Objective To observe the influence of icariin on ultrastructural changes of hippocampus in Alzheimer disease rat and explore the mechanism o f icariin to AD. Methods After the Morris water maze test, 18 Wistar rats were randomly divided into three groups： normal control group, model group, Icariin treatment group. The i.P. Injection of LPS for rats of model group and Icariin treatment group were administrated, Morris water maze test was used to evaluate the memory dysfunction and electron microscope was used to observe the ultrastructure of hippocampus neuron. Results In Water maze test , data of the escape latency of rats in each group was recorded, there was a significant difference between the model group and the normal control group（P 〈0.05）, as well as the same result between the treatment group and the model group（P 〈0. 05）;In spatial probe test, average time percentage spent on the target quadrant of model group was significantly decreased（P 〈0.05）while the time were significantly increased of treatment group than those of model group（P 〈 0.05）; In the model group, the cellular membrane was irregular and discontinue, nuclear membrane was cloudinessl. The nuclear chromatin was aggregate. The mitochondria was oncootic and vacuolate. The golgi apparatus was oncootic;The endoplasmic reticulum was dilative. These ultrastructural pathological changes in the treatment group were not apparent. Conclusion In this experiment, AD model mice which administrated LPS appeared decrease in memory dysfunction , it indicated that a successful AD rat model has been made. Icariin is beneficial for improving the memory dysfunction, attenuating the neural damage of AD model rat.
