Sodium-potassium-chloride cotransporter 1 (NKCC1) and potassium-chloride cotransporter 2 (KCC2) are associated with the transmission of peripheral pain.We investigated whether the increase of NKCC1 and KCC2 is associated with peripheral pain transmission in dorsal root ganglion neurons.To this aim,rats with persistent hyperalgesia were randomly divided into four groups.Rats in the control group received no treatment,and the rat sciatic nerve was only exposed in the sham group.Rats in the chronic constriction injury group were established into chronic constriction injury models by ligating sciatic nerve and rats were given bumetanide,an inhibitor of NKCC1,based on chronic constriction injury modeling in the chronic constriction injury + bumetanide group.In the experiment measuring thermal withdrawal latency,bumetanide (15 mg/kg) was intravenously administered.In the patch clamp experiment,bumetanide (10 μg/μL) and acutely isolated dorsal root ganglion neurons (on day 14) were incubated for 1 hour,or bumetanide (5 μg/μL) was intrathecally injected.The Hargreaves test was conducted to detect changes in thermal hyperalgesia in rats.We found that the thermal withdrawal latency of rats was significantly decreased on days 7,14,and 21 after model establishment.After intravenous injection of bumetanide,the reduction in thermal retraction latency caused by model establishment was significantly inhibited.Immunohistochemistry and western blot assay results revealed that the immune response and protein expression of NKCC1 in dorsal root ganglion neurons of the chronic constriction injury group increased significantly on days 7,14,and 21 after model establishment.No immune response or protein expression of KCC2 was observed in dorsal root ganglion neurons before and after model establishment.The Cl^– (chloride ion) fluorescent probe technique was used to evaluate the change of Cl^– concentration in dorsal root ganglion neurons of chronic constriction injury model rats.We found that the relative optical density of N-(et 展开更多

BACKGROUND:Penehyclidine is a newly developed anticholinergic agent.We aimed to investigate the role of penehyclidine in acute organophosphorus pesticide poisoning(OP)patients.METHODS:We searched the Pubmed,Cochrane library,EMBASE,Chinese National Knowledge Infrastructure(CNKI),Chinese Biomedical literature(CBM)and Wanfang databases.Randomized controlled trials(RCTs)recruiting acute OP patients were identifi ed for meta-analysis.Main outcomes included cure rate,mortality rate,time to atropinization,time to 60%normal acetylcholinesterase(AchE)level,rate of intermediate syndrome(IMS)and rate of adverse drug reactions(ADR).RESULTS:Sixteen RCTs involving 1,334 patients were identifi ed.Compared with the atropineor penehyclidine-alone groups,atropine combined with penehyclidine significantly increased the cure rate(penehyclidine+atropine vs.atropine,0.97 vs.0.86,RR 1.13,95%CI[1.07–1.19];penehyclidine+atropine vs.penehyclidine,0.93 vs.0.80,RR 1.08,95%CI[1.01–1.15])and reduced the mortality rate(penehyclidine+atropine vs.atropine,0.015 vs.0.11,RR 0.17,95%CI[0.06–0.49];penehyclidine+atropine vs.penehyclidine,0.13 vs.0.08,RR 0.23,95%CI[0.04–1.28]).Atropine combined with penehyclidine in OP patients also helped reduce the time to atropinization and AchE recovery,the rate of IMS and the rate of ADR.Compared with a single dose of atropine,a single dose of penehyclidine also signifi cantly elevated the cure rate,reduced times to atropinization,AchE recovery,and rate of IMS.CONCLUSION:Atropine combined with penehyclidine benefi ts OP patients by enhancing the cure rate,mortality rate,time to atropinization,AchE recovery,IMS rate,total ADR and duration of hospitalization.Penehyclidine combined with atropine is likely a better initial therapy for OP patients than atropine alone. 展开更多

Zebrafish and human genomes are highly homologous;however,despite this genomic similarity,adult zebrafish can achieve neuronal proliferation,regeneration and functional restoration within 6–8 weeks after spinal cord injury,whereas humans cannot.To analyze differentially expressed zebrafish genes between axon-regenerated neurons and axon-non-regenerated neurons after spinal cord injury,and to explore the key genes and pathways of axonal regeneration after spinal cord injury,microarray GSE56842 was analyzed using the online tool,GEO2R,in the Gene Expression Omnibus database.Gene ontology and protein-protein interaction networks were used to analyze the identified differentially expressed genes.Finally,we screened for genes and pathways that may play a role in spinal cord injury repair in zebrafish and mammals.A total of 636 differentially expressed genes were obtained,including 255 up-regulated and 381 down-regulated differentially expressed genes in axon-regenerated neurons.Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment results were also obtained.A protein-protein interaction network contained 480 node genes and 1976 node connections.We also obtained the 10 hub genes with the highest correlation and the two modules with the highest score.The results showed that spectrin may promote axonal regeneration after spinal cord injury in zebrafish.Transforming growth factor beta signaling may inhibit repair after spinal cord injury in zebrafish.Focal adhesion or tight junctions may play an important role in the migration and proliferation of some cells,such as Schwann cells or neural progenitor cells,after spinal cord injury in zebrafish.Bioinformatic analysis identified key candidate genes and pathways in axonal regeneration after spinal cord injury in zebrafish,providing targets for treatment of spinal cord injury in mammals. 展开更多

目的了解2017年上海市细菌耐药性监测结果。方法采用纸片扩散法或自动化仪器法对上海市49所医院的临床分离菌进行药敏试验。包括30所三级医院(含3所儿童医院)和19所二级医院。采用美国临床和实验室标准化协会(CLSI)2017年版标准判断结... 目的了解2017年上海市细菌耐药性监测结果。方法采用纸片扩散法或自动化仪器法对上海市49所医院的临床分离菌进行药敏试验。包括30所三级医院(含3所儿童医院)和19所二级医院。采用美国临床和实验室标准化协会(CLSI)2017年版标准判断结果。结果共获134 078株临床分离菌,其中革兰阳性菌40 243株,占30.0%;革兰阴性菌93 835株,占70.0%。二级医院和三级医院菌株分别占29.0%和71.0%。在二、三级医院中革兰阳性菌和革兰阴性菌各为25.4%和74.6%、31.9%和68.1%。MRSA和MRCNS在各自菌种中的检出率分别为49.1%和79.7%。二级医院、三级医院的MRSA和MRCNS检出率分别为53.2%和72.1%、47.7%和82.2%。葡萄球菌属中未发现万古霉素耐药株。1 041株儿童非脑膜炎肺炎链球菌分离株中青霉素敏感(PSSP)、中介(PISP)和耐药株(PRSP)分别占84.3%、7.8%和7.9%;330株成人分离株PSSP、PISP和PRSP分别为89.4%、4.2%和6.4%。检出5株(0.1%)万古霉素耐药粪肠球菌、64株(1.2%)万古霉素耐药屎肠球菌,经过PCR测序确认的菌株中主要为vanA基因型。大肠埃希菌、肺炎克雷伯菌和奇异变形杆菌中ESBL的检出率分别为54.9%、35.1%和47.6%。上述产ESBL菌株在二级医院和三级医院的检出率分别为53.7%、31.5%、44.0%和55.5%、37.0%、50.0%。肠杆菌科细菌对碳青霉烯类抗生素仍高度敏感,对亚胺培南和美罗培南的总耐药率分别为11.1%和11.0%,二级医院、三级医院分别为9.0%和9.2%、12.2%和11.9%,碳青霉烯类耐药肠杆菌科细菌(CRE)的检出率为11.9%,其中73.4%是肺炎克雷伯菌。鲍曼不动杆菌、肺炎克雷伯菌、铜绿假单胞菌和大肠埃希菌中出现对大多数测试药物耐药而仅对1~2种抗菌药物敏感的广泛耐药株,二级医院和三级医院分别为64株、357株、7株、25株和412株、1 129株、38株、45株。结论目前主要临床分离菌对于常用抗菌药物的耐药性仍甚为严重,尤其是多重耐药 展开更多

Aim: The aim of the research is to determine characteristic of the correlation between climate and stroke through the analysis of relevant documents. Methods: We found 115 researches of temperature and season on stroke mortality/incidence and these were categorized according to location, meteorology, date span, data source and research sites’ latitudes. We divided the globe into 5 temperature zones based on latitude ranges: 0 - 10, 10 - 23.5, 23.5 - 40, 40 - 50, and >50 degrees, and allotted a zone to each country. Stroke mortality by five temperature zones was calculated per 100,000 people. Results: 70% of the researches came from latitudes greater than 40 degrees with 92% of researches located in Europe, USA, and Asia. The mortality range was 42 - 63 per 105 individuals within latitudes 0 - 40 degrees. In latitudes 40 - 66.5 degrees, a linear upward trend (y = 69.82x &#8722;22.823, R2 = 0.99) was noticed. More than 75% of the research reports indicated a negative correlation between climate and stroke, and the proportion was almost 6 and 7 times greater than that of the fluctuation and non-correlation, respectively. The most frequently used research methods were regression analysis and time series analysis. Conclusion: All of the research results confirmed that lower temperature is associated with higher mortality and incidence of stroke, while higher latitude is correlated with higher stroke mortality, consistent with the temperature zones. 展开更多

以聚偏氟乙烯-六氟丙烯(PVDF-HFP)/聚碳酸丙烯酯(PPC)为聚合物基体,在聚合物中添加Li5.5La3Zr1.75Ta0.25O12(LLZTO)颗粒、并利用无纺布作为骨架来制备复合凝胶聚合物电解质(CGPE)。当CGPE中的PVDF-HFP/PPC质量比为2∶1、LLZTO颗粒在聚... 以聚偏氟乙烯-六氟丙烯(PVDF-HFP)/聚碳酸丙烯酯(PPC)为聚合物基体,在聚合物中添加Li5.5La3Zr1.75Ta0.25O12(LLZTO)颗粒、并利用无纺布作为骨架来制备复合凝胶聚合物电解质(CGPE)。当CGPE中的PVDF-HFP/PPC质量比为2∶1、LLZTO颗粒在聚合物中的质量百分比为5wt.%时,CGPE表现出3.5×10-4S cm-1的离子电导率。同时,CGPE膜中有无纺布做支撑,具有较好的机械强度,在同样大小的拉伸强度下,CGPE的伸长量是商用隔膜的三倍,表现出良好的韧性。以该CGPE为电解质的锂硫电池表现出良好的循环稳定性能,0.1C下充放电,首次放电比容量为896mAh g-1,首次库伦效率高于液态电池,循环100次后仍有74%的容量剩余,容量保持率高于液态电池。 展开更多

两种栓塞方法行超选择性肝动脉栓塞术治疗多囊肝病的对比研究

目的比较两种栓塞方法行超选择性肝动脉栓塞术(TAE)治疗多囊肝病的疗效及安全性。方法回顾分析解放军总医院第一医学中心2008年6月至2016年6月行TAE的41例多囊肝病患者资料,其中男性5例,女性36例,年龄36~68岁。根据栓塞方案不同分为观察... 目的比较两种栓塞方法行超选择性肝动脉栓塞术(TAE)治疗多囊肝病的疗效及安全性。方法回顾分析解放军总医院第一医学中心2008年6月至2016年6月行TAE的41例多囊肝病患者资料,其中男性5例,女性36例,年龄36~68岁。根据栓塞方案不同分为观察组(n=14)和对照组(n=27)。观察组采用博来霉素+α氰基丙烯酸正丁酯+碘油,对照组为α氰基丙烯酸正丁酯+碘油。记录术后有无并发症。术后3、6个月及此后每6个月定期复查,评估症状改善情况、腹围、肝内囊肿总体积及肝脏总体积。结果两组均成功行TAE,无严重并发症。观察组术后3、6、12、24个月症状明显改善率为92.9%(13/14)、92.9%(13/14)、92.9%(13/14)、92.3%(12/13),术后24个月失访1例。对照组术后3、6、12、24个月症状明显改善率为88.9%(24/27)、85.2%(23/27)、84.6%(22/26)、81.8%(18/22)。术后12个月失访1例,术后24个月失访5例。与本组术前比较,两组术后腹围均减小,术后6、12、24个月肝内囊肿总体积及肝脏总体积均减小,差异有统计学意义(P<0.05)。结论应用博来霉素+α氰基丙烯酸正丁酯+碘油及α氰基丙烯酸正丁酯+碘油行TAE治疗多囊肝病安全、有效,且前者疗效优于后者。 展开更多

【目的】逐步采用机械化直播的集约栽培模式已是水稻种植的主要趋势之一,本文研究了机械化水稻直播条件下,缓释尿素与普通尿素不同配施对水稻生长和产量的影响特征,为直播稻的高产高效施肥技术提供理论基础。【方法】以优质杂交籼稻‘... 【目的】逐步采用机械化直播的集约栽培模式已是水稻种植的主要趋势之一,本文研究了机械化水稻直播条件下,缓释尿素与普通尿素不同配施对水稻生长和产量的影响特征,为直播稻的高产高效施肥技术提供理论基础。【方法】以优质杂交籼稻‘宜香优2115’为材料,采用两因素裂区设计,主区为直播(CD)和手插(CH)两种栽植模式;副区为氮肥运筹方式,总施氮量150kg/hm^2,设普通尿素按基、蘖、穗肥配比为3∶3∶4(U30),普通尿素全量基施(U100),缓释尿素全量基施(S100)及缓释尿素(基肥)与普通尿素(穗肥)配比为8∶2(S80)、6∶4(S60)、4∶6(S40)6种肥料运筹方式,以不施氮肥(CK)为对照。调查了直播水稻的叶色变化、叶片生长、LAI、剑叶光合作用和叶片碳氮代谢及其与产量形成的关系。【结果】1)直播稻产量略高于手插稻,前者有效穗数优势显著,后者优势在于每穗粒数,其产量差异表现取决于有效穗数和每穗粒数是否充分互补,具体体现在总颖花数,优势肥料处理在有效穗数和每穗粒数上协调性更强。2)缓释尿素与普通尿素的配施处理下叶色表现,直播稻和手插稻分别呈“三黑三黄”和“二黑二黄”交替变化,直播稻的叶色相对较浅;直播稻叶片生长前快后慢,齐穗期LAI及齐穗后剑叶Pn值均较低,但其高效叶面积率较高,表明后期群体上部功能叶叶面积较大,群体质量较高;在叶片碳氮代谢上,直播稻中、后期叶片NSC/N较高,碳代谢较旺盛,两种栽植方式中高产处理后期叶片NSC/N相近,均较低,但代谢途径不同,直播稻主要通过增强氮代谢,手插稻则通过增强碳水化合物的同化进行调节。增加穗肥占比可有效优化直播稻中后期的叶片质量。3)相关分析显示,孕穗期的叶片增长率、LAI、剑叶Pn及叶片NSC/N均与产量呈显著或极显著相关,表明孕穗期的叶片质量是影响产量形成的关键。4)直播稻和� 展开更多

BACKGROUND Unconjugated bilirubin (UCB) is generally considered toxic but has gained recent prominence for its anti-inflammatory properties. However, the effects of it on the interaction between intestinal flora and organisms and how it influences immune responses remain unresolved. AIM To investigate the role of UCB in intestinal barrier function and immune inflammation in mice with dextran-sulfate-sodium-induced colitis. METHODS Acute colitis was induced by 3%(w/v) dextran sulfate sodium salt in drinking water for 6 d followed by untreated water for 2 d. Concurrently, mice with colitis were administered 0.2 mL UCB (400 μmol/L) by intra-gastric gavage for 7 d. Disease activity index (DAI) was monitored daily. Mice were sacrificed at the end of the experiment. The length of the colon and weight of the spleen were recorded. Serum level of D-lactate, intestinal digestive proteases activity, and changes to the gut flora were analyzed. In addition, colonic specimens were analyzed by histology and for expression of inflammatory markers and proteins.RESULTS Mice treated with UCB had significantly relieved severity of colitis, including lower DAI, longer colon length, and lower spleen weight (colon length: 4.92 ± 0.09 cm vs 3.9 ± 0.15 cm;spleen weight: 0.33 ± 0.04 vs 0.74 ± 0.04, P < 0.001). UCB administration inactivated digestive proteases (chymotrypsin: 18.70 ± 0.69 U/g vs 44.81 ± 8.60 U/g;trypsin: 1.52 ± 0.23 U/g vs 9.05 ± 1.77 U/g, P < 0.01), increased expression of tight junction (0.99 ± 0.05 vs 0.57 ± 0.03, P < 0.001), decreased serum level of D-lactate (31.76 ± 3.37 μmol/L vs 54.25 ± 1.45 μmol/L, P < 0.001), and lowered histopathological score (4 ± 0.57 vs 7 ± 0.57, P < 0.001) and activity of myeloperoxidase (46.79 ± 2.57 U/g vs 110.32 ± 19.19 U/g, P < 0.001). Contact inhibition adjusts organ size to the proper size and ensures the cultured cells growing to a monolayer.By regulating the downstream coordinator YAP,the evolutionarily conserved Hippo transduction pathway attunes cell growth and death in response to cell contact inhibition,polarity,self-renewal,and differentiation.Dysregulation of this pathway is involved in various diseases such as cancer.RNA-binding protein QKI regulates cell proliferation,metabolism,division,and immunity in various cancer models,but its role in cancer cell contact inhibition remains unclear.In this study,we aimed to clarify the relationship between QKI and YAP,and the role of their interaction in cell contact inhibition.We found a lower QKI expression level in sparse condition,whereas a higher expression level in confluent condition by western blot analysis and immunofluorescence assay.QKI knockdown elevated cell proliferation and invasion both in vitro and in vivo.Strikingly,the results of CCK-8 assay,colony formation assay,and transwell assay showed that the phenomenon was in accord with the expression level of pYAP and reverse with YAP.Higher levels of Wnt3a and β-catenin were also found in xenografts of QKI-knockdown clear cell renal cell carcinoma (ccRCC) CAKI-1 cells by western blot analysis and immumohistochemical staining.Finally,a positive correlation between QKI and pYAP was found in clinical specimens by immunohistochemistry.Thus,as a negative regulator of YAP,QKI attuned the cell contact inhibition,leading to inhibition of cancer cell proliferation and invasion through Wnt and GPCR pathway. 展开更多

This work aimed to investigate the activity of loureirin A against Candida albicans biofilms. 2, 3-Bis-(2-methoxy-4-nitro-5-sulfophenyl)-2 H-tetrazolium-5-carboxanilide(XTT) reduction assay and scanning electron microscopy were used to investigate the anti-biofilm effect.Minimal inhibitory concentration testing and time-kill curve assay were used to evaluate fungicidal activity. Cell surface hydrophobicity(CSH) assay and hyphal formation experiment were respectively carried out to investigate adhesion and morphological transition,two virulence traits of C. albicans. Real-time RT-PCR was used to investigate gene expression. Galleria mellonella-C. albicans and Caenorhabditis elegans-C. albicans infection models were used to evaluate the in-vivo antifungal effect. Human umbilical vein endothelial cells and C. elegans nematodes were used to evaluate the toxicity of loureirin A. Our data indicated that loureirin A had a significant effect on inhibiting C. albicans biofilms, decreasing CSH, and suppressing hyphal formation. Consistently, loureirin A down-regulated the expression of some adhesion-related genes and hypha/biofilm-related genes. Moreover, loureirin A prolonged the survival of Galleria mellonella and Caenorhabditis elegans in C. albicans infection models and exhibited low toxicity. Collectively,loureirin A inhibits fungal biofilms, and this effect may be associated with the suppression of pathogenic traits, adhesion and hyphal formation. 展开更多

Objective Identification of new risk factors is needed to improve prediction of adverse outcomes in patients with three-vessel disease(TVD). The present study aimed to evaluate the prognostic values of serum chloride and sodium levels in patients with TVD. Methods We used data from a prospective cohort of consecutive patients with angiographically confirmed TVD. The primary endpoint was all-cause death. Cox proportional hazard regression was used to analyze the relationship of serum chloride and sodium levels with long-term outcomes of TVD patients. Results A total of 8,318 participants with available serum chloride and sodium data were included in this analysis. At baseline, patients in the low tertiles group of serum chloride level(≤ 102.0 mmol/L) or serum sodium level(≤ 139.0 mmol/L) had more severe disease conditions. During a median follow-up of 7.5-year, both low serum chloride level and low serum sodium level were found to be associated with an increased risk for mortality in univariate analysis. However, when both parameters were incorporated into a multivariate model, only low serum sodium level remained to be an independent predictor of all-cause death(hazard ratio: 1.16, 95% confidence interval: 1.01-1.34, P = 0.041). Modest but significant improvement of discrimination was observed after incorporating serum sodium level into the Synergy between percutaneous coronary intervention(PCI) with Taxus and Cardiac Surgery score. Conclusion Serum sodium level is more strongly associated with long-term outcomes of TVD patients compared with serum chloride level. Low serum sodium level is an independent risk factor for mortality, but only provides modest prognostic information beyond an established risk model. 展开更多

Objective The aim of this study was to determine whether low calf circumference(CC) could predict nutritional risk and the cutoff values of CC for predicting nutritional risk in hospitalized patients aged ≥ 80 years.Methods A total of 1,234 consecutive patients aged ≥ 80 years were enrolled in this study.On admission, demographic data, CC, and laboratory parameters were obtained.Patients with Nutritional Risk Screening 2002(NRS-2002) total score ≥ 3 were considered as having nutritional risk.Results CC values were significantly lower in patients with nutritional risk compared to those in patients without nutritional risk [27.00(24.50-31.00) vs.31.00(29.00-33.50], P < 0.001].CC was negatively correlated with age and nutritional risk scores.Logistic regression analysis of nutritional risk revealed that body mass index, albumin level, hemoglobin level, cerebral infarction, neoplasms, and CC(OR, 0.897;95% confidence interval, 0.856-0.941;P < 0.001) were independent impact factors of nutritional risk.Nutritional risk scores increased with a decrease in CC.In men, the best CC cutoff value for predicting nutritional risk according to the NRS-2002 was 29.75 cm.In women, the cutoff value was 28.25 cm.Conclusion CC is a simple, noninvasive, and valid anthropometric measure to predict nutritional risk for hospitalized patients aged ≥ 80 years. 展开更多

Objective: To evaluate the effects of Celastrus Orbiculatus extracts(COE) on metastasis in hypoxiainduced hepatocellular carcinoma cells(Hep G2) and to explore the underlying molecular mechanisms. Methods:The effect of COE(160, 200 and 240 μg/mL) on cell viability, scratch-wound, invasion and migration were studied by 3-4,5-dimethyl-2-thiazolyl-2,5-diphenyl-2-H-tetrazolium bromide(MTT), scratch-wound and transwell assays, respectively. Co Cl2 was used to establish a hypoxia model in vitro. Effects of COE on the expressions of E-cadherin, vimentin and N-cadherin were investigated with Western blot and immuno?uorescence analysis,respectively. Results: COE inhibited proliferation and metastasis of hypoxia-induced hepatocellular carcinoma cells in a dose-dependent manner(P<0.01). Furthermore, the expression of epithelial-mesenchymal transition(EMT) related markers were also remarkably suppressed in a dose-dependent manner(P<0.01). In addition, the upstream signaling pathways, including the hypoxia-inducible factor 1α(Hif-1α) and Twist1 were suppressed by COE. Additionally, the Hif-1α inhibitor 3-5’-hydroxymethyl-2’-furyl)-1-benzylindazole(YC-1), potently suppressed cell invasion and migration as well as expression of EMT in hypoxia-induced Hep G2 cells. Similarly, the combined treatment with COE and YC-1 showed a synergistic effect(P<0.01) compared with the treatment with COE or YC-1 alone in hypoxia-induced Hep G2 cells. Conclusions: COE signi?cantly inhibited the tumor metastasis and EMT by suppressing Hif-1α/Twist1 signaling pathway in hypoxia-induced Hep G2 cell. Thus, COE might have potential effect to inhibit the progression of Hep G2 in the context of tumor hypoxia. 展开更多
