噬菌体cocktail JS-SP1对沙门菌流行菌株混合感染的裂菌动力学及灭菌效果研究 预览

摘要 目的 研究沙门菌噬菌体cocktail JS-SP1对沙门菌流行菌株混合感染的裂菌动力学及灭菌效果。方法 从50份污水及粪样样品中分离获得6株高裂菌活性的沙门菌噬菌体,将其按等比例混合制成沙门菌噬菌体cocktail JS-SP1,通过测定A600分析其对6株沙门菌流行菌株混合菌液的裂菌动力学,并且对0、4和10h这3个时间点的活细菌计数,分析JS-SP1的灭菌效果。结果与混合细菌组、单株噬菌体处理组相比,JS-SP1处理组可以有效而且快速降低混合液的A600值。与其他处理组相比,4h时,JSSP1处理组细菌数下降极显著(P〈0.01);10h时,降到10CFU以下(P〈0.01)。结论 噬菌体cocktail JS-SP1对混合细菌感染具有快速彻底的灭菌能力,其有可能成为一种有效控制沙门菌污染的潜在治疗手段和产品。 Objective To study the bactericidal effects and the kinetics of phage cocktail JS-SP1 against mixed epidemic Salmonella strains. Methods Six strains of high lytic Salmonella phages were isolated from 50 sewage and fecal samples, and then they were mixed to make a Salmonella phage cocktail JS-SP1 by equal proportion. The bactericidal kinetics of JS-SP1 against the mixture of the six host strainswere determined by A600, and the number of bacteria at 0, 4 and 10h were counted to assess the antibacterial effects of JS-SP1. Results Compared with the mixed bacteria group and thesingle phage group, the JS-SP1 group can effectively and rapidly reduce the A600 value of the culture. Compared with other treatment groups, the number of bacteria of JS-SP1 group decreased significantly (P〈0.01) at 4h, and was less than 10CFU at 10h (P〈0.01). Conclusion Cocktail JS-SP1 had a rapid and complete bactericidal capability for the mixed bacterial infections, and it maybe a potential therapeutic tool or product to control the Salmonella contamination.
