目的探讨低氧对肺动脉平滑肌细胞中胱硫醚-γ-裂解酶(CSE)基因表达的影响和硫化氢对肺动脉平滑肌细胞增殖的调节作用.方法人工培养大鼠的肺动脉平滑肌细胞,实验分为1.对照组;2.硫氢化钠(NaHS)组:10-4 mol/L NaHS处理24 h;3.低氧组:氧浓度1%以下24 h;4.低氧+NaHS组:低氧24 h,10-4 mol/L NaHS.每组各6孔细胞.用3H-TdR掺入估计细胞增殖,用定量竞争性RT-PCR方法测定CSE mRNA含量,评价CSE基因表达.结果低氧组平滑肌细胞中的CSE mRNA含量较对照组减少67%(P<0.01).低氧组细胞的3H-TdR掺入较对照组增加66%(P<0.01);NaHS组细胞的3H-TdR掺入较对照组减少37%(P<0.01);低氧+NaHS组的3H-TdR掺入较低氧组减少29%(P<0.01).结论低氧使人工培养的大鼠肺动脉平滑肌细胞中CSE基因表达下调,硫化氢对大鼠肺动脉平滑肌细胞增殖有抑制作用.
Objective To explore the influence of hypoxia on cystathionine gamma lyase (CSE) gene expr ession of rat cultured pulmonary artery smooth muscle cells (PASMCs) and the mod ulatory effect of hydrogen sulfide on PASMCs proliferation.Methods Rat′s cultured PASMCs were randomly divided into contr ol group,NaHS group,hypoxia group and hypoxia +NaHS group. Each group contains 6 wells of cells. Hypoxic challenge was performed with oxygen concentr ation≤1%. The cell proliferation rate was measured by 3H-thymidine i ncorporation into DNA of the PASMCs. The expression of CSE mRNA was measured by quantitative competitive reverse-transcription polymerase chain reaction (RT-PCR ).Results For the PASMCs in hypoxia group, the amount of CSE mRN A was down-regulated about 67% (P<0.01) compared with control group. Compare d with control group, the 3H-thymidine incorporation was increased by 66% (P<0.01) for PASMCs in hypoxia group; compared with control group, the 3H-thymidine incorporation was decreased by 37% for PASMCs in NaHS gr oup; compared with hypoxia group,the 3H-thymidine incorporation was d ecreased by 29% for PASMCs in hypoxia+NaHS group.Conclusion Hypoxia challenge can down-regulate the CSE gene exp ression;and hydrogen sulfide can suppress the proliferation of rat′s PASMCs.
