败酱总黄酮提取纯化工艺优化及纯化前后体外抗肝肿瘤药效对比研究

The process optimization of extraction and purification for total flavonoids and comparative study on the efficacy of anti-liver tumors in vitro before and after purification

摘要 目的优选败酱总黄酮的提取纯化工艺,并结合体外药效实验,考察纯化前后的活性,为败酱合理开发和利用提供依据。方法采用正交试验设计法,以总黄酮提取量为指标,优选提取工艺;采用大孔吸附树脂法,以总黄酮含量为考察指标,优选纯化工艺;采用MTT法比较总黄酮纯化前后体外肝癌细胞抑制作用。结果败酱总黄酮最佳提取工艺为15倍量60%乙醇回流提取,提取3次,每次1h。最佳纯化工艺为HPD-300树脂,上样浓度0.1 g饮片/mL,0.9 g/mL(饮片/湿树脂)比例、6倍水除杂,4倍70%乙醇洗脱,收集洗脱液水浴蒸干,即得败酱总黄酮富集物;纯化后总黄酮体外抗肝肿瘤IC50值比纯化前降低81.16%。结论该工艺方法先进合理,适合工业生产,具有简便、快速、准确等优点,为败酱的进一步研究及开发奠定基础。 Objective Optimization the extraction and purification process for total flavonoids from Patrinia scabiosaefolia Fisch.,study the pharmacodynamic experiment in vitro,investigate the activity before and after purification,provide rational development and utilization.Methods The best extraction process of total flavonoids was carried out with the orthogonal experiment,the best purification process of total flavonoids was carried out with the pore adsorption resin method,comparison of inhibitory effects of total flavonoids on hepatoma cells in vitro with the method of MTT.Results The best extracting technology for Patrina villosa Juss was determined with 60%ethanol,15 times,refluxing extraction 3 times and every time by 1 hour.The optimal purification technology was determined in the condition of the concentration 0.1 g/mL of the Patrinia extract,using HPD-300 macro porous resin,with the ration of 0.75 g/mL(content of medicinal materials against resin)and sample impurity with water,6 times and elution with 70%ethanol4 times.Collect the eluent water bath and evaporated,obtain the total flavonoids from Valerianaceae,the total flavonoids had an IC50 value of 81.6%lower than purification.Conclusion This technology and method was useful and suitable for industrial production,it was simple,rapid and accurate,it established foundation for further research and development for sauce herbal medicine.
