枯草芽孢杆菌溶血相关基因yqxC的表达及功能鉴定 预览 被引量:1

Expression and functional characterization of a hemolysis-associated gene yqxCfrom Bacillus subtilis

摘要 【目的】确定枯草芽孢杆菌溶血相关基因yqxC的功能。【方法】采用PCR方法,扩增枯草芽孢杆菌溶血相关基因yqxC,构建GST融合表达载体,并在大肠杆菌中进行表达。使用含体积分数5%绵羊血的琼脂平板,检测可溶性融合蛋白GST-YqxC的溶血活性。此外,采用RT-PCR检测yqxC基因在枯草芽孢杆菌中的转录情况。【结果】获得的可溶性融合蛋白GST-YqxC,在绵羊血琼脂平板上显示出较强的溶血活力;通过RT-PCR,检测到yqxC基因可以在枯草芽孢杆菌中转录。【结论】枯草芽孢杆菌yqxC基因编码蛋白YqxC有较强的溶血活性,且能够发生转录,yqxC很可能是引起枯草芽孢杆菌溶血的基因之一。 【Objective】The study was done to identify the hemolysis-associated gene yqxCfromBacillus subtilis.【Method】A hemolysis-associated gene yqxCwas amplified from the genomic DNA of B.subtilis by PCR.GST fusion expression vectors were constructed and fusion proteins were expressed in Escherichia coli.5% sheep blood agar plates were used to measure the hemolytic activity of the expressed soluble fusion protein.In addition,the transcription of yqxCwas studied by RT-PCR.【Result】The soluble fusion protein GST-YqxC sh...
