To extract and determine content of the flavone of Rhodiola rosea L. The extraction condition by uniform design was optimized,and U5（ 53） was selected to do the uniform design experiment. The uniform design method was applied to investigate the effects of alcohol content,liquid ratio,and extraction temperature on the result of extraction. The content of the flavone of the Rhodiola rosea L. was determined by ultraviolet-visible spectrop hotometry. The extraction conditions were optimized with the extraction yield and the mass fraction of flavone of the Rhodiola rosea L. in the extract as indexes. Exacting process were optimized of the flavone of Rhodiola rosea L. as follows： Temperature is 80 ℃,the alcohol content is 70% and liquid ratio of 1： 5（ g / m L）,the extraction ratio of flavone is 3. 75%. The result of this experiment can provide evidence for the further development and application of the extraction technology of the flavone in the Rhodiola rosea L.
