摘要
目的:观察不同剂量咖啡酸对白细胞及血小板减少症小鼠凝血功能的影响。方法:采用阿糖胞苷制备白细胞及血小板减少症小鼠模型,同时每天一次灌胃给予低(100mg/kg)、中(150mg/kg)、高(200mg/kg)不同剂量咖啡酸。给药第10天,取血浆检测凝血系统凝血酶时间(TT)、凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、纤维蛋白原(FIB)以及抗凝血系统血浆蛋白C(PC)、纤溶酶原(PLG)、抗凝血酶III(AT III)水平。结果:咖啡酸各剂量组TT及PT均明显低于阴性对照组及正常组。咖啡酸中剂量组APTT明显低于阴性对照组及正常组。咖啡酸低剂量及中剂量组FIB含量明显高于阴性对照组及正常组。各组PC水平低下,且相互间比较无显著性差异。各组血浆样本中PLG均未达到检出水平。咖啡酸各剂量组AT III明显高于阴性对照组。结论:咖啡酸能够缩短TT、PT、APTT,激活内源性、外源性凝血途径,提高纤维蛋白原含量,发挥增强止血和凝血作用。咖啡酸也明显升高AT III,说明其具有增强凝血及抗凝血双向调节作用,能够调节机体维持止血及抗血栓平衡,以适应机体维持血液系统稳态所需。
Objective: To explore the effects of different doses of caffeic acid (CA) on coagulation function in leucopenia and thrembocytopenia mice. Methods: Myclosuppressed model mice were prepared by chemotherapy drug cytosine arabinoside (Ara-C), and treated with low-dose CA (100 mg/kg), intermediate-dose CA (150 mg/kg), and high-dose CA (200 mg/kg), meanwhile 0.5% carboxymethylccllttlose sodium (vehicle) was used as negative controls, once daily by gavage, and interleukin-11 was used as positive control once daily by subcutaneous injection. On day 10, we measured the level of TT, PT, APTr, FIB, PC, PLG and AT III in plasma. Results: Tr and PT in all the groups of caffeic acid are significantly lower than that in the negative control group and normal group. APTT in the intermediate-dose CA group is significantly lower than that i,n the negative control group and normal group. FIB in the intermediate-dose CA and high-dose CA groups are significantly higher than that in the negative control group and normal group. There is no difference between each group in PC and PLG in each group is under the detectable level. AT Ⅲ in all the groups of caffeic acid are significantly higher than that in the negative control group. Conclusion: Coffeic acids can shorten the time of TT, PT, APTT, activate endogenous and exogenous pathways of blood coagulation, increase Fibfinogen content and therefore enhance the function of hemostasis. Caffeic acid can also markedly elevate AT Ⅲ, which means it can regulate coagulation and anticoagulation system at the same time, adjusting and maintaining bcmostasis and antithrombotic balance.
