白藜芦醇通过激活腺苷酸活化蛋白激酶信号途径在体外对大鼠炎症性肠病改善作用的实验研究

Experimental Study on Improvement Effects of Resveratrol in Inflammatory Bowel Disease in Rats by Activating AMPK Signaling Pathway

摘要 目的:研究白藜芦醇通过腺苷酸活化蛋白激酶(AMPK)信号途径在体外对大鼠炎症性肠病的改善作用。方法:选取48只健康SD大鼠将其随机分为对照组(n=12)、模型组(n=12)、美沙拉嗪组(n=12)和白藜芦醇组(n=12);除对照组大鼠外,均采用三硝基苯磺酸(TNBS)灌胃以建模,而对照组大鼠采用同等体积的乙醇溶液灌胃;建模成功后,美沙拉嗪组大鼠给予美沙拉嗪2 g/(kg·d)灌胃,白藜芦醇组大鼠给予白藜芦醇100 mg/(kg·d)灌胃,而正常组和模型组大鼠给予蒸馏水100 mg/(kg·d)灌胃,喂养期间观察各组大鼠的体质量、大便性状和隐血情况,并比较各组大鼠按疾病活动度(DAI)量表评分差异,以及各组大鼠喂养7 d后采集其空腹静脉血采用ELISA法检测大鼠血清中PGE2,NO,IL-1β,TNF-α水平,同时处死大鼠取其结肠观察其形态,并采用Western Blotting法检测大鼠肠黏膜中SIRT1、AMPK和NF-кB p65蛋白水平值的变化。结果:模型组、美沙拉嗪组和白藜芦醇组大鼠的DAI评分值均高于对照组(P<0.05);而美沙拉嗪组和白藜芦醇组大鼠的DAI评分值明显低于模型组(P<0.05);白藜芦醇组大鼠的DAI评分值低于美沙拉嗪组(P<0.05);模型组、美沙拉嗪组和白藜芦醇组大鼠的PGE2,NO,IL-1Β,TNF-α水平测得值高于对照组(P<0.05);而美沙拉嗪组和白藜芦醇组大鼠的PGE2,NO,IL-1Β,TNF-α水平测得值均明显低于模型组(P<0.05);白藜芦醇组大鼠的PGE2,NO,IL-1Β,TNF-α水平测得值均低于美沙拉嗪组(P<0.05);模型组、美沙拉嗪组和白藜芦醇组大鼠的肠黏膜中SIRT1,AMPK低于对照组(P<0.05),而NF-кB p65蛋白高于对照组(P<0.05);美沙拉嗪组和白藜芦醇组大鼠的SIRT1、AMPK明显高于模型组(P<0.05),而NF-кB p65蛋白低于模型组(P<0.05);白藜芦醇组大鼠的SIRT1、AMPK明显高于美沙拉嗪组,而NF-кB p65蛋白低于美沙拉嗪组(P<0.05)。结论:白藜芦醇对炎症性肠病具有改善作用,有效降低炎症因子的表达;其机制 Objective: To study the improvement effects of Resveratrol in inflam-matory bowel disease in rats by activating AMPK signaling pathway in vitro. Methods: 48 healthy SD rats were randomly divided into control group(n=12), model group(n=12), mesalazine group(n=12) and resveratrol group(n=12). In addition to the control group, all rats were treated with TNBS for modeling, while the control group was treated with ethanol solution of the same volume. After the success of the modeling, the salad oxazine group rats are given the salad oxazine 2 g/(kg·d) to fill the stomach, resveratrol group rats given 100 mg/(kg · d) to fill the stomach, the normal group and model group rats given distilled water 100 mg/(kg · d) to fill the stomach;observing between groups of rats during the period of feeding body quality,traits and stool occult blood;based on the disease activity and comparing each group rats(DAI) rating scale differences;and in rats of groups its fasting venous blood was taken from the 7 d by ELISA method to detect the rat serum PGE2, NO, IL-1β. TNF-α levels were detected, and the colons of rats were sacrificed and the changes of SIRT1, AMPK and NF-кB p65 protein levels in the intestinal mucosa were detected by Western Blotting. Results: DAI scores of the model group, mesalazine group and resveratrol group were all higher than those of the control group(P<0.05). DAI scores of mesalazine group and resveratrol group were significantly lower than those of model group(P<0.05). DAI score of the resveratrol group was lower than that of the mesalazine group(P<0.05). Model group, the salad lamictal and resveratrol group rats PGE2, NO, IL-1β, TNF-α level measured value is higher than the control group(P<0.05). And the salad lamictal and resveratrol group rats PGE2, NO,IL-1β, TNF-α level measured values were significantly lower than those of model group(P<0.05). Resveratrol group of PGE2 in rats, NO, IL-1β, TNF-α level measured values are lower than the salad oxazine group(P<0.05). The SIRT1 and AMPK levels in the
