Objective: To investigate the effects of Qingre Huayu Decoction Ⅱ on the expressions of IL-6 and IL-8 in ischemic cortex of rats with cerebral ischemia reperfusion(I/R) injury in rats. Methods: All SD rats were randomly divided into two groups, ninety-six rats were randomly divided into four groups according to random number table, the normal control group(group A, n=6), sham operation group(group B, n=30), ischemia-reperfusion group(group C, n=30) and Qingre Huayu Decoction Ⅱgroup(group D, n=30). The rat model of reperfusion after focal cerebral ischemia was established by thread embolization.Except the normal control group, other groups were further divided into 5 subgroups according to 3h, 6h, 12 h, 24 h and 48 h after I/R. After sacrificed one group was randomly selected for HE staining and histopathological changes of brain were observed in each group. The changes of IL-6/IL-8 expression on the ischemic side of hippocampus were measured by radioimmunoassay(RIA).Results: HE staining results showed that the brain tissue of group A and group B had no significant changes in pathological morphology, group C and group D had different degrees of neuronal injury, and the neuronal damage of group D was significantly lower than that of group C at each time point(P<0.05). The RIA results showed that: compared with the group B, the expressions of IL-6 and IL-8 in group C and group D were significantly higher at 3h, reached the peak at 24 h, and remained at a high level after 48h(P<0.05). Compared with group C, the expressions of IL-6 and IL-8 at 12 h, 24 h and 48 h after operation in group D decreased significantly(P<0.01). Conclusion: Qingre Huayu Decoction Ⅱ may reduce the inflammatory cascade and the cerebral ischemia reperfusion injury by reducing the release of inflammatory factors IL-6 and IL-8 to play a protective role of neuron.
