目的:探讨伸筋草总生物碱的提取纯化方法,并对其急性毒性及抗炎活性进行研究。方法:采用阳离子交换树脂法提取纯化伸筋草药材中的总生物碱成分,以苦参碱为对照品,采用酸性染料比色法,对提取物中的总生物碱含量进行测定。对伸筋草总生物碱提取物进行急性毒性试验,以弗氏完全佐剂致大鼠足跖肿胀试验和大鼠棉球肉芽肿增生试验,对伸筋草总生物碱的抗炎活性进行研究。结果:500 g伸筋草药材共提取纯化制得伸筋草总生物碱提取物92 g,提取率为18.4%。伸筋草总生物碱含量测定方法的精密度(RSD=0.86%)、重复性(RSD值为1.71%)、稳定性(RSD=0.92%)、加样回收率(平均值=99.22%,RSD=1.34%)试验结果显示,均在误差范围内,伸筋草提取物中所含总生物碱的质量分数为30.47μg/mg。伸筋草总生物碱提取物的LD50为11.62 g/kg。伸筋草高、中、低剂量组均能够改善佐剂型关节炎(AA)大鼠的足跖肿胀并降低大鼠棉球肉芽肿的增生情况,且呈现剂量依赖性,其中伸筋草高、中剂量组与模型组相比,改善效果显著(P<0.05)。结论:伸筋草总生物碱提取物未见明显毒性,为伸筋草药材的抗炎活性成分,具有较好的抗炎作用。
Objective:To investigate the extraction and purification method of total alkaloids from Lycopodii Herba,and to study its acute toxicity and anti-inflammatory activity.Methods:The total alkaloids in Lycopodii Herba were extracted and purified by cation exchange resin method.The total alkaloids in the extract were determined by acid dye colorimetry with matrine as the reference substance.The acute toxicity test of the total alkaloid was carried out,and the anti-inflammatory activity of the total alkaloids was studied by the toe swelling test and cotton ball granuloma test.Results:Total alkaloid extracted from 500 g Lycopodii Herba amounted to 92 g,and the extraction rate was 18.4%.The extraction indexes of the total alkaloid from Lycopodii Herba were all within the error range[accuracy(RSD=0.86%),repeatability(RSD=1.71%),stability(RSD=0.92%),recovery rate(mean=99.22%,RSD=1.34%)],and the mass fraction of total alkaloids in the extract was 30.47μg/mg.The LD50 of total alkaloid extract was 11.62 g/kg.The high-dose,medium-dose and low-dose groups were all able to improve the plantar swelling of the rats with adjuvant arthritis(AA)and reduce the hyperplasia of cotton ball granuloma in a dose-dependent way,among which the high-dose and medium-dose groups had a significant improvement compared to the model group(P<0.05).Conclusion:The total alkaloid extract from Lycopodii Herba has no obvious toxicity,which is the anti-inflammatory active component of Lycopodii Herba with significant effect.
