生物活性多孔钽骨修复材料促骨生长的实验研究 被引量:1

Effect of TGF-β1 combined with porous tantalum on repairing maxillofacial bone defects

摘要 目的通过将转化生长因子-β1(transforming growth factor-β1,TGF-β1)与多孔钽颗粒复合,制备生物活性多孔钽修复材料,并观察该材料的促骨生长作用。方法 6只Beagle犬,建立下颌骨缺损模型共计36个,单侧3个骨缺损分别为单纯多孔钽颗粒修复组(对照组)、TGF-β1+多孔钽颗粒修复组(实验组)和无干预组(空白对照组)。于材料植入后30、60、90 d处死动物,取下颌骨行大体观察、X线检测、甲苯胺蓝染色、四环素荧光标记检测、CD31相关抗原抗体染色。结果 30 d时实验组新骨生成面积及新生微血管数目显著大于对照组和空白对照组(P〈0.05);60 d时对照组新骨生成面积及新生微血管数目显著大于实验组和空白对照组(P〈0.05);90 d时3个时间点成骨情况差异无统计学意义(P〉0.05)。结论 TGF-β1复合多孔钽生物材料具有高促骨活性,能加速多孔钽与周围骨组织的早期生物性结合,缩短愈合时间。 Objective To prepare biological composite combining porous tantalum and TGF-β1,and observe its effect on bone growth. Methods A total of 36 mandibular defect models were established in 6Beagle dogs, TGF-β1 treated porous tantalum particles and non-treated porous tantalum particles were implanted into the defect areas of bilateral mandibular bone in Beagle dogs. Blank control group was also established. Animals were sacrificed on day 30,day 60 and day 90 after implantation. Mandibular bones were examined by X-ray detection,dissection,toluidine blue staining,tetracycline fluorescence assay,and CD31 associated antigen antibody assay. Results Larger area of new bone and more newly formed microvessels were found in the experimental group than in the control group on day 30 and day 60( P〈 0. 05). On day 90,no difference was observed among the three groups( P〉〈0. 05). Conclusion Porous tantalum combined with TGF-β1 can effectively promote bone formation and facilitate the biological adhesion of porous tantalum to surrounding bone tissues,thus shortening the healing time.
