清热化瘀Ⅱ号方对大鼠脑缺血再灌注损伤IL-6及IL-8表达的影响 被引量:2

Effects of Qingre Huayu Decoction Ⅱ on the expressions of IL-6 and IL-8 in rats with cerebral ischemia reperfusion injury

摘要 目的:研究清热化瘀Ⅱ号方对大鼠脑缺血再灌注损伤缺血侧皮层白细胞介素-6(IL-6)、白细胞介素-8(IL-8)表达的影响。方法:将成年健康SD大鼠按随机数字表分为两大组,每大组再随机分为4个组,空白组、假手术组、模型组、清热化瘀Ⅱ号方组(QRHYⅡ组)。采用线栓法制备大鼠局灶性脑缺血再灌注模型,除空白组外其余3组分别按缺血再灌注2h后3、6、12、24、48h 5个时间点分为5个亚组。取材后,随机选择一组进行HE染色并观察各组脑组织病理形态学改变,另一组应用放射免疫法检测大鼠缺血侧皮层IL-6、IL-8的表达。结果:HE染色结果:空白组、假手术组均未见神经元损伤,模型组及QRHYⅡ组有不同程度的神经元损伤,QRHYⅡ组在各时间点的神经元受损较模型组组明显减轻(P<0.05);放射免疫法结果:与假手术组比较,模型组及QRHYⅡ组IL-6、IL-8在3h开始明显升高,24h达到高峰,48h后仍维持在较高水平,差异有统计学意义(P<0.05);与模型组比较,QRHYⅡ组随着存活时间的延续,IL-6、IL-8的表达在同一时间点明显下降,具有显著性的差异(P<0.01)。结论:清热化瘀Ⅱ号方可能通过下调炎性因子IL-6、IL-8的释放,减轻炎性级联反应,减轻脑缺血再灌注损伤,发挥神经元保护作用。 Objective: To investigate the effects of Qingre Huayu Decoction Ⅱ on the expressions of IL-6 and IL-8 in ischemic cortex of rats with cerebral ischemia reperfusion(I/R) injury in rats. Methods: All SD rats were randomly divided into two groups, ninety-six rats were randomly divided into four groups according to random number table, the normal control group(group A, n=6), sham operation group(group B, n=30), ischemia-reperfusion group(group C, n=30) and Qingre Huayu Decoction Ⅱgroup(group D, n=30). The rat model of reperfusion after focal cerebral ischemia was established by thread embolization.Except the normal control group, other groups were further divided into 5 subgroups according to 3h, 6h, 12 h, 24 h and 48 h after I/R. After sacrificed one group was randomly selected for HE staining and histopathological changes of brain were observed in each group. The changes of IL-6/IL-8 expression on the ischemic side of hippocampus were measured by radioimmunoassay(RIA).Results: HE staining results showed that the brain tissue of group A and group B had no significant changes in pathological morphology, group C and group D had different degrees of neuronal injury, and the neuronal damage of group D was significantly lower than that of group C at each time point(P<0.05). The RIA results showed that: compared with the group B, the expressions of IL-6 and IL-8 in group C and group D were significantly higher at 3h, reached the peak at 24 h, and remained at a high level after 48h(P<0.05). Compared with group C, the expressions of IL-6 and IL-8 at 12 h, 24 h and 48 h after operation in group D decreased significantly(P<0.01). Conclusion: Qingre Huayu Decoction Ⅱ may reduce the inflammatory cascade and the cerebral ischemia reperfusion injury by reducing the release of inflammatory factors IL-6 and IL-8 to play a protective role of neuron.
