目的观察分析两种STR基因座检测试剂盒中共有基因座D6S474和D5S2500的等位基因分型结果的差异。方法采用Chelex法对20名无关个体的血样提取DNA,并结合DNA参考物质9947A,分别采用Investigator HDplex试剂盒和AGCU21＋1试剂盒,经PCR复合扩增STR基因座,毛细管电泳分离扩增产物和激光扫描片段分析,比较两种试剂中共有STR基因座D6S474和D5S2500等位基因分型的差异。结果 HDplex试剂中D6S474基因座等位基因分型与AGCU 21＋1试剂中该基因座分型相比均减少一个重复序列;而两种试剂中D5S2500基因座的等位基因分型具有显著差异。结论不同厂商对同一个STR基因座的引物设计上存在差异,导致该基因座等位基因分型结果存在差异。实验室在应用多套STR基因座检测试剂盒检测时,应关注共有基因座分型结果的一致性。
Objective：To analysis and ensure the consistency of genotype results for D6 S474 and D5 S2500 loci of two STR kits. Methods：20 unrelated individual blood DNA was extracted by Chelex method and DNA reference material 9947 a,respectively,the STR loci of Investigator HDplex kit and AGCU 21 ＋ 1 kit were multiplexed by PCR and the amplified products were separated by capillary electrophoresis,DNA fragments were scanned by laser,compared the differences with the genotyping of D6 S474 and D5 S2500 in two STR kits. Results：There was difference of one repeat fragment in D6 S474 locus between the HDplex kit and AGCU 21＋1 kit;D5 S2500 locus in two kits have significant differences. Conclusion：The different designs of primers of the same STR loci producted by different manufacturers,will lead to the differences of the genotyping. In the laboratory,we should pay attention to the genotyping of the same STR loci which are owned by several STR kits.
