金钻蔓绿绒的离体培养研究 被引量:6

摘要 本研究以金钻蔓绿绒根茎、叶片、叶基、叶柄为试验材料,研究外植体类型与取材时间、消毒处理方法,植物激素种类与浓度筛选出适宜诱导愈伤组织、不定芽与继代培养基。结果表明:叶片和叶基最佳灭菌时间:75%酒精30 s+0.1%升汞10 min,叶柄:75%酒精30 s+0.1%升汞16 min,根茎:75%酒精30 s+0.1%升汞20 min;最佳取材时间为夏季,愈伤诱导率高,且诱导时间短;叶片、叶基、叶柄最佳诱导愈伤培养基:MS+2.0 mg/L TDZ+0.2 mg/L 2,4-D,增殖培养基:MS+3.0 mg/L 6-BA+0.2 mg/L NAA;根茎诱导不定芽及继代增殖最佳培养基分别为:MS+1 mg/L 6-BA+0.3 mg/L NAA+1 mg/L GA3与MS+0.8 mg/L 6-BA+0.2 mg/LIBA。 In this research, the rhizome, leaf, leaf base and leaf petiolas of Philodendron ‘con-go'were used as experimental materials to investigate the sampling time, disinfection method explant types, the types and concentration of plant hormone and filtering the suitable medium for callus induction, adventitious buds and subculture. The result showed the optimum disinfecting time of leaf and leaf base: 75% alcohol 30 s +0.1% Hg Cl210min; petiole: 75% alcohol 30 s +0.1% Hg Cl216 min, rhizome: 75% alcohol 30s+0.1% Hg Cl220 min; Summer was the best sampling time, it had the high rate of callus induction and the short induction time. The best callus induction and proliferation medium of leaves, leaf base and petiole respectively: MS +2.0 mg/ LTDZ +0.2 mg/L2,4-D; MS+3.0 mg/L6-BA+0.2 mg/LNAA; The rhizome induced adventitious buds and the best subculture multiplication medium respectively: MS +1 mg/L6-BA +0.3 mg/L NAA +1 mg/L GA3; MS +0.8 mg/L 6-BA +0.2 mg/L IBA.
