转OvBAN/bar双价基因的紫花苜蓿对虫蚀及除草剂的耐受性分析 预览

以"甘农3号"紫花苜蓿为受体,利用农杆菌介导法导入双价基因OvBAN/bar,通过PCR检测和Southern杂交分析表明,4个转基因株系含有目的基因且均出现杂交条带,其中2个株系为单拷贝,1个株系为双拷贝,1个株系为三拷贝;对获得的4株转... 以"甘农3号"紫花苜蓿为受体,利用农杆菌介导法导入双价基因OvBAN/bar,通过PCR检测和Southern杂交分析表明,4个转基因株系含有目的基因且均出现杂交条带,其中2个株系为单拷贝,1个株系为双拷贝,1个株系为三拷贝;对获得的4株转基因苜蓿进行qRT-PCR分析表明,具有三拷贝数的1个转基因株系OvBAN基因的表达量最高,其次为双拷贝数的2个转基因株系,并且这些株系的花青素还原酶活性及缩合单宁含量均显著高于野生型苜蓿。以过量表达OvBAN基因的3株转基因苜蓿为试验材料,进行抗蚜性和除草剂耐受性分析。结果表明,与野生型苜蓿相比,转OvBAN/bar基因苜蓿对苜蓿蚜都具有较好的抗性,蚜虫抑制率为78%~93%;用0.5%的市售草铵膦喷施各株系,10d后各转基因株系除个别叶片枯萎死亡,其他叶片及茎秆生长状况良好,而野生型苜蓿全部枯萎死亡,表明bar基因已整合到转基因苜蓿基因组中并稳定表达。综上所述,转OvBAN/bar基因紫花苜蓿表现出良好的抗虫性和除草剂耐受性。 展开更多

Children presenting with partial physeal arrest and significant remaining growth may benefit from physeal bar resection,although the operation is a technique demanding procedure.This study evaluates the treatment of post-traumatic pediatric ankle varus deformity using physeal bar resection and hemi-epiphysiodesis with the assistance of two operative methods.Forty-five patients presenting with a distal tibial medial physeal bridge as well as ankle varus deformity following traumatic ankle physeal injury between 2009 and 2017 were followed.These patients were treated w让h physeal bar resection and hemi-epiphysiodesis,with the assistance of either fluoroscopy (10 cases) or intraoperative three-dimensional navigation (35 cases).Of the 45 cases,the median age was 9.0 years (range:3-14 years) with 28 male and 17 female patients.The median of pre-operation ankle varus angle was 20 degrees (IQR 15-25) and 5 degrees (IQR 0-20) at the time of final follow up,representing a statistically significant difference (P<0.05).No differences were observed with regards to age,gender,and surgical history between effective group and ineffective group (P>0.05).The median of pre-operative ankle varus angles of the navigation and fluoroscopy groups were both 20 degrees (P>0.05).The median correction angle of the navigation and fluoroscopy groups was 10 and 15 degrees,respectively (P>0.05).Our results indicate that physeal bar resection and hemiepiphysiodesis are effective treatments for correcting ankle varus deformity due to traumatic medial physeal arrest of the distal tibia.We observe no difference in outcome between fluoroscopy group and three-dimensional navigation group during the procedures. 展开更多

A prime requirement for hydrological applications,such as sediment budgeting or numerical modelling,is that produced Digital Terrain Models(DTMs)accurately represent the shape of landforms,especially for river reaches where data are not homogeneous.DTM error is a function of data point measurement accuracy and density and also of the field survey strategy when limited amounts of data will be acquired.This paper aims to advance the importance of the field survey strategy for the specific,but common cases,where only limited topographic data will be available.This methodology is based on the idea that any feature can be properly described by a set of cross sections and breaklines describing both main and secondary directions of the flow.Then,a longitudinal linear interpolation can be applied to the defined homogeneous zones.This morphologically oriented(MO)method that includes data acquisition strategy and interpolation,was validated using a reference DTM derived from LiDAR measurements.An estimation of the uncertainties also is suggested based on the distance of the nearest point and the local slope using a geographically weighted regression.The proposed MO method is typically applicable to Alpine river reaches characterized by multiple channels that may always be underwater and not navigable such as an alternate bar system with secondary and transverse channels. 展开更多

Based on the characteristics of hydrodynamics and sediment transport in the bar area in the Modaomen Estuary,a flume experiment was performed to study the evolution of the longitudinal profile of the mouth bar.The mouth bar evolution was investigated under the impacts of floods with different return periods as well as flood-wave interaction.The results showed that floods with different return periods had significant influences on the evolution of the river mouth bar.Particularly on the inner slope of the mouth bar,the sediment was substantially active and moveable.The inner slope and the bar crest tended to be remarkably scoured.The erosion was intensified with the increase of the magnitude of floods.Moreover,the bar crest moved seawards,while the elevation of the bar crest barely changed.Under the flood-wave interaction,a remarkable amount of erosion on the inner and outer slopes of the mouth bar was also found.The seaward displacement of the bar crest under the interaction of floods and waves was less than it was under only the impact of floods,while more deposition was found on the crest of the mouth bar in this case. 展开更多

Core, well logging and seismic data were used to investigate sandbody architectural characteristics within Lower Member of Minghuazhen Formation in Neogene, Bohai BZ25 Oilfield, and to analyze the sedimentary microfacies, distribution and internal architecture characteristics of the bar finger within shoal water delta front. The branched sand body within shoal water delta front is the bar finger, consisting of the mouth bar, distributary channel over bar, and levee. The distributary channel cuts through the mouth bar, and the thin levee covers the mouth bar which is located at both sides of distributary channel. The bar finger is commonly sinuous and its sinuosity increases basinward. The distributary channel changes from deeply incising the mouth bar to shallowly incising top of the mouth bar.The aspect ratio ranges from 25 to 50 and there is a double logarithmic linear positive relationship between the width and thickness for the bar finger, which is controlled by base-level changing in study area. For the bar finger, injection and production in the same distributary channel should be avoided during water flooding development. In addition, middle–upper distributary channel and undrilled mouth bar are focus of tapping remaining oil. 展开更多

Cemented tailings backfill(CTB)have increasingly been used in recent years to improve the stability of mining stopes in deep underground mines.Deep mining processes are often associated with rock bursting and high-speed dynamic loading conditions.Therefore,it is important to investigate the characteristics and dynamic mechanical behavior of CTB.This paper presents the results of dynamic tests on CTB specimens with different cement and solid contents using a split Hopkinson pressure bar(SHPB).The results showed that some CTB specimens exhibited one to two lower stress peaks after reaching dynamic peak stress before they completely failed.The greater the cement-to-tailings ratio is,the more obvious the strain reaction.This property mainly manifested as follows.First,the dynamic peak stress increased with the increase of the cement-to-tailings ratio when the impact velocity was fixed.Second,the dynamic peak stress had a quadratic relationship with the average stress rate.Third,the cement-to-tailings ratio could enhance the increase rate of dynamic peak stress with strain rate.In addition,the dynamic strength enhancement factor K increased with the increase of strain rate,and its value was larger than that of the rock samples.The failure modes of CTB specimens under low-speed impact were tensile failure and X conjugate shear failure,where were nearly the same as those under static uniaxial and triaxial compression.The CTB specimens were crushed and broken under critical strain,a failure mode similar to that of low-strength concrete.The results of the experimental research can improve the understanding of the dynamic mechanical properties of CTB and guide the strength design of deep mining backfills. 展开更多

本研究成功建立了一个全新的以博落回愈伤为外植体的遗传转化体系。该遗传体系将抗草丁膦的Bar基因整合到pcambia2301质粒,构建植物载体,通过农杆菌介导转染博落回愈伤,选用Bar基因为筛选标记,采用PPT进行筛选,对转染的愈伤和组培... 本研究成功建立了一个全新的以博落回愈伤为外植体的遗传转化体系。该遗传体系将抗草丁膦的Bar基因整合到pcambia2301质粒,构建植物载体,通过农杆菌介导转染博落回愈伤,选用Bar基因为筛选标记,采用PPT进行筛选,对转染的愈伤和组培苗用PCR、RT-PCR、GUS染色等方法验证为阳性,获得了抗除草剂的博落回植株。该遗传体系相比以叶片为外植体、抗生素为筛选标记的遗传体系,更能加快转化时间,缩短转化周期,而且假阳性更低,对于博落回生物学研究具有一定帮助。 展开更多
