摘要 目的:建立高效液相色谱法同时测定护肝片中绿原酸、牡荆苷、五味子醇甲、五味子甲素和五味子乙素的含量。方法:使用ZORBAXSB-C18柱(4.6mm×150mm,5μm),柱温35℃,流动相为甲醇-0.2%磷酸水溶液梯度洗脱,流速1mL·min-1,检测波长0~20min为340nm,20~40min为254nm,进样量20μL。结果:绿原酸、牡荆苷、五味子醇甲、五味子甲素和五味子乙素在各自浓度范围内线性关系良好,r≥0.9999,平均加样回收率在86.46%~101.21%,RSD为1.51%~2.90%(n=6)。对3个批次护肝片样品进行了含量测定,其中五味子醇甲含量符合药典标准。结论:该方法简便准确、稳定可靠,可用于护肝片有效成分的含量测定。 Objective: To establish a high performance liquid chromatography (HPLC) method for simultaneously determining the contents of chlorogenic acid, vitexin, schisandrol A, schisandrin A and schisandrin B in Hugan tablets. Methods: ZORBAX SB-C18 column (4.6 mm × 150 mm, 5 μm) was used and the column temperature was 35 ℃. The mobile phase consisted of methanol and 0.2% phosphoric acid solution which were linear gradient eluting at the flow rate of 1 mL·min-1. The detection wavelengths were 340 nm from 0 to 20 minutes and 254 nm from 20 to 40 minutes. The sample size was 20 μL. Results: Chlorogenic acid, vitexin, schisandrol A, schisandrin A and schisandrin B had a good linear relation (r ≥ 0.999 9) in their respective concentration ranges. The average recoveries were in the range of 86.46%– 101.21% and the RSD were in the range of 1.51%– 2.90%(n = 6). The contents of three batches of Hugan tablets were determined, in which the content of schisandrol A was in accordance with the pharmacopoeia standard. Conclusion: The method was simple, accurate, stable and reliable, which could be used for the determination of effective components in Hugan tablets.
