酪氨酸蛋白激酶受体EphA2在人脑星形细胞瘤中的表达及其与肿瘤血管生成的关系研究

Research on the clinical expressions of tyrosine kinase receptor EphA2 in the human brain astrocytoma and its correlation with the genesis of tumor vessels

摘要 目的:研究人脑星形细胞瘤中酪氨酸蛋白激酶受体Eph A2的表达水平,分析Eph A2表达与肿瘤血管生成的关系。方法:临床纳入44例我院2014年9月至2017年9月期间收治的人脑星形细胞瘤患者作为研究组,另选取同期因脑出血需要脑叶切除的6例患者作为对照组。应用Western Blot及免疫组化法检测不同级别肿瘤组织中Eph A2表达水平及微血管密度(MVD)差异。结果:Eph A2蛋白主要表达于肿瘤细胞的胞浆和胞膜,在星形细胞瘤中仅4例低级别星形细胞瘤未检测到Eph A2蛋白表达,其余星形细胞瘤中均检测到Eph A2蛋白表达,阳性表达率高达90.91%。正常脑组织中均未检测到Eph A2蛋白表达。两组患者的Eph A2蛋白表达有统计学差异(P〈0.05)。低级别(Ⅰ-Ⅱ级)和高级别(Ⅲ-Ⅳ级)人脑星形细胞瘤中0-3级染色程度MVD水平均有明显差异(P〈0.05);3级MVD〉2级MVD〉1级MVD〉0级MVD(均P〈0.05)。人脑星形细胞瘤中Eph A2蛋白表达水平、病理分级以及染色程度均与MVD呈正相关(P〈0.05)。结论:Eph A2蛋白表达水平与人脑星形细胞瘤病理分级和恶性程度具有明显的相关性,且与肿瘤微血管的生成息息相关。 Objective: To analyze the clinical expressions of tyrosine kinase receptor EphA2 in the human brain astrocytoma and its correlation with the genesis of tumor vessels. Methods: 44 patients with human brain astrocytoma treated from September 2014 to September 2017 in our hospital were selected as study group; 6 cerebral hemorrhage patients who need lobectomy were selected as control group. Through the Western Blot and immunohistochemical method,the clinical expressions of EphA2 in the human brain astrocytoma and the microvessel density(MVD) were observed. Results: The EphA2 proteins were majorly distributed in the tumor cell cytoplasm and membrane. EphA2 proteins were not detected only in 4 cases of low-grade astrocytoma,and they were detected in the other astrocytoma cases. The positive rate of EphA2 was 90. 91%. The EphA2 proteins were not detected in the normal cerebral tissues. The EphA2 expressions for two groups were significantly different(P〈0. 05). For low-grade(Grade Ⅰ - Ⅱ) and high-grade(Grade Ⅲ - Ⅳ) astrocytoma,the MVD of patients with Grade 0 - 3 staining degree was significantly different(P〈0. 05). The MVD ranged from high to low: Grade 3,Grade 2,Grade 1 and Grade 0(P〈0. 05). In the human brain astrocytoma,the EphA2 expressions,pathological grading and staining degree were positively correlated with the MVD(P〈0. 05). Conclusion: The EphA2 protein expressions have an obvious correlation with the pathological grading and malignant degree as well as the genesis of tumor microvessel.
