两种佐剂在H9N2亚型禽流感灭活疫苗中的效果比较 认领

摘要 为提高H9N2亚型禽流感灭活疫苗的免疫效力,拟从Carbigen和Marcol-52两种佐剂中选择一种适合的疫苗佐剂。利用两种佐剂分别制备H9N2亚型禽流感灭活疫苗,通过SPF鸡免疫后7、14、21、24 d的抗体水平、黏膜系统SIgA含量及攻毒保护率三个方面,对Carbigen和Marcol-52佐剂进行探讨。结果显示,Carbigen佐剂和Marcol-52佐剂疫苗均可以刺激机体产生呼吸道黏膜SIgA和血清抗体。在免疫后第7天至第21天,Carbigen佐剂疫苗组血清抗体水平均高于Marcol-52佐剂疫苗组;Carbigen佐剂疫苗组SIgA产生早于Marcol-52,在第14天Carbigen佐剂疫苗组SIgA含量明显高于Marcol-52佐剂疫苗组(P<0.05),产生SIgA主要部位位于气管叉和肺脏;Carbigen佐剂疫苗组攻毒后的保护率高于Marcol-52佐剂疫苗组。Carbigen佐剂配置的疫苗可刺激接种动物快速产生免疫应答反应,其效果优于Marcol-52佐剂,有望成为H9N2灭活疫苗的候选佐剂。 To improve the immune efficacy of inactivated H9N2 avian influenza vaccine,a suitable adjuvant of vaccine was selected from Carbigen and Marcol-52.First of all,two kinds of inactivated H9N2 avian influenza vaccines were prepared with Carbigen and Marcol-52.The effects after immunization by SPF birds were discussed from three aspects:antibody levels in day 7,day 14,day 21 and day 24 after immunization,SIgA contents in mucosal system and challenge protection test.The results showed that both vaccines with Carbigen and Marcol-52 could stimulate the production of SIgA in respiratory mucosa and serum antibodies.From 7 to 21 days after immunization,the serum antibody level of Carbigen group vaccine was higher than that of Marcol-52 group vaccine;The SIgA production of Carbigen group vaccine was earlier than that of Marcol-52 group vaccine and significantly higher than that of Marcol-52 group vaccine on 14 days(P<0.05),the main part of arisen SIgA was tracheal fork and lung;the protection rate of Carbigen group vaccine was higher after challenge.The vaccine with Carbigen can stimulate the immune response quickly after animal vaccinated and the effect is better than with Marcol-52.It is expected to be a candidate adjuvant for H9N2 inactivated vaccine.
