火龙果幼苗离体辐射诱变及其对低温胁迫的生理响应 预览

Radiation Mutagenesis of Pitaya(Hylocereus undulatus Britt )Seedlings in vitro and Its Physiological Responces to the Low Temperature Stress

摘要 为探讨火龙果幼苗离体辐射诱变及其对低温胁迫的生理响应机理,以‘紫红龙’无菌苗为试材,通过~(60)Coγ进行辐照处理,以20 mmol/L的羟脯氨酸(HYP)为选择压,筛选出拟变异植株,并对其进行初步的抗寒性生理检测。结果表明,低辐照强度(≤15 Gy)对无菌苗的成活及分化无明显影响,随着辐射剂量的增大,无菌苗的成活率和分化率显著降低,且对其生长的抑制作用越明显。以半致死剂量(LD_(50))和临界致死剂量(LD_(40))确定火龙果无菌苗辐照的适宜剂量范围为38.5~42.4 Gy。经过LD_(50)(38.5 Gy)辐射及20 mmol/L的HYP筛选成活的无菌苗,其相对电导率(REC)和丙二醛(MDA)含量均随低温处理时间的延长而逐渐增加,但增幅远低于对照;可溶性糖、可溶性蛋白和脯氨酸(Pro)含量随时间延长呈先增加后降低的变化趋势,且显著高于对照;超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)及抗坏血酸过氧化物酶(APX)活性与可溶性糖等渗透调节物质含量的变化规律相似,整个低温锻炼过程中,不同酶活性较对照分别升高了7.9%~61.5%、7.7%~47.1%、16.1%~28.4%和6.4%~63.7%。综上表明,经辐射诱变和HYP筛选的材料可能产生了一定的变异,其抗寒能力较对照大大提高。 To discuss the radiation mutagenesis of pitaya seedlings in vitro and its physiological reaction to low temperature stress, an experiment was carried out with aseptic plantlets induced from 'Zi Honglong' adult trees as the test materials to investigate the effect of 60Co-γ-rays irradiation on the survival and growth of pitaya seedlings in vitro and its physiological responses to cold stress under selected pressure of 20 mmol/L HYP. The results showed that there was no significant effect of low irradiation dose(≤15 Gy)on the survival and differentiation of the seedlings in vitro. The survival and differentiation rate decreased significantly with the increase of irradiation does, and higher irradiation dose led to a more obvious effect. The feasible irradiation dose range determined by LD50 and LD40 for 60Co-γ, mutation was 38.5-42.4 Gy. Relative electrical conductivity (REC)and malonaldehyde (MDA)content of the seedlings increased with the time extending of low temperature, no matter using 60Coγ mutagens and 20 mmol/L HYP or not. There was a remarkable influence of low temperature on soluble sugar, soluble protein and proline, and all of them increased at first and decreased subsequently with the time of low temperature processing prolonged. Most changes of osmotic regulation substances completed in a relatively short period of time. All kinds of protective enzymes, including superoxide dismutase(SOD), peroxidase(POD), catalase (CAT)and ascorbate peroxidase(APX)activity changed inconsistently with the time extension of low temperature, and they were up to7.9%-61.5%, 7.7%-47.1%, 16.1%-28.4% and 6.4%-63.7%, respectively, compared with thecontrol. Most of the enzyme activity increased notably in the early stage of low temperature treatment, but declined gradually with the temperature decline and treating time prolongation.
