白扁豆提取物对幽门螺杆菌损伤人胃黏膜上皮细胞的修复作用 预览

Repairing Effects of Dolichos Bean Seed Extract on Helicobacter pylori-Injured Human Gastric Epithelial Cells

摘要 对数种中药提取物进行筛选后发现,白扁豆提取物对人胃黏膜上皮(human gastric epithelial cell line,GES-1)细胞增殖具有良好的促进作用,在500.00 μg/mL浓度作用下GES-1细胞存活率可提高到125.80%。同时,白扁豆提取物可明显抑制幽门螺杆菌(Helicobacter pylori,Hp)生长。以15.62μg/mL白扁豆提取物作用24h后,对Hp抑制率达62.61%。白扁豆提取物对脲酶活性也有明显的抑制作用,其IC50值为397.14μg/mL。以Hp侵染GES-1细胞24 h建立GES-1细胞损伤模型,经500μg/mL白扁豆提取物作用12h后,GES-1细胞存活率可达到95%以上。因此,白扁豆可能通过促进GES-1细胞增殖的同时抑制Hp及脲酶活性,从而对Hp损伤GES-1细胞起到修复的作用。 Effects of several traditional Chinese herbs extracts on human gastric epithelial cells (GES-1) were analyzed. The Dolichos bean seed (DBS) extract demonstrated promotive effects on the proliferation of GES-1 cells. The survival rate of GES-1 cells reached 125.80 % at a concentration of 500.00 μg/mL. In the meantime, DBS extract could significantly inhibit Helicobacter pylori (Hp) growth. The inhibition rate of Hp reached 62.61 % after treatment with 15.62 μg/mL DBS extract for 24 h. Moreover, DBS extract showed significant inhibitory effect on urease activity, with IC50 value of 397.14 μg/mL. The GES-1 cell injury model was established by infecting GES-1 cells with Hp for 24 h. The survival rate of GES-1 cells was over 95 % after treatment with 500 μg/mL DBS extract for 12 h. Therefore, DBS extract might repair Hp-injured GES-1 cells by promoting the proliferation of GES-1 cells and inhibiting Hp and urease activity.
