目的探讨右侧海马注射β-淀粉样蛋白(Aβ)对大鼠学习记忆能力和乙酰胆碱酯酶(AchE)的影响及何首乌的作用.方法将Aβ1-40 10μg/μl 注入右侧海马建立大鼠AD模型,测定其学习记忆能力和脑AchE活性及何首乌的影响作用.结果 Aβ注射后出现较持久的学习记忆障碍,Aβ组呈渐进性加重,治疗组有一定的改善.Aβ组海马区、额叶皮质和基底前脑区的AchE的活性较对照组明显减少;治疗组较对照组减少,但较Aβ组有明显提高.结论何首乌能改善AD模型大鼠学习记忆获得能力,提高AchE活性.
Objective To investigate whether amyloid-beta protein injected into hippocampus might affect memory ability and activity of acetylcholinesterase in rats,and the effects of Radix Polygonum Multiflorum Thumb(PMT). Methods The AD rat model was established by injection of amyloid-beta protein (Aβ1-40,10μg) into right hippocampus of rats. Learning and memory ability,the brain acetylcholinesterase activity were studied. Possible protective effects of Radix Polygoni Multiflori Preparata were also observed. Results Learning and memory disorder aggravated progressively in experimental group and ameliorated. Obviously in PMEG-administrated group at the 20th day. AchE activity in hippocampus,Meynert nucleus and cortex fields decreased remarkablely in experimental group,and that in PMEG-administrated group reduced obviously than in control group,but higher than in experimental group. Conclusion PMEG showes good activity,and can improve the activity of AchE.
