依据COI基因及形态特征鉴定蛀害红松和樟子松的梢斑螟

Identification of Dioryctria species Boring Pinus koraiensis and P.sylvestris var.mongolica with COI Gene Sequences and Morphological Characteristics

摘要 为确定危害黑龙江地区红松(Pinus koraiensis)、樟子松(P.sylvestris var.mongolica)树干及侧枝的梢斑螟属(Dioryctria)昆虫种类,通过比对幼虫毛序、成虫外部形态及外生殖器特征对其进行鉴定。同时,利用分子生物学技术辅助鉴定,依据线粒体COI基因计算样本与其他梢斑螟种类间的遗传距离,分别构建ML树和NJ树,以确定其系统发育关系。结果显示:样本的形态特征与赤松梢斑螟(D.sylvestrella)相同;样本的COI基因序列相似度较高,且种群间存在共享单倍型(Hap3);样本与在GenBank记载的赤松梢斑螟间的遗传距离(0.004)处于种内遗传距离范围(0~0.005),且在系统发育树中与赤松梢斑螟聚为一支。因此,可以确定所鉴定的样本均为赤松梢斑螟。依据COI基因序列的分析结果,能够有效确定物种及分类地位,并且与传统划分结果相吻合,说明COI基因适用于梢斑螟属昆虫的鉴定和系统发育的研究。 In order to determine the species of Dioryctria,which is boring the trunk and lateral branches of Pinus koraiensis and P.sylvestris var.mongolica in Heilongjiang,the specimens were identified by comparing the larval chaetotary,the outer morphology of adult and the characteristics of external genitalia.At the same time,molecular biology technology was used to assist identification.With mitochondrial COI gene,the genetic distance between the specimens and other species of Dioryctria was calculated,and ML tree and NJ tree were constructed to determine their phylogenetic relationship,respectively.The morphological characteristics of specimens were the same as that of D.sylvestrella.The sequence similarity of COI gene in the specimens was very high,and haplotype(Hap3)was found in different populations.The genetic distance(0.004)between the specimens and D.sylvestrella recorded in GenBank was within the range of intraspecific genetic distance(0-0.005).And the specimens were clustered as one branch in phylogenetic tree,together with D.sylvestrella.Therefore,it can be determined that the identified specimens are D.sylvestrella.From COI gene sequence,the taxonomic status of species can be effectively determined,and it is consistent with the traditional classification of species.These results indicated that COI gene was suitable for the identification and phylogenetic study of Dioryctria species.
