Injury-Resistant Action of Metallothionein and Its Relationship with Endothelin

摘要 为探讨金属硫蛋白(MT)的细胞保护机制及与内皮素(ET)的关系,在大鼠止血带休克模型上观察到MT明显改善动物休克状态的同时显著抑制其血浆ET含量的增加。离体实验发现MT呈剂量依赖地抑制ET的缩血管作用和抑制血管紧张素Ⅱ刺激ET的释放。提示MT可以通过抑制ET的释放和拮抗ET的缩血管效应,发挥其内源性抗损伤物质的作用。 In order to investigate the relationship between the cytoprotective mechanism of metallothione (MT) and its effect on endothelin (E T) , the model of rat torniq uet shock was produced. It was found that the treatment with MT significantly im proved animal shock stale, and inhibited the increase in plasma ET level of shocked rats. It was also observed that in vitro MT markedly antagonized ET-induced vasocontrac-tion and lowered angiotensin Ⅱ-stimulated release of ET in a dose-dependent manner. Tihe resulis suggest that MT could exert its injury-resistant action, as an endogenous defensive substance through inhibition of ET release and anlagonization against ET-induced vasocontraction.
