川崎病并发冠脉损伤患儿血清PAI-1水平对血管内皮细胞活力的影响 预览 被引量:1

Effects on Cell Viability of HUVECs Stimulated by Serum From Childrenwith Coronary Artery Lesions of Kawasaki Disease via Regulating PAI-1

摘要 目的:探讨川崎病(KD)并发冠脉损伤(CALs)患儿血清纤溶酶原激活物抑制物1(PAI-1)水平对血管内皮细胞活力的影响。方法:收集川崎病患儿52例,其中冠脉损伤组患儿25例,非冠脉损伤组患儿27例,另收集健康对照组儿童21例,收集临床资料检测KD-CALs高危及出凝血指标,制备血清标本。RT-qPCR和ELISA法检测血清PAI-1水平。各组血清分别处理人脐静脉内皮细胞(HUVECs) 24h后,RT-qPCR法检测HUVECs中PAI-1表达,CCK-8法检测细胞活力。结果:川崎病并发冠脉损伤组患儿血小板(Platelet)、白细胞(WBC)、红细胞沉降率(ESR)以及凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、纤维蛋白原(FIB)和纤维蛋白原降解产物(FDP)水平明显高于对照组。川崎病并发冠脉损伤组Platelet、ESR、PT、APTT和FDP水平较非冠脉损伤组明显增高(P〈0.05),但FIB水平明显降低,提示KD并发CALs时患儿体内出现明显凝血功能障碍。RT-q PCR和ELISA检测结果显示,与对照组比较,川崎病并发冠脉损伤组和非冠脉损伤组血清PAI-1 mRNA和蛋白水平显著升高,其中并发冠脉损伤组水平增高最显著。细胞实验结果显示,与对照组比较,川崎病并发冠脉损伤组血清处理后,HUVECs中PAI-1表达明显升高,然而HUVECs活力却显著降低。结论:川崎病并发冠脉损伤患儿机体处于严重高凝状态,其患儿血清高水平PAI-1可诱导HUVECs中PAI-1水平明显升高,从而抑制HUVECs活力。 Objective: To investigate the effect and mechanism of serum from children with coronary artery lesions of Kawasaki disease on the viability of human umbilical vein endothelial cells (HUVECs) by regulating PAI-1. Methods: Fifty-two children with Kawasaki disease were collected, including 25 children withcoronary artery injury, 27 children with non - coronary artery injury, and 21 children with healthy controlgroup. Clinical data were collected to detect the high risk of KD-CALs and coagulation index, and serum samples were prepared. Human umbilical vein endothelial cells (HUVECs) were treated with serum for 24 hours.The expression of PAI-1 in HUVECs was detected by RT-qPCR and the cell viability was detected by CCK-8. Results: Platelet, white blood cell (WBC), erythrocyte sedimentation rate ( ESR), prothrombin time(PT), activated partial thromboplastin time (APTT), fibrinogen (FIB) and fibrinogen degradation product (FDP) levels in children with Kawasaki disease complicated with coronary artery injury were significantlyhigher than those in the control group. Conclusion: Children with KD-CALs have significantly elevated levelsof serum PAI-1 in a massive hypercoagulable state, and increased PAI-1 levels in HUVECs leading to the inhibition of viability of HUVECs.
