浸种方法对丝瓜种子萌发的影响 预览

摘要 【目的】探究不同浸种方法对丝瓜种子萌发的影响,得到种子发芽率高、出芽一致性好的丝瓜种子浸种方法。【方法】以丝瓜品种‘圆帅’为试验材料,设置纯净水浸泡(T1)、人工破壳后200mg/LGA3浸泡(T2)、人工破壳后纯净水浸泡(T3)、200mg/LGA3浸泡(T4)4种浸种方法,28℃浸种12h后30℃恒温催芽,测定各处理丝瓜种子的发芽势、发芽率、壮苗指数、酶活性、内源激素含量等指标,并观察各处理丝瓜种子萌发第2天胚轴横切面的解剖结构。【结果】各处理丝瓜种子发芽势与发芽率高低顺序均为T2>T3>T4>T1,T2比T1发芽势提高了38%,发芽率提高了59%,壮苗指数增加了0.007。与其他处理相比,T2处理在种子萌发初期会提升过氧化物酶(POD)、过氧化氢酶(CAT)和超氧化物歧化酶(SOD)活性;增加生长素(IAA)、玉米素(TZ)含量;降低脱落酸(ABA)含量;茉莉酸(JA)、水杨酸(SA)含量在种子萌发时较低,随后其含量先上升后下降,其主要在种子萌发第3天后开始发挥调控作用。从丝瓜种子胚轴横切面可以看出,T2处理维管束横切面面积最大,表明该处理可促进胚轴维管束细胞的发育,使维管束更加发达。【结论】在丝瓜种子育苗过程中,破壳并采用200mg/LGA3浸泡是获得种子发芽率高、出芽一致性好的浸种方法。 【Objective】 This study explored the effects of different soaking methods on germination of loofah seeds to obtain a soaking method with high germination rate and good sprouting consistency.【Method】 The loofah variety ‘Yuanshuai’ was used,and four soaking treatments of pure water soaking (T1),200 mg/L GA 3 soaking (T2) after artificial shelling,and pure water soaking (T3) after artificial shell breaking,and 200 mg/L GA 3 immersion (T4) were setup.After 12 h of seeds soaking at 28 ℃,germination was carried out at constant temperature of 30 ℃.The germination potential,germination rate,seedling index,enzyme activity,and endogenous hormone content were measured,and the anatomical structure of the cross section of the hypocotyl on the 2nd day of each treatment was observed.【Result】 The germination potential and germination rate of loofah seeds were in the order of T2>T3>T4>T1.Compared to T1,T2 increased the germination potential by 38%, increased the germination rate by 59%, and increased the seedling index by 0.007.Compared with other treatments, T2 treatment increased peroxidase (POD), catalase (CAT) and superoxide dismutase (SOD) activities in the early stage of seed germination, increased auxin (IAA) and zeatin (content of TZ),and decreased the content of abscisic acid (ABA). Contents of jasmonic acid (JA) and salicylic acid (SA) were low in seed germination, then the contents increased first before decreasing,and they played a regulatory role after the third day of seed germination.As seen from the cross section of the lobe seed hypocotyls,T2 treatment had the largest cross-sectional area,indicating that the treatment promoted the development of hypocotyl vascular column cells and made the vascular column more developed.【Conclusion】 In the process of seedling cultivation of loofah,the loofah seedlings with high germination rate and good uniformity of sprouting can be obtained by breaking the shell and soaking with 200 mg/L GA 3.
