into nerve cells.This offers the possibility of using EGb761 to induce the differentiation of stem cells,facilitating stem cell transplantation.To provide a comprehensive reference for the future application of EGb761 in stem cell therapy,we reviewed studies investigating the influence of EGb761 on stem cells.These started with the composition and neuropharmacology of EGb761,and eventually led to the finding that EGb761 and some of its important components play important roles in the differentiation of stem cells and the protection of a beneficial microenvironment for stem cell transplantation. 展开更多

OBJECTIVE:To assess the clinical efficacy and safety of ginkgo injections(GIs)combined with conventional treatment(CT)against angina pectoris(AP)due to coronary heart disease(CH D).METHODS:Randomized controlled trials(RCTs)that used GIs to treat AP were searched in SinoMed,PubMed,China National Knowledge Infrastructure Database,Chinese Scientific Journals Database,Wanfang Database,Embase and Cochrane Library until March 2017.The Cochrane "risk of bias" method was used to evaluate the methodological quality of RCTs.Data were analyzed using Stata v13.1 and WinBUGS v1.4.3.RESULTS:A total of 73 RCTs involving 7621 patients were included.A Bayesian network Meta-analysis of RCTs was undertaken,and the advantages of four types of Gl-supplemented CT in AP treatment were determined.CONCLUSION:Gl-assisted CT was more effective against AP than CT alone.However,based on the limitations of the study,additional high-quality RCTs are required to confirm our findings. 展开更多

Ginkgo biloba extracts (GBEs), a natural herb extract w... Abdominal aortic aneurysms (AAAs) are a chronic vascular disease characterized by pathological luminal dilation. Aortic rupture is the fatal consequence of AAAs. Ginkgo biloba extracts (GBEs), a natural herb extract widely used as food supplements, drugs, and cosmetics, has been reported to suppress development of calcium chloride-induced AAAs in mice. Calcium chloride-induced AAAs do not rupture, while angiotensin II (AngII)-induced AAAs in mice have high rate of aortic rupture, implicating potentially different mechanisms from calcium chloride-induced AAAs. This study aimed to determine whether GBE would improve aortic dilation and rupture rate of AngII-induced AAAs. Male apolipoprotein E (apoE) -/- mice were infused with AngII and administered either GBE or its major active ingredients, flavonoids and ginkgolides, individually or in combination. To determine the effects of GBE in mice with established AAAs, male apoE-/- mice were flrstly infused with AngII for 28 days to develop AAAs, and then administered either GBE or vehicle in mice with established AAAs, which were continuously infused with AngII for another 56 days. GBE, but not the two major active components separately or synergistically, prevented aortic rupture, but not aortic dilation. The protection of GBE from aortic rupture was independent of systolic blood pressure, lipid, and inflammation. GBE also did not attenuate either aortic rupture or progressive aortic dilation in mice with established AAAs. GBE did not reduce the atherosclerotic lesion areas, either. In conclusion, GBE prevents aortic rupture in AngII-infused hypercholesterolemic mice, but only in the early phase of the disease development. 展开更多

The response of soil fauna to the litter decomposition process has received considerable attention,but this effect has not been fully examined in agroforestry systems.A 1-year in situ decomposition experiment was carried out in a pure ginkgo plantation and two ginkgo agroforestry systems using a litterbag method(11 different treatments were tested in three systems).We found that the application of different organic materials(crop residues)produced positive effects on the number of soil fauna in the ginkgo planting systems;the mixture of ginkgo leaves and corn leaves was the best performing treatment.Collembola and Acarina were the predominant groups in the litter bags and were mainly responsible for the differences among the treatments.Litter mixing promoted the abundance,richness,and diversity of soil fauna,and significant differences regarding the Shannon–Wiener index of the soil fauna were observed among the 11 treatments in July.Significantly higher soil MBC(microbial biomass carbon)and MBN(microbial biomass nitrogen)were observed in agroforestry systems than in pure ginkgo plantations.These results suggest that the practice of intercrop residue application plays an important role in enhancing soil ecosystem function in ginkgo agroforestry systems and may ultimately contribute to sustainable intercrop production,soil fertility,and local economic diversity. 展开更多

目的:分析银杏达莫配合依达拉奉应用于急性脑梗死,对患者神经功能与应激指标的影响。方法:选取2016年4月至2017年4月朝阳市中心医院收治的急性脑梗死患者112例,按照治疗方案的不同分为对照组(n=54)和观察组(n=58)。对照组予以依达拉奉治... 目的:分析银杏达莫配合依达拉奉应用于急性脑梗死,对患者神经功能与应激指标的影响。方法:选取2016年4月至2017年4月朝阳市中心医院收治的急性脑梗死患者112例,按照治疗方案的不同分为对照组(n=54)和观察组(n=58)。对照组予以依达拉奉治疗,观察组予以依达拉奉+银杏达莫治疗,于治疗前1d、治疗14d后检测2组神经递质指标、应激指标、神经功能NIHSS评分,比较2组治疗14d内的疗效。结果:1)观察组在治疗后AOPP(75.12±3.74)μmol/L、IL-6(71.62±5.01)pg/mL、ox-LDL(85.80±5.26)U/mL、CRP(9.18±0.55)mg/L、MDA(3.08±0.56)nmol/mL、IL-10(22.13±1.62)μg/L显著低于对照组,GSH-Px(39.18±6.28)mg/mL、SOD(162.54±13.15)U/mL显著高于对照组(P<0.05);(2)观察组在治疗后VAP(8.12±1.74)ng/L、NPY(170.52±6.15)μg/L、NSE(12.13±1.62)μg/L、Glu(71.62±4.01)μmol/L显著低于对照组,NGF(89.18±3.28)pg/mL、GABA(9.08±0.86)μmol/L、NTF(5.80±0.76)ng/mL、NAA(429.18±17.55)mmol/L显著高于对照组(P<0.05);3)观察组在治疗后NIHSS评分总分(11.50±1.62)分显著低于对照组(18.00±1.78)分(P<0.05);4)观察组总有效率(96.55%)显著高于对照组(87.04%)(P<0.05)。结论:对于急性脑梗死,可采取银杏达莫辅助依达拉奉治疗,比较单纯应用依达拉奉,能够有效调节患者应激反应、神经递质表达,提高患者神经功能,疗效较好。 展开更多

[Objectives]This study was conducted to study the extraction process and the content determination of flavonoids in ginkgo(Ginkgo biloba L.)leaves.[Methods]Ethanol extraction and methanol extraction of total flavonoids in ginkgo leaves were studied,and the optimal extraction conditions for flavonoids were determined by orthogonal test;and with quercetin as reference substance,total flavonoid content in ginkgo leaves was determined by UV spectrophotometry.[Results]The optimal extraction process was 4 h of Soxhlet extraction with methanol;and the total flavonoid contents had a good linear relation in the range of 0.006 5-0.039 mg/ml(R 2=0.999 9),the average content was stabilized at 1.135%,and the average recovery of the method was 102.0%.[Conclusions]This study selected the optimal extraction process for total flavonoids in ginkgo leaves.The test method is simple with high accuracy and precision,and is suitable for the extraction and determination of total flavonoids in ginkgo leaves. 展开更多

Evidence continues to grow on potential health risks associated with Ginkgo biloba and its constituents.While biflavonoid is a subclass of the flavonoid family in Ginkgo biloba with a plenty of pharmacological properties,the potential toxicological effects of biflavonoids remains largely unknown.Thus,the aim of this study was to investigate the in vitro and in vivo toxicological effects of the biflavonoids from Ginkgo biloba(i.e.,amentoflavone,sciadopitysin,ginkgetin,isoginkgetin,and bilobetin).In the in vitro cytotoxicity test,the five biflavonoids all reduced cell viability in a dose-dependent manner in human renal tubular epithelial cells(HK-2)and human normal hepatocytes(L-02),indicating they might have potential liver and kidney toxicity.In the in vivo experiments,after intragastrical administration of these biflavonoids at 20 mg·kg^–1·d^–1 for 7 days,serum biochemical analysis and histopathological examinations were performed.The activity of alkaline phosphatase was significantly increased after all the bifl avonoid administrations and widespread hydropic degeneration of hepatocytes was observed in ginkgetin or b ilobetin-treated mice.Moreover,the five biflavonoids all induced acute kidney injury in treated mice and the main pathological lesions were confirmed to the tubule,glomeruli,and interstitium injuries.As the in vitro and in vivo results suggested that these biflavonoids may be more toxic to the kidney than the liver,we further detected the mechanism of biflavonoids-induced nephrotoxicity.The increased TUNEL-positive cells were detected in kidney tissues of biflavonoids-treated mice,accompanied by elevated expression of proapoptotic protein BAX and unchanged levels of antiapoptotic protein BCL-2,indicating apoptosis was involved in biflavonoids-induced nephrotoxicity.Taken together,our results suggested that the five biflavonoids from Ginkgo biloba may have potential hepatic and renal toxicity and more attentions should be paid to ensure Ginkgo biloba preparations safety. 展开更多
